(±) Abscisic acid (ABA) injected into petioles of attached transpiring leaves of Pharbitis nil Chois. cv violet reduced the photosynthetic capacity of the mesophyll of these leaves as well as the stomatal conductance to CO2 diffusion. Greater than 75% of the injected ABA was recovered as ABA, suggesting that ABA rather than some metabolite thereof was the active compound. The nonstomatal effect of ABA increased from 30% reduction in photosynthesis at 0.25 micromolar ABA in the leaf blade to 90% reduction at 18 micromolar. Despite the effect of ABA on the nonstomatal component of leaf net CO2 uptake, it was calculated that a substantial part of the reduction in leaf net CO2 uptake (50-80%) could be accounted for by the effect of ABA on stomatal conductance.  相似文献

Dithiothreitol (DTT), an inhibitor of violaxanthin de-epoxidation and zeaxanthin formation in chloroplasts, inhibited blue-light-stimulated stomatal opening in epidermal peels of Vicia faba L. in a concentration-dependent fashion. Complete inhibition was observed at 3 mM DTT. The DTT effect was specific for the stomatal response to blue light, and the red-light-stimulated opening, which depends on photosynthetic reactions in the guard cells, was unaffected. Preirradiation of stomata in epidermal peels with increasing photon fluence rates of red light, prior to an incubation in 10 mol·m-2·s-1 of blue light and 100 mol·m-2·s-1 red light, resulted in a DTT-sensitive, blue-light-stimulated opening that was proportional to the fluence rate of the red light pre-treatment. Guard cells in epidermal peels and guard-cell protoplasts irradiated with red light showed increases in their zeaxanthin content that depended on the fluence rate of red light, or on the incubation time. The increases in zeaxanthin concentration were inhibited by DTT. Day respiration of illuminated C(3) leaves is not well understood and particularly, the metabolic origin of the day respiratory CO(2) production is poorly known. This issue was addressed in leaves of French bean (Phaseolus vulgaris) using (12)C/(13)C stable isotope techniques on illuminated leaves fed with (13)C-enriched glucose or pyruvate. The (13)CO(2) production in light was measured using the deviation of the photosynthetic carbon isotope discrimination induced by the decarboxylation of the (13)C-enriched compounds. Using different positional (13)C-enrichments, it is shown that the Krebs cycle is reduced by 95% in the light and that the pyruvate dehydrogenase reaction is much less reduced, by 27% or less. Glucose molecules are scarcely metabolized to liberate CO(2) in the light, simply suggesting that they can rarely enter glycolysis. Nuclear magnetic resonance analysis confirmed this view; when leaves are fed with (13)C-glucose, leaf sucrose and glucose represent nearly 90% of the leaf (13)C content, demonstrating that glucose is mainly directed to sucrose synthesis. Taken together, these data indicate that several metabolic down-regulations (glycolysis, Krebs cycle) accompany the light/dark transition and emphasize the decrease of the Krebs cycle decarboxylations as a metabolic basis of the light-dependent inhibition of mitochondrial respiration.  相似文献

Drought is a major abiotic stress affecting all levels of plant organization and, in particular, leaf elongation. Several experiments were designed to study the effect of water deficits on maize (Zea mays) leaves at the protein level by taking into account the reduction of leaf elongation. Proteomic analyses of growing maize leaves allowed us to show that two isoforms of caffeic acid/5-hydroxyferulic 3-O-methyltransferase (COMT) accumulated mostly at 10 to 20 cm from the leaf point of insertion and that drought resulted in a shift of this region of maximal accumulation toward basal regions. We showed that this shift was due to the combined effect of reductions in growth and in total amounts of COMT. Several other enzymes involved in lignin and/or flavonoid synthesis (caffeoyl-CoA 3-O-methyltransferase, phenylalanine ammonia lyase, methylenetetrahydrofolate reductase, and several isoforms of S-adenosyl-l-methionine synthase and methionine synthase) were highly correlated with COMT, reinforcing the hypothesis that the zone of maximal accumulation corresponds to a zone of lignification. According to the accumulation profiles of the enzymes, lignification increases in leaves of control plants when their growth decreases before reaching their final size. Lignin levels analyzed by thioacidolysis confirmed that lignin is synthesized in the region where we observed the maximal accumulation of these enzymes. Consistent with the levels of these enzymes, we found that the lignin level was lower in leaves of plants subjected to water deficit than in those of well-watered plants.  相似文献

One hundred to 120 maize recombinant inbred lines at the mature fourth leaf stage derived from F-2 and Io parental lines were grown in a glasshouse and were deprived of water for 9 days in order to detect pertinent markers of the physiological response to water stress which may be used for breeding. Carbohydrate metabolism QTLs were compared to photosynthesis gas exchange QTLs. The locations of these QTLs were further compared with those of morphological trait QTLs when water availability varied. The traits ranged from three enzyme activities (invertase, sucrose-P synthase, ADP glucose pyrophosphorylase) and hexose, sucrose, starch content to CO2 uptake and stomatal conductance, water status, leaf size, root/shoot ratio, and ABA (leaf, root and xylem sap). Four main results were obtained (1) only 14 % of QTLs were common to both drought and watered treatments, confirming the existence of stress specific chromosome regions, (2) the QTLs tended to form clusters, frequently consisting of QTLs from different classes (growth, photosynthesis, water status, carbohydrate metabolism and ABA), (3) carbohydrate metabolism trait QTLs were more frequently co-located with growth trait QTLs than photosynthesis related ones, especially in control conditions, (4) one co-location was observed between the three enzyme activities implied in sucrose and starch metabolism and a corresponding structural gene, which can be considered as a candidate gene for explaining part of the variability of each enzymatic trait (invertase, sucrose-P synthase, ADPglucose pyrophosphorylase). It is concluded that, carbohydrate metabolism provides valuable traits for understanding and improving maize responses to water stress.  相似文献

