Apolipoproteins, extracted from human serum high-density lipoproteins, can be resolved and recovered with high yield from a preparative MicroPak TSK Type 3000SW size-exclusion column using Tris-buffered 6 m urea or 6 m guanidinium chloride mobile phases. Adequate resolution of some apolipoprotein pairs is only achieved at low flow velocities and low sample loads, necessitating repetitive injections of small amounts of material for preparative isolation. An analytical high-performance liquid chromatograph equipped with a simplified sample introduction scheme and low-pressure switching valves for fraction collection was used to isolate milligram quantities of HDL apolipoproteins.  相似文献

In the state of São Paulo in southeastern Brazil, a dangerous decree allows the culture of hybrids and non-native extralimital fish species in the Paraná and East Atlantic River basins. We recorded the illegal importation and sale of the non-native striped catfish Pangasianodon hypophthalmus in northeastern, central-western, southeastern, and southern areas of the country. This species is already consumed across Brazil and is cultivated for ornamental purposes. Besides escapes from fish farms and ‘fish and pays’, P. hypophthalmus can reach natural environments through aquarium dumping and ornamental ponds, motivated by its large size that reach after being sold at small size. The species has been introduced in many countries, and if established in Brazil, could put native Brazilian biodiversity and ecosystems at risk. Although importing specimens of P. hypophthalmus is prohibited by law, Decree 62.243/2016 in the state of São Paulo creates a new opportunity for invasion. In fact, once a novel fish species is moved to a new continent, it is irrational to supposed that it will stay static just in the area for which it was intended. In general, the fish disperse in the novel area on their own, by escapes from captivity or deliberate and illicit releases by persons or associations. The unwise attitude of Brazilian decision-makers and lay people in their attempt to develop aquaculture with non-native species goes against the objectives of responsible aquaculture based in native species.  相似文献

Eucalyptus globulus essential oil was evaluated for its genotoxic potential using a somatic segregation assay and a diploid strain of the fungus Aspergillus nidulans, heterozygous for nutritional and conidia color markers. The main compounds of the current essential oil sample were eucalyptol (49.0 %), α-pinene (8.9), β-pinene (1.5), globulol (6.9), α-eudesmol (1.12), spathulenol (1.42), γ-cadinene (1.45), trans-β-elemenone (1.23) and aromandendrene (2.3), totaling 74 % of oil. Oil at 0.12 and 0.25 μL/mL was found to increase the mitotic instability of the original diploid strain and the number of diploid mitotic recombinants of A. nidulans. The genotoxicity of the oil was associated with the induction of mitotic crossing-over or with oil-broken chromosomes.  相似文献

The antioxidant properties of two series of thiazolidinones and thiazinanones were reported. The novel six‐membered thiazinanones were synthesized from the efficient multicomponent reaction of 2‐picolylamine (2‐aminomethylpyridine), arenaldehydes, and the 3‐mercaptopropionic acid in moderate to excellent yields. These novel compounds were fully identified and characterized by NMR and GC‐MS techniques. In vitro antioxidant activities of all compounds were evaluated by 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH) and 2,2′‐azinobis‐3‐ethylbenzothiazoline‐6‐sulfonic acid (ABTS) tests. The antioxidant assays of thiobarbituric acid reactive species and total thiol content levels in the cerebral cortex and liver of rats were also performed. Thiazinanone 5a showed the best radical scavenging activity in DPPH and ABTS tests, as well as reduced lipid peroxidation and increased total thiol group in biological systems. Altogether, the results may be considered a good starting point for the discovery of a new radical scavenger.  相似文献

A detailed study of the presence of alpha1 AR binding sites and alpha1 AR subtype mRNA expression in human urinary bladder areas involved in the micturition (i.e. detrusor, trigone and neck) is reported here, investigating whether or not there are differences between sexes. Results obtained indicated that alpha1 AR proteins were detectable in each bladder area. In both sexes, the detrusor and the neck expressed similar levels of alpha1 ARs: respectively, detrusor: 14.6 +/- 1.2 in men and 13.1 +/- 1.1 fmol/mg prot in women; neck: 16.9 +/- 3.2 in men and 17.5 +/- 4.1 fmol/mg prot in women. In the trigone, significantly higher alpha1ARs were found in women compared to men (20.6 +/- 1.1 vs 11.7 +/- 0.7 fmol/mg prot). Subtype analysis indicated that in women, each area was endowed with mRNA encoding for each alpha1 AR subtype. The men detrusor expressed alpha1a and alpha1d ARs, while in the trigone and the neck, each subtype was present. Since the detrusor muscle hypertrophy is a marker of bladder obstructive outlet, the selective alpha1 AR subtype targeting arouses much interest, as evidence indicates that there are differences in signalling pathways among the subtypes. Furthermore, the significance of the alpha1 ARs coexpression is still unknown; interestingly, recent papers demonstrate that alpha1 AR subtypes could dimerize. Thus, in the human urinary bladder it may be suggested a potential level of alpha1 AR complexity that could have an impact on drug development.  相似文献

