The diet of fork-tailed (Oceanodroma furcata) and Leach's storm-petrel(O.leucorhoa) was studied at Petrel Island, a small island northof Hippa Island in the Queen Charlotte Islands, British Columbiain 1983. The diet of the two storm-petrel species consistedmostly of the amphipod Paracallisoma coecus and fish. Most fisheswhich could be identified were myctophids, and may have beenacquired by storm-petrels at night. Fishes increased while P.coecusdeclined in importance as a food over the nestling season ofthe storm-petrels. The amount of oil also declined in the Leach'sstorm-petrel diet over the season. Since P.coecus contains muchoil, the seasonal decline of both P.coecus and oil was probablyassociated. Other less important foods were jellyfishes, copepods,isopods, euphausiids, shrimp, squid and octopus. Jellyfishes(Velella velella) only occurred in the diet of Leach's storm-petrels,which may relate to that storm-petrel feeding in open oceanwhere V. velella occurs most frequently. The most numerous copepodwas Neocalanus cristatus, which was only observed in the storm-petreldiet early on during the study period. An abyssal copepod, Bathycalanusbradyi, and the isopod Cirolana californiensis (tentativelyidentified, the latter only previously known from sea canyonsoff California) were surprising food items of storm-petrels.Most euphausiid prey consisted of Thysanoessa spinifera andEuphausia pacifica.  相似文献

ADAM33 (a disintegrin and metalloprotease) was recently found to be a novel asthma susceptibility gene. Domain-specific antibodies were used to study its expression and processing. When the pro-domain and catalytic domain were expressed by a stable-transfected cell line, the pro-domain was removed by cleavage within a putative furin cleavage site. The catalytic domain was active in an alpha(2)-macroglobulin complex formation assay and mutation of the catalytic site glutamic acid (E346A) eliminated activity. In transient transfections using the full-length protein, a pro-form and mature form were detectable and alternate glycosylation was demonstrated at sites within the catalytic domain. ADAM33 was detected on the cell surface, with the majority of protein detected intracellularly. The E346A mutation had no significant effect on protein processing. Endogenous ADAM33 was detected in bronchus tissue, bronchial smooth muscle cells, and MRC-5 fibroblasts, consistent with a role in the pathophysiology of asthma.  相似文献

It is anticipated that a lowering of the water table and reduced soil moisture levels in peatlands may increase peat decomposition rates and consequently affect nutrient availability. However, it is not clear if patterns will be consistent across different peatland types or within peatlands given the natural range of ecohydrological conditions within these systems. We examined the effect of persistent drought on peatland nutrient dynamics by quantifying the effects of an experimentally lowered water table position (drained for a 10-year period) on peat KCl-extractable total inorganic nitrogen (ext-TIN), peat KCl-extractable nitrate (ext-NO3 ?), and water-extractable ortho-phosphorus (ext-PO4 3?) concentrations and net phosphorus (P) and nitrogen (N) mineralization and nitrification rates at natural (control) and drained microforms (hummocks, lawns) of a bog and poor fen near Québec City, Canada. Drainage (water table drawdown) decreased net nitrification rates across the landscape and increased ext-NO3 ? concentrations, but did not affect net N and P mineralization rates or ext-TIN and ext-PO4 3? concentrations. We suggest that the thick capillary fringe at the drained peatland likely maintained sufficient moisture above the water table to limit the effects of drainage on microbial activity, and a 20 cm lowering of the water table does not appear to have been sufficient to create a clear difference in nutrient dynamics in this peatland landscape. We found some evidence of differences in nutrient concentrations with microforms, where concentrations were greater in lawn than hummock microforms at control sites indicating some translocation of nutrients. In general, the same microtopographic differences were not observed at drained sites. The general spatial patterns in nutrient concentrations did not reflect net mineralization/immobilization rates measured at our control or drained peatlands. Rather, the spatial patterns in nutrient availability may be regulated by differences in vegetation (mainly Sphagnum moss) cover between control and drained sites and possibly differences in hydrologic connection between microforms. Our results suggest that microform distribution and composition within a peatland may be important for determining how peatland nutrient dynamics will respond to water table drawdown in northern peatlands, as some evidence of microtopographic differences in nutrient dynamics was found.  相似文献

