CD studies carried out on A23187 indicate a solvent-dependent conformation for the free acid. Alkali metal ions were found to bind to the ionophore weakly. Divalent metal ions such as Mg2+, Ca2+, Sr2+, Ba2+ and Co2+ and trivalent lanthanide metal ions like La3+ were found to form predominantly 2:1 (ionophore-metal ion) complexes at low concentrations of metal ions, but both 2:1 and 1:1 complexes were formed with increasing salt concentration. Mg2+ and Co2+ exhibit similar CD behaviour that differs from that observed for the other divalent and lanthanide metal ions. The structure of 2:1 complexes involves two ligand molecules coordinated to the metal ion through the carboxylate oxygen, benzoxazole nitrogen and keto-pyrrole oxygen from each ligand molecule along with one or more solvent molecules. Values of the binding constant were determined for 2:1 complexes of the ionophore with divalent and lanthanide metal ions. 相似文献

Diabetic Retinopathy (DR) is one of the attenuating complications of diabetes mellitus. The key gene responsible for causing
diabetic retinopathy is protein kinase C beta (PKCβ). Protein kinase C is a family of protein kinase enzymes which are involved in
controlling the function of other proteins through phosphorylation mechanism and plays a crucial role in signal transduction
mechanisms. Among all the PKC isoenzymes, PKCβ could be a significant isoenzyme involved in vascular dysfunction during
hyperglycemia. Studies show that oral administration of PKCβ inhibitor Ruboxistaurin (), decreases vessel permeability
and improves retinal condition. Thus compounds that decrease the PKCβ activation would be helpful in the treatment of diabetic
retinopathy. The compounds similar to Ruboxistaurin are taken from Super Target database and docking analysis was performed.
Maleimide derivative 3 showed highest binding affinities compared to Ruboxistaurin and so we advise that compound may be

The esterification reaction between stearic acid and lactic acid using Rhizomucor miehei lipase and porcine pancreas lipase was optimized for maximum esterification using response surface methodology. The formation
of the ester was found to depend on three parameters namely enzyme/substrate ratio, lactic acid (stearic acid) concentration
and incubation period. The maximum esterification predicted by theoretical equations for both lipases matched well with the
observed experimental values. In the case of R. miehei lipase, stearoyl lactic acid ester formation was found to increase with incubation period and lactic acid (stearic acid)
concentrations with maximum esterification of 26.9% at an enzyme/substrate (E/S) ratio of 125 g mol−1. In the case of porcine pancreas lipase, esterification showed a steady increase with increase in incubation period and lactic
acid (stearic acid) concentration independent of the E/S ratios employed. In the case of PPL, a maximum esterification of
18.9% was observed at an E/S ratio of 25 g mol−1 at a lactic acid (stearic acid) concentration of 0.09 M after an incubation period of 72 h.

An inhibitor of plant lipoxygenase from culture filtrates of Lactobacillus casei was purified by column chromatography and shown to be benzoic acid. The isolated benzoic acid had an IC50 of 350 M against purified soybean lipoxygenase at pH 9. L. casei therefore may have the potential to be used as a preservative against the oxidation of unsaturated fatty acids, thereby preventing undesirable flavours in foods. 相似文献

