Experimental data on selection of Streptococcus lactis producing the polypeptide antibiotic nisin with the method of protoplast fusing, one of the modern methods of cell engineering are presented. Four strains of Streptococcus lactis differing in their nisin-producing levels and difficult for protoplasting were used in the study. It was shown possible to transfer them to the protoplast form when respective conditions for their preliminary cultivation and regeneration are provided. Distinctive features of these strains with respect to the antibiotic resistance, sugar fermentation and growth component requirements were revealed. The protoplast fusing yielded hybrids differing from the parent strains by a number of phenotypical features and nisin-synthesizing activity.  相似文献

To increase the nisin synthesizing activity of the natural strain Lactococcus lactis subsp. lactis 119 isolated from sour milk, UV irradiation in different doses was used followed by isolation of productive clones. The highest mutation effect was observed with the dose of 76,000 erg/mm2, when 11% of the cells increased their productivity by 12.8% at the minimum survival rate. Two-step UV irradiation and adaptive selection on the nisin-contaning medium provided isolation of a strain with the activity 42.6% higher than that of the initial strain (3850 IU/ml). Natural and UV-induced variability of the strain by the nisin synthesis, growth rate, carbohydrate consumption and sensitivity to antibiotics of various groups were studied.  相似文献

Insect pathogenic viruses and parasitoids represent distinct biological entities that exploit a shared host resource and have similar effects in suppressing host populations. This study explores the interactions between the ectoparasitoid Euplectrus plathypenae (Hymenoptera: Eulophidae) and the Spodoptera exigua multiple nucleopolyhedrovirus (SeMNPV) in larvae of S. exigua (Lepidoptera: Noctuidae). Parasitoid progeny failed to complete development in hosts that had been infected prior to parasitism. However, infection of S. exigua fourth instars at 48 h post‐parasitism had no significant effects on the survival of parasitoid progeny. Larval and pupal development times of E. plathypenae that survived on virus‐infected S. exigua did not differ significantly from that of parasitoids on healthy hosts. Virus‐induced mortality and the production of occlusion bodies were very similar in parasitized and non‐parasitized S. exigua. The virus was genetically stable over three passages in parasitized and unparasitized hosts. These results suggest that applications of SeMNPV‐based insecticides are unlikely to disrupt pest control exerted by the parasitoid E. plathypenae in biological pest control programs as long as virus applications are timed not to coincide with parasitoid releases.  相似文献

Syn5 is a marine cyanophage that is propagated on the marine photosynthetic cyanobacterial strain Synechococcus sp. WH8109 under laboratory conditions. Cryoelectron images of this double-stranded DNA (dsDNA) phage reveal an icosahedral capsid with short tail appendages and a single novel hornlike structure at the vertex opposite the tail. Despite the major impact of cyanophages on life in the oceans, there is limited information on cyanophage intracellular assembly processes within their photosynthetic hosts. The one-step growth curve of Syn5 demonstrated a short cycle with an eclipse period of ~45 min, a latent phase of ~60 min, and a burst size of 20 to 30 particles per cell at 28°C. SDS-PAGE and Western blot analysis of cell lysates at different times after infection showed the synthesis of major virion proteins and their increase as the infection progressed. The scaffolding protein of Syn5, absent from virions, was identified in the lysates and expressed from the cloned gene. It migrated anomalously on SDS-PAGE, similar to the phage T7 scaffolding protein. Particles lacking DNA but containing the coat and scaffolding proteins were purified from Syn5-infected cells using CsCl centrifugation followed by sucrose gradient centrifugation. Electron microscopic images of the purified particles showed shells lacking condensed DNA but filled with protein density, presumably scaffolding protein. These findings suggest that the cyanophages form infectious virions through the initial assembly of scaffolding-containing procapsids, similar to the assembly pathways for the enteric dsDNA bacteriophages. Since cyanobacteria predate the enteric bacteria, this procapsid-mediated assembly pathway may have originated with the cyanophages.  相似文献

The marine cyanophage Syn5 can be propagated to a high titer in the laboratory on marine photosynthetic Synechococcus sp. strain WH8109. The purified particles carry a novel slender horn structure projecting from the vertex opposite the tail vertex. The genome of Syn5 includes a number of genes coding for novel proteins. Using immune-electron microscopy with gold-labeled antibodies, we show that two of these novel proteins, products of genes 53 and 54, are part of the horn structure. A third novel protein, the product of gene 58, is assembled onto the icosahedral capsid lattice. Characterization of radioactively labeled precursor procapsids by sucrose gradient centrifugation shows that there appear to be three classes of particles—procapsids, scaffold-deficient procapsids, and expanded capsids. These lack fully assembled horn appendages. The horn presumably assembles onto the virion just before or after DNA packaging. Antibodies raised to the recombinant novel Syn5 proteins did not interfere with phage infectivity, suggesting that the functions of these proteins are not directly involved in phage attachment or infection of the host WH8109. The horn structure may represent some adaption to the marine environment, whose function will require additional investigation.  相似文献

The authors studied the effect of some factors, including the conditions of preincubation, the action of 2-mercaptoethanol, EDTA, alpha-amylase, on protoplast production in four strains of Streptococcus lactis caused by lysozyme. The strains differed in the nisin-producing activity and in the structure of the cell walls that were not affected with lysozyme without either preincubation in 2-mercaptoethanol or in a salt medium with minimal inhibitory concentrations of DL-threonine. EDTA and alpha-amylase increased the lysozyme effect. Among seven buffer systems studied the most favourable for protoplast production in S. lactis is the ammonia-citric buffer with EDTA, and the best regeneration medium is the agar salt medium to which, depending on the strain, either glucose or sucrose should be added as a stabilizer.  相似文献
