Codon Usage in Tetrahymena and Other Ciliates   总被引:6,自引:0,他引:6  
Codon usage in ciliates was examined by analyzing the coding regions of 22 ciliate genes corresponding to a total of 26, 142 nucleotides (8, 714 codons). It was found that Tetrahymena, Paramecium and the hypotrichs ( Oxytricha and Stylonychia ) differed in which synonymous codons were used most frequently by their genes. In fact, the codon choices in highly expressed Tetrahymena genes were more similar to those of yeast genes than those of Paramecium genes. The ciliates do not appear to have unusually strong biases in codon usage frequency when compared to other protists such as yeast. The analysis of the Tetrahymena genes indicated that genes which are highly expressed during normal cell growth have a stronger bias towards using the "preferred" codons than those expressed at lower levels during growth or for brief periods during processes such as conjugation. ABSTRACT Recent work suggests that availability and quality of forage in late summer and early autumn, a time when female ungulates face multiple energetic demands, is critical to reproduction in wild ungulates. Therefore, we examined direct links between nutritional quality of diets, body condition, and reproduction of lactating mule deer. Using captive mule deer, we tested the hypothesis that females consuming diets with lower digestible energy (DE; kJ/g) would have lower DE intake rates (DEI; MJ/day), have less body fat and muscle, have later estrus cycles, and have lower pregnancy and twinning rates. Deer fed lower DE diets had lower DEI during summer and autumn. In turn, deer with lower DEI, regardless of diet DE, had lower body mass, body fat, and muscle thickness. When nutritional quality of diets began to decline earlier in the summer, relationships between food quality, DEI, and body condition were stronger. Although DEI did not influence estrus date for deer that became pregnant before 21 December, deer with lower DEI had a lower probability of becoming pregnant and had a lower probability of producing twins. Measures of body condition in October (i.e., body mass, body fat, and muscle depth) predicted pregnancy and twinning rates in mule deer. Serum concentration of hormones leptin and Insulin Growth Factor 1 were not good predictors of body condition or reproduction. These findings suggest that managers concerned with productivity of mule deer populations should consider focusing on assessing and improving quality of forage available in summer and autumn.  相似文献

Abstract: Dispersal distances and their distribution pattern are important to understanding such phenomena as disease spread and gene flow, but oftentimes dispersal characteristics are modeled as a fixed trait for a given species. We found that dispersal distributions differ for spring and autumn dispersals of yearling male white-tailed deer (Odocoileus virginianus) but that combined data can be adequately modeled based on a log-normal distribution. We modeled distribution of dispersal distances from 3 distinct populations in Pennsylvania and Maryland, USA, based on the relationship between percent forest cover and mean dispersal distance and the relationship between mean and variance of dispersal distances. Our results suggest distributions of distances for dispersing yearling male white-tailed deer can be modeled by simply measuring a readily obtained landscape metric, percent forest cover, which could be used to create generalized spatially explicit disease or gene flow models.  相似文献

Adult beetles in the small subfamily Aulacoscelidinae (superfamily Chrysomeloidea) are known to feed on the foliage and juices of New World cycads (Order Cycadales; family Zamiaceae), but the habits of larvae have long remained a mystery. We provide the first direct evidence that Aulacoscelidinae larvae feed on and develop within the megagametophyte of the Mesoamerican cycad, Dioon merolae (Zamiaceae). Phylogenetic analyses based on partial DNA sequences from 3 genes recover a cycad seed‐feeding larva proposed to belong to Aulacoscelidinae. These observations reveal a more intimate feeding relationship between Aulacoscelidinae and their New World cycad host plants than was previously recognized. Further, adult Aulacoscelidinae have long been noted to resemble Jurassic fossil chrysomeloids in the extinct subfamily Protoscelidinae. The molecular, morphological, ecological and fossil data reported herein are broadly compatible with an early association between Aulacoscelidinae and their gymnosperm hosts.  相似文献

ABSTRACT We estimated loss of butt-end leg bands on male wild turkeys (Meleagris gallapavo) captured in New York, Ohio, and Pennsylvania (USA) during December-March, 2006–2008. We used aluminum rivet leg bands as permanent marks to estimate loss of regular aluminum, enameled aluminum, anodized aluminum, and stainless steel butt-end leg bands placed below the spur. We used band loss information from 887 turkeys recovered between 31 days and 570 days after release ( = 202 days). Band loss was greater for turkeys banded as adults (>1 yr old) than juveniles and was greater for aluminum than stainless steel bands. We estimated band retention was 79–96%, depending on age at banding and type of band, for turkeys recovered 3 months after release. Band retention was <50% for all age classes and band types 15 months after banding. We concluded that use of butt-end leg bands on male wild turkeys is inappropriate for use in mark-recapture studies.  相似文献

A severe handicap in any study of the cellular biochemistry of the brain is the unavailability of sufficient amounts of pure neurones and glial cells. Many efforts to separate one from the other have been made1, but only four basic types of isolation procedures have been described for routine use: (1) brain is treated with a mixture of acetone-glycerol and water before mincing and centrifugation3–5; (2) brain mince is sieved and subjected to gradient centrifugation1,6–8—this separates neuronal cell bodies from neuropil6 or from intact but rather contaminated glial cells8; (3) whole brain is disrupted in a tissue press and zonal centrifugation is used to separate cellular and subcellular components as they leave the zonal centrifuge rotor9; and (4) chopped brain is incubated in oxygen at 37° C in the presence of 1 % (w/v) trypsin10, a treatment which, in our estimation, severely prejudices this technique's general usefulness, because it precludes meaningful comparative investigations of enzymes and non-enzymatic structural and/or soluble cell proteins. Since investigations of the latter type are the aim in our laboratory, we needed a technique in which brain is not subjected to deleterious treatments such as immersion in acetone-glycerol-water mixtures3–5 or trypsinization10 and in which no special instrumentation9, tissue presses9 or homogenizers8 are required. We now report a technique which successfully accomplishes this objective, for it makes possible the isolation of highly purified neuronal perikarya and of intact glial cells from a few grams of brain, in yields which permit the subsequent subcellular fractionation of the isolated cells. We also report two applications of the new technique which have enabled us to determine the in vivo time course of protein synthesis in and the uptake of the radioactive convulsant agent methionine sulphoximine11,12 by, the neurones and the glial cells of the immature rat brain cortex.  相似文献

ABSTRACT Behavioral studies of white-tailed deer (Odocoileus virginianus) often assign mother-offspring relationships based on common capture of juveniles with adult deer, assuming that fawns associate closely with mothers. We tested this assumption using genetic parentage to assess mother-offspring relationships within capture groups based on data from 10 polymorphic microsatellite loci. At the 80% confidence level, we assigned maternity to 43% and 51% of juveniles captured with an adult female in 2 respective study areas. Capture with their mother did not differ by sex of juveniles in either study area, and limiting our analysis to capture groups that most represent family groups (i.e., one ad F with 1–3 juv) did not increase maternity assignment (35%). Our results indicate that common capture may be a poor indicator of mother-offspring relationships in many field settings. We recommend genetic verification of family relationships.  相似文献