Phosphorus and nitrogen retention in five Precambrian shield wetlands   总被引:11,自引:7,他引:4  
Phosphorus and nitrogen mass balances of five wetlands (two beaver ponds, two conifer-Sphagnum swamps and one sedge fen) situated in three catchments in central Ontario, Canada, were measured. Monthly and annual input-output budgets of total phosphorus (TP), total nitrogen (TN), total organic nitrogen (TON), total inorganic nitrogen (TIN), ammonium ion (NH4 + -N), nitrate (NO 3 -N) and dissolved organic carbon (DOC) were estimated for the five wetlands during the 1982–83 and 1983–84 water years. Except for the deepest beaver pond (3.2 m) which had annual TP retention of –44% (–0.030 ± 0.015 g m–2 yr–1), the wetlands retained < 0.001 to 0.015 g M–2 yr–1 ; however, this wasless than 20% of the inputs and the estimated budget uncertainties were equal to or greater than the retention rates. Annual TN retentions ranged from –0.44 to 0.56 g m–2 yr–1 (–12 to 4%) but were not significantly different from zero. The wetlands transformed nitrogen by retaining TIN (16 to 80% RT) and exporting an equivalent amount as TON (–7 to 102% RT). The beaver ponds, however, retained NO 3 while NH 4 + was passed through or the outputs exceeded the inputs. In contrast, the conifer swamps retained both NH 4 + and NO 3 . DOC fluxes into and out of the beaver ponds were equal (–18 and 4% RT) but output from the conifer swamps exceeded input by > 90%. Marked seasonal trends in nutrient retention were observed. Nutrient retention coincided with low stream flow, increased evapotranspiration and biotic uptake during the summer. Net nutrient export occurred during the winter and spring when stream flows were highest and biotic uptake was low.  相似文献

Initiation of wetland features is integral to sustaining landscape eco-hydrological function and meeting defined goals in surface mine reclamation. Within the sub humid climate of the Athabasca Oil Sands, Canada, the water generation mechanisms (external water sources, internal feedback mechanisms) that enable wetlands to form opportunistically on recently reconstructed landscapes are currently unknown, restricting the flexibility in mine closure planning. To address this knowledge gap, we interpret site and local physical characteristics of opportunistic wetlands within the Athabasca Oil Sands through a synoptic survey. Wetlands formed in ~?8% of the random survey transect areas designed and planted for forestlands. Wetlands had vegetation structures characteristic of woody Salix spp. swamps and narrow-leaved Carex spp. marsh wetland types, with minor coverage of open water marshes. Wetlands formed opportunistically over a range of slopes, aspects and topographic positions, across contrasting fine and coarse-textured landforms. However, different wetland establishment and maintenance controls exist on fine and coarse-textured landforms. On coarse-textured landforms with large groundwater transmissivity, wetland formation was influenced by landscape-scale factors; wetlands were restricted to the toes of slopes and areas intersecting groundwater. On fine-textured constructed landforms, small and large wetlands occurred on lower landscape elevations with the potential for the external (cumulative) water sources, and in hydrologically isolated locations with little potential for runoff contribution from adjacent forestlands (saturation and wetland formation through internal feedback mechanisms). Regardless of landscape position, wetlands formed on flat areas and in shallow inward draining endorheic pans with clay rich soils where low water storage potential promotes frequent surface saturation. These findings have important implications in landscape reclamation design, suggesting that passive techniques that support internal feedback mechanisms may offer a more cost effective reclamation approach compared to more active, expensive techniques that aim to develop wetlands with external water sources.

An intranasal DNA vaccine prime followed by a gp41 peptide booster immunization was compared with gp41 peptide and control immunizations. Serum HIV-1-specific IgG and IgA as well as IgA in feces and vaginal and lung secretions were detected after immunizations. Long-term humoral immunity was studied for up to 12 mo after the booster immunization by testing the presence of HIV-1 gp41- and CCR5-specific Abs and IgG/IgA-secreting B lymphocytes in spleen and regional lymph nodes in immunized mice. A long-term IgA-specific response in the intestines, vagina, and lungs was obtained in addition to a systemic immune response. Mice immunized only with gp41 peptides and L3 adjuvant developed a long-term gp41-specific serum IgG response systemically, although over a shorter period (1-9 mo), and long-term mucosal gp41-specific IgA immunity. HIV-1-neutralizing serum Abs were induced that were still present 12 mo after booster immunization. HIV-1 SF2-neutralizing fecal and lung IgA was detectable only in the DNA-primed mouse groups. Intranasal DNA prime followed by one peptide/L3 adjuvant booster immunization, but not a peptide prime followed by a DNA booster, was able to induce B cell memory and HIV-1-neutralizing Abs for at least half of a mouse's life span.  相似文献

