Multipotent stem cells in the body facilitate tissue regeneration, growth, and wound healing throughout life. The microenvironment in which they reside provides signals that direct these progenitors to proliferate, differentiate, or remain dormant; these factors include soluble molecules, the extracellular matrix, neighboring cells, and physical stimuli. Recent advances in the culture of embryonic stem cells and adult progenitors necessitate an increased understanding of these phenomena. Here, we summarize the interactions between stem cells and their local environment, drawing on in vivo observations and tissue culture studies. In addition, we describe novel methods of characterizing the effects of various environmental factors and review new techniques that enable scientists and engineers to more effectively direct stem cell fate.  相似文献

Although a centrifugal bioreactor (CCBR) supports high-density mammalian suspension cell cultures by balancing drag, buoyancy, and centrifugal forces, to date anchorage-dependent cultures have not been tried. Also, steady or intermittent hydrostatic pressures of 8 to 500 kPa, and shears of 0.02 to 1.4 N/m(2) can be simultaneously applied in the CCBR. This article demonstrates the use of a CCBR to stimulate chondrogenesis in a high-density culture. At 3 weeks, histological results show even distribution of glycosaminoglycan (GAG) and collagen, with 1,890 ± 270 cells/mm(2) cell densities that exceed those of 1,470 ± 270 in pellet cultures. Analysis of collagen content reveals similar levels for all treatment groups; 6.8 ± 3.5 and 5.0 ± 0.4 μg collagen/μg DNA for 0.07 and 0.26 MPa CCBR cultures, respectively, in contrast to 6.6 ± 1.9 values for control pellet cultures. GAG levels of 5.6 ± 1.5 and 4.1 ± 0.9 μg GAG /μg DNA are present for cultures stressed at 0.07 and 0.26 MPa, respectively, in comparison to control pellet cultures at the 8.4 ± 0.9 level. Although results to date have not revealed mechanical stress combinations that stimulate chondrogenesis over unstressed controls, system advantages include continuous culture at cell densities above those in the pellet, precise medium control, the ability to independently vary multiple mechanical stresses over a broad range, and the flexibility for integration of scaffold features for future chondrogenesis stimulation studies.  相似文献

Background

In the years 2000 and 2002, the German Environment Agency in Berlin (UBA) published the results of a comprehensive LCA study on beverage containers comprising aluminium cans with volumes of 330 ml and 500 ml. Starting with the aluminium can scenarios and the respective results obtained during the UBA study, additional analyses were performed by IFEU in 2003, a German consultant having been a member of the project team working on the UBA study. The objective was to examine the influence of selected parameters on the LCA profile of carbonated soft drink containers. Data and method were in complete analogy with the LCI and LCA part of the UBA study.

Materials

In 2006, the aluminium industry commissioned a study on further influential factors that help determine the sale of certain types of beer, studying the effects of two selected parameter settings on the comparative results of the aluminium can against the refillable glass bottle. In this scenario, special attention was given to two influential factors, the distribution distance—distinguished by regional and nationwide distribution—and trippage rate.

Results and discussion

The results of the initial LCA from the years 2000 and 2002 showed, for the examined parameters container weight, rate of post-consumer recovery of used containers, degree of recycled content and quality of the recycling routes, that each had a considerable influence on the environmental impact profile of the aluminium can within the given framework. Can weight and recycling rate were sensitive factors in the impact categories of climate change, fossil resources, summer smog (POCP), acidification and terrestrial eutrophication. Can volume affected virtually all impact categories examined.

Conclusions

By now, individual improvement options have already been put into practice in Germany. The environmental profile of the average 330 ml aluminium can on the German market can be expected to be ahead of that of the aluminium can at the time of the UBA study. The introduction of a 500-ml can on the market denotes a fundamental step forward in improving LCA results of the aluminium can as a container for beverages.

