Two forms of exopolygalacturonase increase as peach fruits ripen 总被引:1,自引:0,他引:1
Abstract. Freestone peach cultivars are distinguished from clingstone cultivars by a more extensive softening of the mesocarp tissue, and by the separation of mesocarp and endocarp during ripening. Cultivars of both types have been reported to develop exopolygalacturonase activity during ripening, but the enzyme has not been characterized in any detail. During development of freestone peaches ( Prunus persica L. var Coronet), two exopolygalacturonase enzymes were detected 42, 65 and 85 d after full bloom and in ripe fruit. During ripening one enzyme (exoPG 1) increased 36-fold and the other (exoPG 2) 90-fold but exoPG 2 accounted for a 73% of the total activity in ripe fruit. ExoPG 1 was purified 24-fold and exoPG 2 540-fold. ExoPG 2 is a slightly acidic glycoprotein. ExoPG 1 and exoPG 2 differ slightly in their pH optima and in their responses to calcium: each produces monogalacturonic acid as a reaction product. Cells from the sub-epidermal layer of Pilea cadierei preparedfor electron microscopy using standard fixation and embeddingprocedures appear extremely osmiophilic, with the organellesstanding out in negative relief. However, when tissues werefixed in glutaraldehyde supplemented with either procaine orcaffeine (substances known to complex with phenols) the osmiophilicmaterial was generally limited to the vacuole. After procaineor caffeine fixation the surrounding cytoplasm appeared as inadjacent, nonphenolic cells. Some small aggregates of phenolicmaterial appear in the cisternae of the endoplasmic reticular,indicating these organelles may be involved in either the synthesisor transport of phenolics in Pilea. Since one of the major functionsof phenolics is in light-screening, air blisters may serve toscreen out damaging radiation from the underlying tissue. Introduction
Ultrasonography (US) might have an added value to clinical examination in diagnosing early rheumatoid arthritis (RA) and assessing remission of RA. We aimed to clarify the added value of US in RA in these situations performing a systematic review.Methods
A systematic literature search was performed for RA, US, diagnosis and remission. Methodological quality was assessed; the wide variability in the design of studies prohibited pooling of results.Results
Six papers on the added value of US diagnosing early RA were found, in which at least bilateral metacarpophalangeal (MCP), wrists and metatarsophalangeal (MTP) joints were scanned. Compared to clinical examination, US was superior with regard to detecting synovitis and predicting progression to persistent arthritis or RA. Eleven papers on assessing remission were identified, in which at least the wrist and the MCP joints of the dominant hand were scanned. Often US detected inflammation in patients clinically in remission, irrespective of the remission criteria used. Power Doppler signs of synovitis predicted X-ray progression and future flare in patients clinically in remission.Conclusions
US appears to have added value to clinical examination for diagnosing of RA when scanning at least MCP, wrist and MTP joints, and, when evaluating remission of RA, scanning at least wrist and MCP joints of the dominant hand. Evidence indicates that small heat-shock proteins (Hsps) areinvolved in stress tolerance, but the specific cell componentsor functions that small Hsps protect or repair are mostly unidentified.We recently showed that the chloroplast small Hsps of higherplants (1) are produced in response to many environmental stresses(e.g., heat, oxidative, and high-light stress); and (2) protect(but do not repair) photosynthetic electron transport in vitroduring stress, specifically by interacting with the oxygen-evolving-complexproteins of Photosystem II (PSII) within the thylakoid lumen.However, in vivo evidence of the importance of these Hsps tophotosynthetic stress tolerance is lacking. Here we report positiverelationships between chloroplast small Hsp production and PSIIthermotolerance in (1) a heattolerant genotype of Agrostis palustris(bentgrass) and a heat sensitive genotype which lacks one ormore chloroplast small Hsps produced by the tolerant genotype;(2) ecotypes of Chenopodium album (lambs quarters) from thenorthern vs. southern U.S. (New York vs. Georgia); and (3) nineLycopersicon (tomato) cultivars/species differing in heat tolerance.These in vivo results are consistent with our previous in vitroobservations and indicate that genetic variation in productionof the chloroplast small Hsp is an important determinant ofphotosynthetic and, thereby, whole-plant thermotolerance. Recently,we showed that the mitochondrial small Hsp of plants protectsrespiratory (specifically Complex I) electron transport in vitroduring heat stress, and here we present evidence for previouslyunidentified small Hsps in mitochondria of mammal (rat) cellswhich also protect Complex I during heat stress.
