Abstract: Different neurotransmitter receptor agonists [carbachol, serotonin, noradrenaline, histamine, endothelin-1, and trans -(1 S ,3 R )-aminocyclopentyl-1,3-dicarboxylic acid ( trans -ACPD)], known as stimuli of phospholipase C in brain tissue, were tested for phospholipase D stimulation in [32 P]Pi -prelabeled rat brain cortical and hippocampal slices. The accumulation of [32 P]phosphatidylethanol was measured as an index of phospholipase D-catalyzed transphosphatidylation in the presence of ethanol. Among the six neurotransmitter receptor agonists tested, only noradrenaline, histamine, endothelin-1, and trans -ACPD stimulated phospholipase D in hippocampus and cortex, an effect that was strictly dependent of the presence of millimolar extracellular calcium concentrations. The effect of histamine (EC50 18 µ M ) was inhibited by the H1 receptor antagonist mepyramine with a K i constant of 0.7 n M and was resistant to H2 and H3 receptor antagonists (ranitidine and tioperamide, respectively). Endothelin-1-stimulated phospholipase D (EC50 44 n M ) was not blocked by BQ-123, a specific antagonist of the ETA receptor. Endothelin-3 and the specific ETB receptor agonist safarotoxin 6c were also able to stimulate phospholipase D with efficacies similar to that of endothelin-1, and EC50 values of 16 and 3 n M , respectively. Several commercial polysaccharases have been compared for their ability to liberate protoplasts from fungi. These enzymes were found to contain side activities capable of hydrolysing fungal cell walls. Protoplasts have been commonly isolated from fungi using enzyme systems prepared by workers in their own laboratories. However, these procedures are time consuming and considerable variation may be found between different batches of enzyme. The present study shows that high yields of protoplasts can be prepared from a variety of fungi using relatively cheap commercial enzymes. The yields obtained were normally as good as or better than those previously produced. 相似文献

The açaí fruit depulping produces large amounts of long lignocellulosic fiber bundles that are disposed in the environment. Chemical pretreatments may improve açaí fibers favoring their usage in advanced materials. This work aimed to define optimal alkali reaction parameters to improve the properties of açaí fibers. Two NaOH concentrations (5 % and 10 %) and two reaction temperatures (80 °C and 100 °C) were tested. The raw and treated fibers were analyzed by scanning electron microscopy, Fourier transformed infrared spectroscopy, X‐ray diffraction, and thermal analyses. All the alkali pretreatments separated fibers from the bundles, unblocked pit channels by removing silicon structures, exposed the inner lignin, partially removed non‐cellulosic compounds, and raised the cellulose crystalline index. The highest temperature and NaOH content resulted in better cleaning and isolation of the fibers, while milder conditions better preserved the cellulose crystalline structure and thermal stability. 相似文献

Among the three subspecies generally recognized within the Eld's deer (Cervus eldi)--C. e. eldi, C. e. thamin, and C. e. siamensis--C. e. siamensis is considered to be particularly endangered following its disappearance from a major portion of its original range. The only captive breeding population of this subspecies is in the zoological parks at the Paris Museum of Natural History. Taking into account its low effective population size (Ne = 7) and the increasing levels of inbreeding, the continued breeding of this "micropopulation" in isolation from closely related subspecies and in particular from C. e. thamin, which is much more common in zoos as well as in the nature, is questioned. As an initial step in determining if crosses between these subspecies could be performed without risk of outbreeding depression due, in part, to gross differences in their karyotypes, a comparative chromosome banding analysis (RBG-bands) of C. e. siamensis and C. e. thamin was undertaken. No chromosomal differences were identified between the taxa at the level of resolution obtained. The study suggests that, at least from a karyotypic perspective, no obvious differences delimit the two subspecies, and hybridization between endangered C. e. siamensis and C. e. thamin is not likely to lead to impaired fertility in hybrid animals. 相似文献

Amino acid changes S180A (S-->A at site 180), H197Y, Y277F, T285A, and
A308S are known to shift the maximum wavelength of absorption (lambda max)
of red and green visual pigments toward blue, essentially in an additive
fashion. To test the generality of this "five-sites" rule, we have
determined the partial amino acid sequences of red and green pigments from
five mammalian orders (Artiodactyla, Carnivora, Lagomorpha, Perissodactyla,
and Rodentia). The result suggests that cat (Felis catus), dog (Canis
familiaris), and goat (Capra hircus) pigments all with AHYTA at the five
critical sites have lambda max values of approximately 530 nm, whereas rat
(Rattus norvegicus) pigment with AYYTS has a lambda max value of
approximately 510 nm, which is accurately predicted by the five-sites rule.
However, the observed lambda max values of the orthologous pigments of
European rabbit (Oryctolagus cuniculus), white-tailed deer (Odocoileus
virginianus), gray squirrel (Sciurus carolinensis), and guinea pig (Cavia
procellus) are consistently more than 10 nm higher than the predicted
values, suggesting the existence of additional molecular mechanisms for red
and green color vision. The inferred amino acid sequences of ancestral
organisms suggest that the extant mammalian red and green pigments appear
to have evolved from a single ancestral green-red hybrid pigment by
directed amino acid substitutions.

