Melatonin (N-acetyl-5-methoxytryptamine) as a natural biostimulating substance provides a number of benefits in stimulating plant growth in stress situations due to its natural antioxidant capacity. Rhizobia also play crucial roles in supporting plant growth under environmental stress conditions. The overall goal of this research is to study the possible positive effects of melatonin and rhizobacterium in enhancing the growth and salinity tolerance of common bean. To accomplish this objective, we conducted in vitro experiment to select the optimal melatonin concentration and treatment time of seed priming for the best germination. Also, a greenhouse experiment was performed to investigate the effect of melatonin pre-treatment applied before rhizobial inoculation to improve the fitness of common bean under salinity stress. The experiment was conducted using a completely randomized factorial design with six replications and three treatments: priming treatments (melatonin priming (PM100), hydro priming (PH) and dry (PD)), salinity (0, 4, 8, 10 and 16 dS m?1) and Rhizobium strain (inoculated (RS?+) and uninoculated (RS?)). Our results showed that melatonin priming promoted bacterial colony size in Petri-dishes. The interactive effects of melatonin and RS?+?was found to alleviate reactive oxygen species (ROS) burst, and hence protect common bean chlorophylls a, b and carotenoid and photosynthetic activity and decrease malondialdehyde content through activation of antioxidant enzymes (superoxide dismutase, peroxidase, catalase and ascorbate peroxidase), facilitation of soluble protein synthesis, maintenance of Na+ and K+ homeostasis, and finally increase shoot dry weight (33.2, 39.5 and 31.5%) and seed yield (78.6, 91 and 54.2%) compared to the combination of PD and RS- treatments under 0, 4 and 8 dS m?1 salinity levels, respectively. Thus, our findings suggest that seed priming with melatonin, especially 100 µM melatonin is an effective strategy that can be used to enhance salt tolerance in common bean.

The rust resistance genes Lr53 and Yr35, transferred to common wheat from Triticum dicoccoides, were reported previously to be completely linked on chromosome 6B. Four F 3 families were produced from a cross between a line carrying Lr53 and Yr35 (98M71) and the leaf rust and stripe rust susceptible genotype Avocet “S” and were rust tested using Puccinina triticina pathotype 53-1,(6),(7),10,11 and Puccinia striiformis f. sp. tritici pathotype 110 E143 A+. The homozygous resistant lines produced infection types of “;1−” and “;N” to these pathotypes, respectively. The Chi-squared tests indicated goodness-of-fit of the data for one leaf rust gene and one stripe rust gene segregation. Linkage analysis using this population demonstrated recombination of 3% between the genes. Microsatellite markers located on the short arm of chromosome 6B were used to map the genes, with the markers cfd1 and gwm508 being mapped approximately 1.1 and 4.5 cM, respectively, proximal to Lr53. Additional studies of the relationship between Lr36, also located on the short arm of chromosome 6B, and Lr53 indicated that the two genes were independent.  相似文献

Puccinia triticina (Pt), the causal agent of leaf rust evolves through forming new pathotypes that adversely affect the growth and yield of wheat cultivars. Therefore, continued production of resistant varieties through exploring novel sources of resistance in wild relatives which are abundantly found in Iran and the neighbouring regions is a major task in wheat breeding programs. The aim of the present study was to explore 60 wild wheat genotypes selected from the species Triticum monococcum, Aegilops tauschii, Ae. neglecta, Ae. cylindrica, Ae. triuncialis, Ae. umbellulata, Ae. speltoides, Ae. columnaris, Ae. crassa and Ae. ventricosa for resistance to leaf rust. The cultivar ‘Boolani’ and Thatcher near-isogenic lines were used as controls. Two-week-old seedlings were inoculated using 10 Pt pathotypes, and the infection types were recorded. The genotypes were also analysed for polymorphism using six sequence-tagged sites (STS) and sequence characterized amplified region (SCAR) markers. Forty-eight genotypes produced high infection types (3+) for two pathotypes, but the remaining genotypes produced low infection types of ‘0; =’ to ‘1+CN’ to all pathotypes. The latter included three accessions of Ae. tauschii, two accessions of each Ae. umbellulata, Ae. columnaris and Triticum monococcum, and one accession from each Ae. triuncialis, Ae. ventricosa and Ae. neglecta. Analysis for STS and SCAR markers suggested several genotypes could carry the genes Lr9, Lr10, Lr19, Lr24, Lr26 and Lr37 or their potential orthologs in addition to unknown resistance genes. In conclusion, the identified resistant genotypes could be further characterized and used in wheat breeding programs for leaf rust resistance.  相似文献

Triticum monococcum has recently drawn the attention of biologists to discover and utilize novel genes and alleles. To explore the molecular features of the genetic network governing floral transition in shoot apical meristem (SAM) of spring growth habit T. monococcum, two expressed sequence tag (EST) libraries containing 3,031 ESTs from vegetative SAM (VS) and 2,647 ESTs from early reproductive SAM (RS) were analyzed. Assembly of ESTs resulted in 2,303 unigenes for VS library (368 contigs and 1,935 singletons) and 1,890 unigenes (337 contigs and 1,553 singletons) for RS library. The 67.05 % of VS unigenes and 66.30 % of RS unigenes showed significant similarity with genes of known, putative and or unknown function, whereas the remaining 32.95 % of the VS unigenes and 33.7 % of RS unigenes displayed no significant match with the public protein database. The 1,064 and 866 unigenes of VS and RS libraries were assigned to functional categories using Pageman ontology tool. Further analysis revealed that the switch from VS to RS caused significant changes in the abundance of unigenes assigned to some functional categories. A total of 37 genes were identified which were significantly differentially expressed between vegetative and reproductive stages of T. monococcum SAM. Investigation of the differentially expressed genes revealed the importance of the genes involved in energy metabolism, ubiquitin/26S proteasome system, polyamines biosynthesis and signaling of reactive oxygen species in SAM differentiation towards floral transition in T. monococcum.  相似文献