This study reports bioavailability and metabolism of fucoxanthin (FUCO) from brown algae Padina tetrastromatica in rats. Rats were divided into two groups (n = 25/group). Group one was fed basal diet (control) while the group two received retinol deficient diet (RD group) for 8 weeks. After confirmed RD in blood (0.53 μmol/l), rats were further sub-grouped (n = 5/sub group), intubated a dose of FUCO (0.83 μmol) and killed after 0, 2, 4, 6 and 8 h. The plasma levels (area under curve/8 h) of FUCO (fucoxanthinol (FUOH) + amarouciaxanthin (AAx)) was 2.93 (RD group) and 2.74 pmol/dl (control), respectively. No newly formed retinol was detected in RD rats intubated with FUCO. Besides FUOH (m/z 617 (M+H)+) and AAx (m/z 617 (M+H?)+), other deacetylated, hydrolyzed and demethylated metabolites of bearing molecular mass at m/z 600.6 (FUOH–H2O), m/z 597 (AAx–H2O), m/z 579 (AAx–2H2O+1), m/z 551 (AAx–2H2O–2CH3+2) and m/z 523 (AAx–2H2O–4CH3+4) were also detected in plasma and liver by LC-MS (APCI). The biochemical pathways followed by a mixed bacterial culture and one of its constituent strains, Sarcina maxima, MTCC 5216 (hitherto unreported) during the degradation of mononitrophenol isomers was studied using extensive nuclear magnetic
resonance (NMR) spectroscopy (One- and Two-Dimensional Heteronuclear Multiple Quantum Coherence Transfer-2D HMQCT NMR). NMR
investigations revealed that o-nitrophenol (ONP) could be degraded by the consortium to metabolites such as catechol, cis, cis-muconic acid, γ-hydroxymuconic semialdehyde, maleylacetate and β-ketoadipate. The spectra of ONP reaction mixture degraded
by S.␣maxima showed that formation of maleylacetate from γ-hydroxymuconic semialdehyde should go through a new metabolite γ-hydroxymaleylacetate,
hitherto unreported. The consortium could breakdown m-nitrophenol (MNP) to 4-aminocatechol indicating that it came from 3-hydroxyaminophenol. However, S. maxima MTCC 5216, could convert MNP to hitherto unreported 2-nitrohydroquinone and the subsequent 2-hydroxylaminohydroquinone to
1,2,4-benzenetriol along with γ-hydroxymuconic semialdehyde, muconolactone and maleylacetate. The pathway followed by the
consortium during p-nitrophenol (PNP) degradation was by the formation of 4-nitrocatechol, maleylacetate and β-ketoadipate. PNP reaction mixture
of S.␣maxima, MTCC 5216 on the other hand, showed that the pathway could proceed through the formation of p-hydroquinone as the initial metabolite. The present study conclusively established the nitrophenol-degrading ability of both

A study was conducted using ten genetically diverse genotypes along with their 45F1 (generated by diallel mating) under normal and salt stress conditions. Although, tomato (Lycopersicon esculentum Mill.) is moderately sensitive to salinity but more attention to salinity is yet to be required in the production of tomato. In present study, germination rate, speed of germination, dry weight ratio and Na+/K+ ratio in root and shoot, were the parameters assayed on three salinity levels; control, 1.0 % NaCl and 3.0 % NaCl with Hoagland’s solution. Increasing salt stress negatively affected growth and development of tomato. When salt concentration increased, germination of tomato seed was reduced and the time needed to complete germination lengthened, root/shoot dry weight ratio was higher and Na+ content increased but K+ content decreased. Among the varieties, Sel-7 followed by Arka Vikas and crosses involving them as a parent were found to be the more tolerant genotypes in the present study on the basis of studied parameters. 相似文献

Preparation of o-palmitoyl alkyl lactates with methyl, ethyl, propyl, isopropyl and butyl lactates were attempted in a complex esterification reaction using lipases as catalysts. Compared to lactic acid, alkyl lactates were found to be less inhibitory in nature as they resulted in slightly better yields at shake-flask level. Of the alkyl lactates tested, butyl lactate showed better esterification. Porcine pancreas lipase gave higher yields of esters than Rhizomucor miehei lipase (Lipozyme IM20). 相似文献

Genetically modified (GM) crops undergo large scale multi-location field trials to characterize agronomics, composition, and the concentration of newly expressed protein(s) [herein referred to as transgenic protein(s)]. The concentration of transgenic proteins in different plant tissues and across the developmental stages of the plant is considered in the safety assessment of GM crops. Reference or housekeeping proteins are expected to maintain a relatively stable expression pattern in healthy plants given their role in cellular functions. Understanding the effects of genotype, growth stage and location on the concentration of endogenous housekeeping proteins may provide insight into the contribution these factors could have on transgenic protein concentrations in GM crops. The concentrations of three endogenous proteins (actin, elongation factor 1-alpha, and glyceraldehyde 3-phosphate dehydrogenase) were measured in several different maize hybrids grown across multiple field locations over 2 years. Leaf samples were collected from healthy plants at three developmental stages across the growing seasons, and protein concentrations were quantified by indirect enzyme-linked immunosorbent assay (ELISA) for each protein. In general, the concentrations of these three endogenous proteins were relatively consistent across hybrid backgrounds, when compared within one growth stage and location (2–26%CV), whereas the concentrations of proteins in the same hybrid and growth stage across different locations were more variable (12–64%CV). In general, the protein concentrations in 2013 and 2014 show similar trends in variability. Some degree of variability in protein concentrations should be expected for both transgenic and endogenous plant-expressed proteins. In the case of GM crops, the potential variation in protein concentrations due to location effects is captured in the current model of multi-location field testing. 相似文献