In Brazil capybara, the biggest existing rodent species, and associated tick species, Amblyomma cajennense and Amblyomma dubitatum, are undergoing an unplanned host and parasite population expansion in both urban and rural areas. However, scientific information
about such issue, particularly in urban areas, is scanty. Such rodent and ticks are associated in some municipalities, particularly
in southeastern Brazil, with the transmission of the highly lethal Rickettsia rickettsia caused spotted-fever. In this study ecological aspects related to the establishment and expansion of capybaras and ticks
in urban areas of Uberlandia, Minas Gerais State, Brazil were evaluated. For this purpose, capybara and tick abundance in
four urban areas and an ecological reserve was determined. Abundance of capybaras varied between areas and over the sampling
period and these differences were related to human activities. A positive correlation was found between capybara and tick
abundance, however, the tick species had an uneven distribution within the municipality and environmental factors rather than
host availability were blamed for such. On the whole these observations show that capybara populations in urban areas are
associated to high environmental infestation of ticks and the increased risk of bites and of pathogen transmission to humans.
At the same time the uneven distribution of tick species might implicate in an unequal risk of tick-borne diseases within

Background
The karyotypes of Leptodactylus species usually consist of 22 bi-armed chromosomes, but morphological variations in some chromosomes and even differences in the 2n have been reported. To better understand the mechanisms responsible for these differences, eight species were analysed using classical and molecular cytogenetic techniques, including replication banding with BrdU incorporation.Results
Distinct chromosome numbers were found: 2n = 22 in Leptodactylus chaquensis, L. labyrinthicus, L. pentadactylus, L. petersii, L. podicipinus, and L. rhodomystax; 2n = 20 in Leptodactylus sp. (aff. podicipinus); and 2n = 24 in L. marmoratus. Among the species with 2n = 22, only three had the same basic karyotype. Leptodactylus pentadactylus presented multiple translocations, L. petersii displayed chromosome morphological discrepancy, and L. podicipinus had four pairs of telocentric chromosomes. Replication banding was crucial for characterising this variability and for explaining the reduced 2n in Leptodactylus sp. (aff. podicipinus). Leptodactylus marmoratus had few chromosomes with a similar banding patterns to the 2n = 22 karyotypes. The majority of the species presented a single NOR-bearing pair, which was confirmed using Ag-impregnation and FISH with an rDNA probe. In general, the NOR-bearing chromosomes corresponded to chromosome 8, but NORs were found on chromosome 3 or 4 in some species. Leptodactylus marmoratus had NORs on chromosome pairs 6 and 8. The data from C-banding, fluorochrome staining, and FISH using the telomeric probe helped in characterising the repetitive sequences. Even though hybridisation did occur on the chromosome ends, telomere-like repetitive sequences outside of the telomere region were identified. Metaphase I cells from L. pentadactylus confirmed its complex karyotype constitution because 12 chromosomes appeared as ring-shaped chain in addition to five bivalents.Conclusions
Species of Leptodactylus exhibited both major and minor karyotypic differences which were identified by classical and molecular cytogenetic techniques. Replication banding, which is a unique procedure that has been used to obtain longitudinal multiple band patterns in amphibian chromosomes, allowed us to outline the general mechanisms responsible for these karyotype differences. The findings also suggested that L. marmoratus, which was formerly included in the genus Adenomera, may have undergone great chromosomal repatterning.8.

Mycoplasma mastitis is often difficult to control due to a lack of rapid and accurate diagnostic tools. The aim of the current study was to develop a loop-mediated isothermal amplification (LAMP) assay for the detection of Mycoplasma bovis (M. bovis) in mastitic milk. The assay was developed using primers designed for three different target genes: uvrC, 16S rRNA, and gyrB, and validated using mastitic milk samples previously found positive for the target pathogen. Specificity of the developed assay was determined by testing cross-reactivity of LAMP primers against closely related bovine mastitis bacterial pathogens. The sensitivity was found to be higher compared to conventional polymerase chain reaction (PCR). The LAMP assay was also capable of detecting M. bovis in PCR-negative milk samples of cows with clinical mastitis. The uvrC primers were found to be more sensitive, while gyrB primers were more specific; however, 16S rRNA primers were less specific and sensitive compared to either uvrC or gyrB primers. Cohen’s kappa values for uvrC, gyrB, and 16S rRNA primers used in the LAMP assays were 0.940, 0.970, and 0.807, respectively. There was a high level of agreement between the test results and the true-disease status as indicated by the receiver operating characteristic (ROC) curve. Our findings suggest that the newly developed LAMP assays targeting the uvrC and gyrB genes could be a useful tool for rapid and accurate diagnosis of mastitis caused by M. bovis. 相似文献

Tick-borne diseases (TBDs) remain to be a global animal health threat. Developing countries like the Philippines is not exempt to this. Despite the potential impact TBDs can give to these countries, local government initiatives and researches remain to be limited. In the Philippines, most epidemiological studies were confined only to specific areas, and predominantly in the Northern Area. Due to its unique geography and limited studies, the current nationwide status of most TBDs could not be clearly established. This review mainly covered published studies and presented challenges in the conduct of TBD research in the Philippines, which may be similar to other Southeast Asian or developing countries. To date, reported livestock TBD pathogens in the Philippines include Anaplasma, Babesia, Theileria, and Mycoplasma spp. With the ubiquitous presence of the Rhipicephalus microplus ticks in the country, it is highly probable that other pathogens transmitted by these vectors could be present. Despite studies on different TBDs in the livestock sector, the Philippine government has not yet heightened its efforts to implement tick control measures as part of the routine animal health program for local farmers. Further studies might be needed to determine the nationwide prevalence of TBDs and the presence of other possible tick species and TBD pathogens. The Philippine scenario may present situations that are similar to other developing countries. 相似文献