Cyclic electron flow around photosystem I drives additional proton pumping into the thylakoid lumen, which enhances the protective non-photochemical quenching and increases ATP synthesis. It involves several pathways activated independently. In whole barley leaves, P700 oxidation under far-red illumination and subsequent P700(+) dark reduction kinetics provide a major probe of the activation of cyclic pathways. Two 'intermediate' and 'slow' exponential reduction phases are always observed and they become faster after high light illumination, but dark inactivation of the Benson-Calvin cycle causes the emergence of both a transient in the P700 oxidation and a 'fast' phase in the P700(+) reduction. We investigate here the afterglow (AG) thermoluminescence emission as another tool to detect the activation of cyclic electron pathways from stroma reductants to the acceptor side of photosystem II. This transfer is activated by warming, yielding an AG band at about 45°C. However, treatments that accelerate the 'intermediate' and 'slow' P700(+) reduction phases (brief anoxia, hexose infiltration, fast dehydration of excised leaves) also produced a downshift of this AG band. This pathway ascribable to NADPH dehydrogenase (NDH) would be triggered by a deficit in ATP, while the 'fast' reduction phase corresponding to the ferredoxin plastoquinone reductase pathway is triggered by an overreduction of the photosystem I acceptor pool and is undetected in thermoluminescence. Contrastingly, slow dehydration of unwatered plants did not cause faster reduction of P700(+) nor temperature downshift of the AG band, that is no induction of the NDH pathway, whereas an increased intensity of the AG band indicated a strong NADPH + ATP assimilatory potential.  相似文献

The uptake and accumulation of exogenously supplied glycine betaine (GB) by canola (which never accumulates GB in response to stress) leaf discs has been found to induce damage to some of their structural and functional components. As a consequence some free amino acids were accumulated, particularly glutamine and glycine. Similar results were obtained with leaf discs of Arabidopsis thaliana i.e. another cruciferous plant that does not naturally produce significant amounts of GB. In contrast no changes in glutamine and glycine contents were observed in response to the GB treatment in leaf discs of spinach, a natural producer of GB. The change in glutamine content might be related to the senescing effects caused by the GB treatment. Glycine accumulation in response to GB has been more thoroughly studied with canola leaf discs. It only occurred under light conditions and was suppressed under non-photorespiratory conditions. The accumulation of glycine in canola leaf discs in response to GB was either restricted when GB was added in the presence of aminooxyacetate (an inhibitor of transaminases) or enhanced when added in the presence of aminoacetonitrile (an inhibitor of glycine decarboxylation by mitochondria). Both compounds are known to block the glycolate pathway. Glycine accumulation was not found in leaf discs of Zea mays treated in the light in the presence of GB. These results suggest that the absorbed GB could exert destabilizing effects on the photorespiration of the C3 cruciferous plants canola and Arabidopsis via competitive effects between GB and glycine at the mitochondrial step of the glycolate pathway. The mechanism of the GB effect remains to be elucidated as well as that of its apparent compatibility in spinach, the well known natural producer of GB.  相似文献

Haberlea rhodopensis is a homoiochlorophyllous resurrection plant that shows a low rate of leaf net CO2 uptake (4–6 μmol m?2 s?1) under saturating photosynthetic photon flux densities in air (21% O2 and about 390 ppm CO2). However, leaf net CO2 uptake reaches values of 17–18 μmol m?2 s?1 under saturating CO2 and light. H. rhodopensis leaves have a very low mesophyll CO2 conductance that can partly explain the low rate of leaf net CO2 uptake in normal air. Experimental evidences suggest that mesophyll conductance is not sensitive to temperature in the 20–35 °C range. In addition, it is shown that the (1) transpiration rate of H. rhodopensis is nearly linearly related to the vapour pressure difference between the leaf and the ambient air within the interval from 0.5 kPa to 2.5 kPa at a leaf temperature of 25 °C and (2) leaf net CO2 uptake in normal air under saturating light does not change much with leaf temperature (between 20 °C and 30 °C). At a leaf relative water content of between 90% and 30%, the decrease of leaf net CO2 assimilation during drought can be explained by a decrease of leaf CO2 diffusional conductance. Accordingly the non-photochemical chlorophyll fluorescence quenching decreases only at relative water contents lower than 20%, indicating that photosynthetic activity maintains a trans-thylakoidal proton gradient over a wide range of leaf water contents. Moreover, PSII photochemistry (as estimated by the Fv/Fm ratio and the thermoluminescence B band intensity) is only affected at leaf relative water contents lower than about 20%, thus confirming that primary photosynthetic reactions are resistant to drought. Interestingly, the effect of leaf desiccation on photosynthetic capacity, measured at very high ambient CO2 molar ratios under saturating PPFD, is identical to that observed for three non-resurrection C3 mesophytes. This demonstrates that the photosynthetic apparatus of H. rhodopensis is not more resistant to desiccation when compared to other C3 plants. Since the leaf area decreases by more than 50% when the leaf relative water content is reduced to about 40% during drought it is supposed, following Farrant et al. [Farrant, J.M., Vander, W.C., Lofell, D.A., Bartsch, S., Whittaker, A., 2003. An investigation into the role of light during desiccation of three angiosperms resurrection plants.