An increasing demand for products such as tissues, proteins, and antibodies from mammalian cell suspension cultures is driving interest in increasing production through high‐cell density bioreactors. The centrifugal bioreactor (CCBR) retains cells by balancing settling forces with surface drag forces due to medium throughput and is capable of maintaining cell densities above 108 cells/mL. This article builds on a previous study where the fluid mechanics of an empty CCBR were investigated showing fluid flow is nonuniform and dominated by Coriolis forces, raising concerns about nutrient and cell distribution. In this article, we demonstrate that the previously reported Coriolis forces are still present in the CCBR, but masked by the presence of cells. Experimental dye injection observations during culture of 15 μm hybridoma cells show a continual uniform darkening of the cell bed, indicating the region of the reactor containing cells is well mixed. Simulation results also indicate the cell bed is well mixed during culture of mammalian cells ranging in size from 10 to 20 μm. However, simulations also allow for a slight concentration gradient to be identified and attributed to Coriolis forces. Experimental results show cell density increases from 0.16 to 0.26 when centrifugal force is doubled by increasing RPM from 650 to 920 at a constant inlet velocity of 6.5 cm/s; an effect also observed in the simulation. Results presented in this article indicate cells maintained in the CCBR behave as a high‐density fluidized bed of cells providing a homogeneous environment to ensure optimal growth conditions. © 2010 American Institute of Chemical Engineers Biotechnol. A mathematical model has been developed to describe the mechanism for internal mass transfer and enzyme reaction kinetics of an amperometric conductive matrix enzyme electrode. The model is simplified and solved analytically to arrive at a representation for the response slope in the linear range as well as for the response time. This is the first time that the response time of an enzyme electrode is described by a mathematical model. Simulations give information on how the design parameters influence the performance of the electrode for a glucose oxidase catalyzed sensing reaction process. Based on this information, several designs were constructed and tested showing suitable agreement with theoretical predictions. Finally, an optimized electrode was designed and validated.  相似文献

Background and purpose

Numerous life cycle assessments (LCAs) have been conducted on the environmental impacts of beverage packaging systems. With such a potentially rich source of knowledge available, it seemed worthwhile to conduct a comprehensive evaluation of those existing studies. This paper describes a recent ‘meta analysis’, whose goal it was to provide a structured overview of LCAs on beverage cartons and other packaging systems from past years in order to answer two key questions: (1) Is it possible to draw general conclusions regarding the environmental performance (in terms of strengths and weaknesses) of beverage cartons in comparison to alternative packaging systems from these existing LCAs? (2) If certain trends arise across these LCA studies regarding the environmental performance of beverage cartons compared to other packaging systems for beverages, what can be said on their validity and limitations?

Methods

The meta analysis presented covers 22 LCA studies, all of which fulfil three criteria: (1) full life cycle approach, (2) beverage carton must be among the products evaluated in study, and (3) comparative approach. Each of these studies was categorised either as a core study (if focussed on Europe, conducted in 2000 or later, and peer reviewed) or as a basic study. Next to providing detailed comparisons of the analysed studies, the full report on the meta analysis was designed to allow a quick understanding of their main characteristics (or ‘profiles’). Similarities and differences were highlighted both in terms of results and the applied methodologies (e.g., key settings) and the validity and limitations of the findings were stated. Additionally, further environment-related topics of special interest to stakeholders in the beverage packaging industry were addressed.

Results, discussion, and conclusions

For certain environmental impact indicators/inventory categories, the LCA studies covered in this meta analysis indicate general trends regarding the performance of beverage cartons versus alternative packaging systems. For climate change, cumulated energy demand/fossil resource consumption, and acidification, all regarded by the majority of all studies, beverage cartons mostly have the most favourable results, while in terms of land use for forestry, they clearly require the largest area. For summer smog and terrestrial eutrophication, the result ‘pattern’ points towards a favourable picture for beverage cartons; however, fewer LCA studies provide results for these impact categories. For other environmental aspects, where the results of the analysed studies vary strongly, no clear pattern can be made out. Several aspects were covered in too few LCA studies in order for an overall trend or lack thereof to become visible, and still others—which in part have been receiving increased attention in the past years—are not addressed in any of the analysed core or basic studies.

