O-Linked glycans were isolated from human skim milk mucins or mucin-derived high-molecular weight glycopeptides and fractionated by anion exchange chromatography into neutral and acidic alditols. Major oligosaccharides contained in the acidic fraction were purified by high performance liquid chromatography and structurally characterized by a combination of fast atom bombardment mass spectrometry, methylation analysis and 500 MHz 1H-nuclear magnetic resonance spectroscopy. The structural aspects exhibited by these major species in the acidic fraction resemble those established previously for the neutral oligosaccharides from human skim milk mucins: 1) the size of the alditols varies from tri- to decasaccharides, 2) the core structure is of the ubiquitous type 2, 3) the backbone sequences are of the poly-N-acetyllactosamine type with a particular preponderance of linearly extended GlcNAc beta(1-3)Gal (major) or GlcNAc beta(1-6)Gal units (minor). 相似文献

Novel enterobacterial core oligosaccharides were isolated from Citrobacter O4 and O36 lipopolysaccharides, and their structures were determined by methylation analysis, Smith degradation and enzymatic degradations, gas chromatography/mass spectrometry, and two-dimensional phase-sensitive correlated, relayed coherence transfer, double-quantum, triple-quantum-filtered, and nuclear Overhauser effect (NOE) 1H NMR spectroscopy at 500 MHz. In the formulas, all hexose residues are D-hexopyranoses, and heptoses are L-glycero-D-manno-heptopyranoses; the alternative locations of the side-chain heptose and pyrophosphorylethanolamine (PPEtN) residues are marked by dashed lines; dOclA stands for 3-deoxy-D-manno-octulosonic acid. A postprandial increase in ammonia nitrogen excretion and oxygen consumption rates was observed in juvenile pike fed a natural diet or an artificial dry diet. Specific growth rate of natural diet fed pike (2.4%) was lower than that of pike fed the artificial diet (3.1%). Fifty per cent of ingesta was evacuated within 5–6 h in pike of 25 mg body weight and 9–10 h in those weighing 150mg. Daily nitrogen excretion rates were related to body weight. Respiratory quotient and energy retention efficiency were affected by the nature of the diet ingested by pike. The structures of the oligosaccharides of the hemagglutinin of fowl plague virus [influenza A/FPV/Rostock/34 (H7N1)] have been elucidated by one- and two-dimensional 1H n.m.r. spectroscopy at 500 MHz and by microscale methylation analysis. N-Glycosidic oligosaccharides of the oligomannosidic (OM) and of the N-acetyllactosaminic type have been found, the latter type comprising biantennary structures, without (A) or with (E) bisecting N-acetylglucosamine, and triantennary (C) structures. Analysis of the tryptic and thermolytic glycopeptides of the hemagglutinin allowed the allocation of these oligosaccharides to the individual glycosylation sites. Each attachment site contained a unique set of oligosaccharides. Asn12 contains predominantly structures C and E which are highly fucosylated. Asn28 contains OM and A structures that lack fucose and sulfate. Asn123 shows A that has incomplete antennae but is highly fucosylated and sulfated. Asn149 has fucosylated A and E. Asn231 shows fucosylated A and E with incomplete antennae. Asn406 has OM oligosaccharides. Asn478 has A and E with little fucose. Localization of the oligosaccharides on the three-dimensional structure of the hemagglutinin revealed that the oligomannosidic glycans are attached to glycosylation sites at which the enzymes responsible for carbohydrate processing do not have proper access. These observations demonstrate that an important structural determinant for the oligosaccharide side chains is the structure of the glycoprotein itself. The depth limits for visual feeding by pollan larvae and fry, Coregonus pollan , were determined in Lough Neagh, Northern Ireland, by combining laboratory estimates of the minimum light threshold for feeding activity with field measurements of spectral, diurnal and seasonal changes in light penetration. Although feeding thresholds were lowest in the blue light (400–490 nm), the rapid attenuation of this region in the lake meant that the depth limits of feeding in situ were determined by response to the green region (490–595 nm). Effects of changes in irradiance and light penetration on the depth of feeding are considered during a seasonal study. Increased sensitivity, as the juvenile stages develop, may be offset by reduced light penetration due to algal growth. Over an annual period the maximum feeding depth for pollan varied between 2.6 and 4.1 m. In the juvenile stages of two other predatory species from the lake, perch, Perca fluviatilis , and pike, Esox lucius , the corresponding ranges were 1.1–1.9 m and 2.7–4.6 m respectively. 相似文献

Yellow perch ( Perca flavescens ) have become one of the focal points in aquaculture in the Great Lakes region of the United States. In this paper, we reviewed existing information on nutrition of yellow perch and identified critical areas in need of development. Yellow perch are small at hatching (4–7 mm total length), and are difficult to feed. Zooplankton cultures remain the only viable means of mass propagation, but larvae can be trained to accept formulated feeds in ponds at the age of 30–45 days. Few nutritional requirements exist for yellow perch, thus there are no diets formulated specifically for this species. The current recommendations are use of feeds formulated for trout and salmon. Ongoing research efforts are underway that will improve our ability to feed yellow perch at all life history stages. 相似文献