This study evaluated the diet and feeding selectivity of the catfish Imparfinis mirini in streams with different degrees of urbanization and the effect of rainfall on the availability of prey. The diet was based especially on Chironomidae and Trichoptera. Significant spatial differences in diet were found between the streams; the diet of the fish was similar in the rural and peri‐urban streams, and differed from that in the urban stream. Seasonality was an unimportant factor affecting the species' diet, which did not differ significantly between the rainy and dry periods in any of the streams. Fish from the urban stream fed more according to what was available in the environment, while fish from the peri‐urban and rural streams showed higher degrees of selectivity. The results indicate that environmental conditions influenced the diet and prey selection of this species in response to the differences in diversity and abundance of the food organisms. They indicate that trophic studies of benthivorous fishes, such as I. mirini, may represent an alternative way to assess human effects on streams.  相似文献

In the present study, we reported the efficient synthesis of 11 3‐(pyrimidin‐2‐yl)‐thiazolidinones in good yields using molecular sieve as the desiccant agent. In addition, we have evaluated the antioxidant capacity of the synthesized compounds by the 2,2‐diphenyl‐2‐picrylhydrazyl hydrate (DPPH?) and the 2,2‐azinobis(3‐ethylbenzothiazoline‐6‐sulfonic acid) diammonium salt (ABTS+?) radicals scavenging assay. Six compounds showed antioxidant activity towards DPPH? (EC50 between 16.13 and 49.94 µg/mL) and also demonstrated excellent activity regarding ABTS+? (TEAC: 10.32–53.52). These results showed that compounds 3‐(pyrimidin‐2‐yl)‐thiazolidinones may be easily synthesized by a less expensive procedure and could be a good starting point to the development of new antioxidant compounds. © 2013 Wiley Periodicals, Inc. This study investigated whether aquaculture facilities drive the introduction and establishment of the non-native Nile tilapia (Orechromis niloticus) in Neotropical streams, Brazil. Samples were taken from nine streams with different aquaculture occupation intensities (no, moderate, and intense) using the percentage of occupation of micro-watersheds by aquaculture ponds as a proxy for propagule pressure. The presence of aquaculture facilities and the percentage of aquaculture occupation were good predictors of the catch frequency and of densities of tilapias in the adjacent natural environment. In the streams under intense propagule pressure, females prevailed and high densities of young individuals comparing to adults were recorded. It suggests that the species was reproductively successful in the natural environment. In the streams under moderate propagule pressure, males prevailed, which indicates the likely capture of individuals escaped from aquaculture facilities. In general, our results show the positive influence of propagule pressure on the introduction and establishment of Nile tilapia in natural ecosystems, showing that aquaculture expansion of O. niloticus poses a threat to the conservation of aquatic biodiversity.

The aim of this study was describe functional faunal assemblages occurring in streams with high influence of urbanization in USA and Brazil to answer the question whether similar biological traits of fish assemblages would be found between streams with high influence of urbanization in USA and Brazil. We compiled data on the structure and composition of freshwater fishes in 46 urban streams in the USA and 10 urban streams in Brazil. Data on functional traits was established in accordance with ordinal data on abundance. Cluster analysis was conducted to examine relationships among species composition across all sites sampled and to uncover patterns in functional traits of the fish assemblages by nonmetric multidimensional scaling (NMS). The cluster analysis separated the biogeographic regions in relation to the fish fauna composition. However, the ordination analysis (NMS) did not reveal differences between functional traits in Brazil and USA urban fish assemblages, suggesting functional similarity of very disparate zoogeographic assemblages.  相似文献