Increasing demand for tissues, proteins, and antibodies derived from cell culture is necessitating the development and implementation of high cell density bioreactors. A system for studying high density culture is the centrifugal bioreactor (CCBR), which retains cells by increasing settling velocities through system rotation, thereby eliminating diffusional limitations associated with mechanical cell retention devices. This article focuses on the fluid mechanics of the CCBR system by considering Coriolis effects. Such considerations for centrifugal bioprocessing have heretofore been ignored; therefore, a simpler analysis of an empty chamber will be performed. Comparisons are made between numerical simulations and bromophenol blue dye injection experiments. For the non‐rotating bioreactor with an inlet velocity of 4.3 cm/s, both the numerical and experimental results show the formation of a teardrop shaped plume of dye following streamlines through the reactor. However, as the reactor is rotated, the simulation predicts the development of vortices and a flow profile dominated by Coriolis forces resulting in the majority of flow up the leading wall of the reactor as dye initially enters the chamber, results are confirmed by experimental observations. As the reactor continues to fill with dye, the simulation predicts dye movement up both walls while experimental observations show the reactor fills with dye from the exit to the inlet. Differences between the simulation and experimental observations can be explained by excessive diffusion required for simulation convergence, and a slight density difference between dyed and un‐dyed solutions. Local extinctions are often non‐randomly associated with range size, dispersal ability and habitat specificity, as well as body size, sexual dimorphism and phylogeny. We used a large data set of the Orthoptera species (bush crickets, crickets, grasshoppers) occurring in Germany and compared the number of occupied grid cells before 1980 to those occupied after 1980, corrected for monitoring intensity. The number of grid cells in which a species went extinct was non‐linearly related to the number of occupied grid cells per species. Using generalized linear modelling we analysed extinction in relation to national distribution (the number of occupied grid cells before 1980), dispersal ability (derived from a large body of literature concerning wing development, colonization dynamics and within‐habitat mobility), habitat specificity (moisture specialists versus generalists), potential reproduction (the number of ovarioles), the degree of sexual size dimorphism and phylogeny (twelve clades). Species with a large global range size also had a large national range size. Species with a large range experienced more total extinction events than species with smaller ranges but relatively fewer compared to range size. The latter relationship was largely shaped by the dispersal ability of the species: the interactions of range size×dispersal ability and range size×habitat specificity explained almost one third of the variation in the number of extinction events. Species with high dispersal ability went extinct in a similar number of grid cells irrespective of their range size. By contrast, species with low dispersal ability went extinct in proportion to their range size. Therefore, comparing the speed of extinction across species in the conventional way of extinction rates (that is the percentage of range contraction) might be flawed because it only applies to species with low dispersal ability. Sexual size dimorphism was not a significant predictor of extinction. Extinction was not concentrated on particular clades.  相似文献

Intestinal barrier dysfunction is associated with chronic gastrointestinal tract inflammation and diseases such as IBD and IBS. Serum-derived bovine immunoglobulin/protein isolate (SBI) is a specially formulated protein preparation (>90%) for oral administration. The composition of SBI is greater than 60% immunoglobulin including contributions from IgG, IgA, and IgM. Immunoglobulin within the lumen of the gut has been recognized to have anti-inflammatory properties and is involved in maintaining gut homeostasis. The binding of common intestinal antigens (LPS and Lipid A) and the ligand Pam3CSK4, by IgG, IgA, and IgM in SBI was shown using a modified ELISA technique. Each of these antigens stimulated IL-8 and TNF-α cytokine production by THP-1 monocytes. Immune exclusion occurred as SBI (≤50 mg/mL) bound free antigen in a dose dependent manner that inhibited cytokine production by THP-1 monocytes in response to 10 ng/mL LPS or 200 ng/mL Lipid A. Conversely, Pam3CSK4 stimulation of THP-1 monocytes was unaffected by SBI/antigen binding. A co-culture model of the intestinal epithelium consisted of a C2BBe1 monolayer separating an apical compartment from a basal compartment containing THP-1 monocytes. The C2BBe1 monolayer was permeabilized with dimethyl palmitoyl ammonio propanesulfonate (PPS) to simulate a damaged epithelial barrier. Results indicate that Pam3CSK4 was able to translocate across the PPS-damaged C2BBe1 monolayer. However, binding of Pam3CSK4 by immunoglobulins in SBI prevented Pam3CSK4 translocation across the damaged C2BBe1 barrier. These results demonstrated steric exclusion of antigen by SBI which prevented apical to basal translocation of antigen due to changes in the physical properties of Pam3CSK4, most likely as a result of immunoglobulin binding. This study demonstrates that immunoglobulins in SBI can reduce antigen-associated inflammation through immune and steric exclusion mechanisms and furthers the mechanistic understanding of how SBI might improve immune status and reduce inflammation in various intestinal disease states.  相似文献