During paramyxovirus entry into a host cell, receptor engagement by a specialized binding protein triggers conformational changes in the adjacent fusion protein (F), leading to fusion between the viral and cell membranes. According to the existing paradigm of paramyxovirus membrane fusion, the initial activation of F by the receptor binding protein sets off a spring-loaded mechanism whereby the F protein progresses independently through the subsequent steps in the fusion process, ending in membrane merger. For human parainfluenza virus type 3 (HPIV3), the receptor binding protein (hemagglutinin-neuraminidase [HN]) has three functions: receptor binding, receptor cleaving, and activating F. We report that continuous receptor engagement by HN activates F to advance through the series of structural rearrangements required for fusion. In contrast to the prevailing model, the role of HN-receptor engagement in the fusion process is required beyond an initiating step, i.e., it is still required even after the insertion of the fusion peptide into the target cell membrane, enabling F to mediate membrane merger. We also report that for Nipah virus, whose receptor binding protein has no receptor-cleaving activity, the continuous stimulation of the F protein by a receptor-engaged binding protein is key for fusion. We suggest a general model for paramyxovirus fusion activation in which receptor engagement plays an active role in F activation, and the continued engagement of the receptor binding protein is essential to F protein function until the onset of membrane merger. This model has broad implications for the mechanism of paramyxovirus fusion and for strategies to prevent viral entry.  相似文献

The timing of transitions between life history stages should be affected by factors that influence survival and growth of organisms in adjacent life history stages. In a series of laboratory experiments, we examined the influence of predation risk as a cue to trigger a life history switch in amphibians. In the Oregon Cascade Mountains, some populations of Pacific treefrogs ( Hyla regilla ) and Cascades frogs ( Rana cascadae ) are under intense egg predation by predatory leeches (families Glossiphonidae and Erpobdellidae). We document that both treefrogs and Cascades frogs show plasticity in hatching characteristics in response to the threat of egg predation. Pacific treefrogs hatch sooner and at an earlier developmental stage when either predatory leeches or non-predatory earthworms are allowed direct contact with the developing egg mass. The same response is elicited even without direct contact. Chemical cues of predatory leeches and chemicals released from injured eggs appear to elicit the same early hatching response in treefrogs. For Cascades frogs, cues of leeches, but not those of injured eggs, elicit an early hatching response. Hatching early in response to egg predators may reduce predation. Plasticity of hatching characteristics has rarely been examined. However, we suspect that it may be common, particularly in populations or species that experience high variability in predation pressure between years.  相似文献

Sulphate (SO 4 –2 ) concentrations in 34 intensively measured Canadian Shield streams near the Dorset Research Centre, central Ontario, were used to test a hydrogeologic model that uses simple measures of wetland area and till depth to identify catchments that produce SO 4 –2 pulses. Mean annual measured maximum SO 4 –2 concentrations were significantly greater in shallow till (<1 m depth) catchments containing wetlands than catchments covered with deeper tills (>1 m depth) containing wetlands or catchments with no wetlands. Average maximum SO 4 –2 concentrations in wetland catchments during years with dry summers were >20 mg/L in 19 of 20 catchments with average till depths of <1 m, whereas concentrations were <20 mg/L in 5 of 6 watersheds with average till depths of >1 m. Peaks in mean annual maximum SO 4 –2 concentrations from wetland catchments with shallow till occurred during summers with rain fall 150–200 mm less than potential evaporation estimates. There were no significant differences in mean average annual SO 4 –2 concentration among the different catchments during wet summers, with SO 4 –2 concentrations ranging from 6 to 13 mg/L. These observations suggest that a large portion of the temporal and spatial variation in SO 4 –2 chemistry and export can be predicted in headwater catchments of the Canadian Shield and perhaps in other landscapes where till depth influences upland-wetland hydrologic connections.  相似文献
