Production of ethanol by coupling fermentation and solvent extraction   总被引:2,自引:0,他引:2  
Summary A new technology of fermentation is proposed. The inhibitor product is removed continuously by coupling fermentation and solvent extraction. Applied to ethanol fermentation this technology is suitable to any case where the terminal product is inhibitory.The proposed technology uses both plug flow reactor and liquid-liquid extraction to achieve continuously the extractive fermentation of ethanol. The solvent used for liquid-liquid extraction is dodecanol. A new reactor was used. It is a column packed with a porous material . The fermentation broth is pulsed (a) to increase the interfacial area between the liquid medium and the dodecanol, and (b) to: decrease the gas hold up.Alcoholic fermentations were performed on glucose syrup at 35°C using Saccharomyces cerevisiae, with adsorbed cells as reference, with adsorbed cells and extractive fermentation. The results show that the fermentation is substantially improved. By this new method the ethanol productivity was multiplied by 5 and a solution of 407 g/l of glucose was totally fermented with a yeast which cannot normally transform more than 200 g/l glucose.  相似文献

Dispersal and mating features strongly influence the evolutionary dynamics and the spatial genetic structure (SGS) of marine populations. For the first time in a marine invertebrate, we examined individual reproductive success, by conducting larval paternity assignments after a natural spawning event, combined with a small‐scale SGS analysis within a population of the gorgonian Paramuricea clavata. Thirty four percent of the larvae were sired by male colonies surrounding the brooding female colonies, revealing that the bulk of the mating was accomplished by males from outside the studied area. Male success increased with male height and decreased with increasing male to female distance. The parentage analyses, with a strong level of self‐recruitment (25%), unveiled the occurrence of a complex family structure at a small spatial scale, consistent with the limited larval dispersal of this species. However, no evidence of small scale SGS was revealed despite this family structure. Furthermore, temporal genetic structure was not observed, which appears to be related to the rather large effective population size. The low level of inbreeding found suggests a pattern of random mating in this species, which disagrees with expectations that limited larval dispersal should lead to biparental inbreeding. Surface brooding and investment in sexual reproduction in P. clavata contribute to multiple paternity (on average 6.4 fathers were assigned per brood), which enhance genetic diversity of the brood. Several factors may have contributed to the lack of biparental inbreeding in our study such as (i) the lack of sperm limitation at a small scale, (ii) multiple paternity, and (iii) the large effective population size. Thus, our results indicate that limited larval dispersal and complex family structure do not necessarily lead to biparental inbreeding and SGS. In the framework of conservation purposes, our results suggested that colony size, proximity among colonies and the population size should be taken into consideration for restoration projects.  相似文献

Amyotrophic Lateral Sclerosis is a fatal, progressive neurodegenerative disease characterized by loss of motor neuron function for which there is no effective treatment. One of the main difficulties in developing new therapies lies on the multiple events that contribute to motor neuron death in amyotrophic lateral sclerosis. Several pathological mechanisms have been identified as underlying events of the disease process, including excitotoxicity, mitochondrial dysfunction, oxidative stress, altered axonal transport, proteasome dysfunction, synaptic deficits, glial cell contribution, and disrupted clearance of misfolded proteins. Our approach in this study was based on a holistic vision of these mechanisms and the use of computational tools to identify polypharmacology for targeting multiple etiopathogenic pathways. By using a repositioning analysis based on systems biology approach (TPMS technology), we identified and validated the neuroprotective potential of two new drug combinations: Aliretinoin and Pranlukast, and Aliretinoin and Mefloquine. In addition, we estimated their molecular mechanisms of action in silico and validated some of these results in a well-established in vitro model of amyotrophic lateral sclerosis based on cultured spinal cord slices. The results verified that Aliretinoin and Pranlukast, and Aliretinoin and Mefloquine promote neuroprotection of motor neurons and reduce microgliosis.  相似文献

We characterize the binding affinity and the thermodynamics of hybridization of triplex-forming antiparallel purine-hairpins composed of two antiparallel purine domains linked by a loop directed toward single-stranded and double-stranded DNA (ssDNA, dsDNA). Gel retardation assays and melting experiments reveal that a 13-mer purine-hairpin binds specifically and with a K ( d ) of 8 x 10(8) M to polypyrimidine ssDNA to form a triple helical structure. Remarkably, we show that purine-hairpins also bind polypurine/polypyrimidine stretches included in a dsDNA of several hundred bp in length. Binding of purine-hairpins to dsDNA occurs by triplex formation with the polypyrimidine strand, causing displacement of the polypurine strand. Because triplex formation is restricted to polypurine/polypyrimidine stretches of dsDNA, we studied the triplex formation between purine-hairpins and polypyrimidine targets containing purine interruptions. We found that an 11-mer purine-hairpin with an adenine opposite to a guanine interruption in the polypyrimidine track binds to ssDNA and dsDNA, allowing expansion of the possible target sites and increase in the length of purine-hairpins. Thus, when using a 20-mer purine-hairpin targeting an interruption-containing polypyrimidine target, the binding affinity is increased compared to its 13-mer antiparallel purine-hairpin counterpart. Surprisingly, this increase is much more pronounced than that observed for a tail-clamp purine-hairpin extended up to 20 nt in the Watson-Crick domain only. Thus, triplexforming antiparallel purine-hairpins can be a potentially useful strategy for both single-strand and double-strand nucleic acid recognition.  相似文献

