Virus filtration is a key clearance unit operation in the manufacture of recombinant protein, monoclonal antibody, and plasma-derived biopharmaceuticals. Recently, a consensus has developed among filter manufacturers and end users about the desirability of a common nomenclature and a standardized test for classifying and identifying virus-retentive filters. The Parenteral Drug Association virus filter task force has chosen PR772 as the model bacteriophage to standardize nomenclature for large-pore-size virus-retentive filters (filters designed to retain viruses larger than 50 to 60 nm in size). Previously, the coliphage PR772 (Tectiviridae family) has been used in some filtration studies as a surrogate for mammalian viruses of around 50 to 60 nm. In this report, we describe specific properties of PR772 critical to the support of its use for the standardization of virus filters. The complete genomic sequence of virulent phage PR772 was determined. Its genome contains 14,946 bp with an overall G+C content of 48.3 mol%, and 32 open reading frames of at least 40 codons. Comparison of the PR772 nucleotide sequence with the genome of Tectiviridae family prototype phage PRD1 revealed 97.2% identity at the DNA level. By dynamic light-scattering analysis, its hydrodynamic diameter was measured as 82 +/- 6 nm, consistent with use in testing large-virus-retentive filters. Finally, dynamic light-scattering analysis of PR772 preparations purified on CsCl gradients showed that the phage preparations are largely monodispersed. In summary, PR772 appears to be an appropriate model bacteriophage for standardization of nomenclature for larger-pore-size virus-retentive filters. 相似文献

【目的】研究分离自四川凉山州新银合欢根瘤菌的遗传多样性和共生有效性。【方法】采用16S rRNA RFLP、BOX-PCR、AFLP、多位点持家基因序列的联合分析及无氮水培法对33株供试新银合欢根瘤菌的遗传多样性和共生有效性进行研究。【结果】分析表明,3种方法在属水平的分群结果具有较好的一致性,有1个Mesorhizobium属的菌株、3个Bradyrhizobium属的菌株、3个Rhizobium属的菌株,26个相似度较高的菌株属Sinorhizobium。16S rRNA-recA-atpD-glnII序列联合构建的新银合欢根瘤菌系统发育树表明,SCAU203、SCAU211可能分别是Rhizobium和Bradyrhizobium的新类群,另外3个代表菌株分别位于Sinorhizobium、Mesorhizobium、Bradyrhizobium分支,分别与S.americanum、M.Plurifarium、R.huautlense亲缘关系最近。无氮水培接种试验筛选出2个共生固氮效果好、与不接种对照处理差异达显著水平的菌株SCAU229和SCAU307,有3个菌株不仅不具共生有效性,甚至不利于宿主的生长,其余84%的供试菌为低效或无效菌株。【结论】凉山州新银合欢根瘤菌具有丰富的遗传多样性,分布于4个属:Rhizobium、Bradyrhizobium、Mesorhizobium、Bradyrhizobium,79%为Sinorhizobium属的菌株,优势菌群为Sinorhizobium。该区的新银合欢根瘤菌大多数的共生有效性差。 相似文献

Background
This study aimed to construct a biological age assessment formula for the Chinese population and to explore the effectiveness of double filtration plasmapheresis for anti-ageing and longevity.Methods
915 subjects were recruited, including 584 (63.8%) males and 331 females (36.2%). Male age was 50.94±10.60 (mean±SD), and female age was 51.20±11.84 (mean±SD). 34 blood markers were detected in the laboratory. The ageing biomarkers were determined by statistical correlation analysis and redundancy analysis, and the biological age assessment formula was established by multiple linear regression analysis. Paired sample T test was used to analyse the elimination effect of double filtration plasmapheresis on aging biomarkers.Results
Based on the comprehensive blood test and analysis, the ageing biomarkers were screened, and the male and female biological age assessment formulas were established. Then, the elimination of ageing biomarkers by double filtration plasmapheresis was examined. Double filtration plasmapheresis can eliminate ageing biomarkers, with an average of 4.47?years decrease in age for males and 8.36?years for females.Conclusion
So, biological age provides a scientific tool for assessing ageing, and double filtration plasmapheresis is safe and might be effective for anti-ageing and longevity. However, the effect of plasmapheresis is expected to be transient, so further studies are needed to plan the number and range of the plasmapheresis procedures necessary to consistently lower the parameters under study.7.

Intelligent gene delivery systems based on physiologically triggered reversible shielding technology have evinced enormous interest due to their potential in vivo applications. In the present work, an acid-labile block copolymer consisting of poly(ethylene glycol) and poly(2-(dimethylamino)ethyl methacrylate) segments connected through a cyclic ortho ester linkage (PEG- a-PDMAEMA) was synthesized by atom transfer radical polymerization of DMAEMA using a PEG macroinitiator with an acid-cleavable end group. PEG- a-PDMAEMA condensed with plasmid DNA formed polyplex nanoparticles with an acid-triggered reversible PEG shield. The pH-dependent shielding/deshielding effect of PEG chains on the polyplex particles were evaluated by zeta potential and size measurements. At pH 7.4, polyplexes generated from PEG- a-PDMAEMA exhibited smaller particle size, lower surface charge, reduced interaction with erythrocytes, and less cytotoxicity compared to PDMAEMA-derived polyplexes. At pH 5.0, zeta potential of polyplexes formed from PEG- a-PDMAEMA increased, leveled up after 2 h of incubation and gradual aggregation occurred in the presence of bovine serum albumin (BSA). In contrast, the stably shielded polyplexes formed by DNA and an acid-stable block copolymer, PEG- b-PDMAEMA, did not change in size and zeta potential in 6 h. In vitro transfection efficiency of the acid-labile copolymer greatly increased after 6 h incubation at pH 5.0, approaching the same level of PDMAEMA, whereas there was only slight increase in efficiency for the stable copolymer, PEG- b-PDMAEMA. 相似文献

(HK-ZJ) isolated from the mangrove plant Aegiceras corniculatum led to isolation of six isoindolones derivatives termed as emerimidine A and B and emeriphenolicins A and D, and six previously reported compounds named aspernidine A and B, austin, austinol, dehydroaustin, and acetoxydehydroaustin, respectively. A hybrid heterojunction and solid‐state photoelectrochemical solar cell based on graphene woven fabrics (GWFs) and silicon is designed and fabricated. The GWFs are transferred onto n‐Si to form a Schottky junction with an embedded polyvinyl alcohol based solid electrolyte. In the hybrid solar cell, solid electrolyte serves three purposes simutaneously; it is an anti‐reflection layer, a chemical modification carrier, and a photoelectrochemical channel. The open‐circuit voltage, short‐circuit current density, and fill factor are all significantly improved, achieving an impressive power conversion efficiency of 11%. Solar cell models are constructed to confirm the hybrid working mechanism, with the heterojunction junction and photoelectrochemical effect functioning synergistically. 相似文献