The impact of a disturbance is normally measured in terms of its effects on adult mortality, whilst the less obvious impact on reproductive biology is generally ignored. Although previous studies have contributed to understanding the effects of a mass mortality of gorgonians in 1999 at NW Mediterranean Sea, none of them have considered its impact on reproduction. The goal of this study was to determine the impact of mass mortality events on the reproduction of the gorgonian Paramuricea clavata, and examine the effect of the damage 1 year (June 2000) and 2 years (June 2001) after the event. The reproductive parameters of female colonies were more affected than those of males. In female colonies that were moderately or severely damaged, the proportion of fertile polyps decreased by about 22–35%, whilst in the worst affected males there was only a 12% decrease. Female colonies showed a progressive decrease in gonadal biomass with increasing damage to a maximum reduction of 73–75% of oocyte production observed in colonies with >33% injury. In contrast, in males, the gonadal biomass decreased rapidly for colonies with 5% of injured surface, with a reduction in spermary production of 49–64%. The same pattern of decrease in gonadal output compared to the extent of the injury was observed in 2001, two years after the mass mortality event. This indicated that the observed pattern was a response to the extent of the injury rather than a direct effect of the event. These severe effects on the reproduction of the red gorgonian species have implications for the recovery of affected populations in the long-term. This study demonstrated that an evaluation of the impact over different demographic parameters is needed to understand all the consequences of these disturbances.  相似文献

Many organisms are expanding their ranges in response to changing environmental conditions. Understanding the patterns of genetic diversity and adaptation along an expansion front is crucial to assessing a species’ long‐term success. While next‐generation sequencing techniques can reveal these changes in fine detail, ascribing them to a particular species can be difficult for organisms that live in close association with symbionts. Using a novel modified restriction site‐associated DNA sequencing (RAD‐Seq) protocol to target coral DNA, we collected 595 coral‐specific single nucleotide polymorphisms from 189 colonies of the invasive coral Oculina patagonica from the Spanish Mediterranean coast, including established core populations and two expansion fronts. Surprisingly, populations from the recent northern expansion are genetically distinct from the westward expansion and core populations and also harbour greater genetic diversity. We found that temperature may have driven adaptation along the northern expansion, as genome scans for selection found three candidate loci associated with temperature in the north but none in the west. We found no genomic signature of selection associated with artificial substrate, which has been proposed for explaining the rapid spread of O. patagonica. This suggests that this coral is simply an opportunistic colonizer of free space made available by coastal habitat modifications. Our results suggest that unique genetic variation, possibly due to limited dispersal across the Ibiza Channel, an influx of individuals from different depths and/or adaptation to cooler temperatures along the northern expansion front may have facilitated the northward range expansion of O. patagonica in the western Mediterranean.  相似文献

Environmentally friendly films exhibiting both antibacterial and antioxidative properties were elaborated from chitosan and tetrahydrocurcuminoids (THCs). Two tetrahydrocurcuminoids, THC1 (5-hydroxy-1,7-bis(4-hydroxy-3-methoxyphenyl)hept-4-en-3-one) and THC2 (5-hydroxy-1,7-bis(4-hydroxy-3,5-dimethoxyphenyl)hept-4-en-3-one), were incorporated into a chitosan film. THC1 could be prepared from natural curcumin extracted from turmeric roots (Curcuma longa L.). The resulting tetrahydrocurcuminoid–chitosan films exhibited a high free-radical scavenging activity against 2,2-diphenyl-1-picrylhydrazyl (DPPH) in methanol, which was due to a progressive release of the THCs into the solvent. The release kinetics was governed both by molecular interactions between chitosan and THCs and probably by electrostatic forces between the ammonium units in chitosan and the aromatic rings in THCs. These interactions were clearly evidenced by the presence of new absorption bands in the visible regions of the electronic absorption spectra of the THCs. The molecular nature of these interactions was shown using glucosamine, the main monomer of chitosan. When associated with THCs, chitosan retained its bioactivity against Listeria innocua; THCs alone were not bioactive enough against listerial strains.  相似文献
