Veillonella alcalescens contained a membrane-bound lactate oxidase system. Studies on the effect of inhibitors on lactate oxidase showed the participation of non-heme iron, quinone and cytochromesb andd. Superoxide anion radicals ( ) and H2O2 were shown to be formed at lactate oxidation and presumably arose from cyanide- and azide-resistant side chains of the respiratory system. The H+/O ratio withL-lactate as a hydrogen donor was 2.3. When an anaerobic culture growing on lactate was shifted to a high dissolved oxygen tension (d.o.t.=15 kPa) rapid inhibition of growth and lactate conversion occurred. This could be correlated with a rapid inactivation of lactate dehydrogenase. The effects of high d.o.t.'s on lactate dehydrogenase, lactate conversion and growth were reversible. After a shift to low d.o.t.'s (<2.5 kPa) growth ofV. alcalescens continued for one or two doublings whereafter lysis did occur. Acetate and pyruvate were the main fermentation products. P/O ratio's were calculated from molar growth yields and fermentation balances. A P/O value of 0.66 was found after a shift to a very low oxygen supply at which the d.o.t. presumably was zero. Shifts to higher d. o. t.'s gave much lower growth yields. Presumably, under these conditions uncoupling between growth and energy production occurred. Both coral‐associated bacteria and endosymbiotic algae (Symbiodiniaceae spp.) are vitally important for the biological function of corals. Yet little is known about their co‐occurrence within corals, how their diversity varies across coral species, or how they are impacted by anthropogenic disturbances. Here, we sampled coral colonies (n = 472) from seven species, encompassing a range of life history traits, across a gradient of chronic human disturbance (n = 11 sites on Kiritimati [Christmas] atoll) in the central equatorial Pacific, and quantified the sequence assemblages and community structure of their associated Symbiodiniaceae and bacterial communities. Although Symbiodiniaceae alpha diversity did not vary with chronic human disturbance, disturbance was consistently associated with higher bacterial Shannon diversity and richness, with bacterial richness by sample almost doubling from sites with low to very high disturbance. Chronic disturbance was also associated with altered microbial beta diversity for Symbiodiniaceae and bacteria, including changes in community structure for both and increased variation (dispersion) of the Symbiodiniaceae communities. We also found concordance between Symbiodiniaceae and bacterial community structure, when all corals were considered together, and individually for two massive species, Hydnophora microconos and Porites lobata, implying that symbionts and bacteria respond similarly to human disturbance in these species. Finally, we found that the dominant Symbiodiniaceae ancestral lineage in a coral colony was associated with differential abundances of several distinct bacterial taxa. These results suggest that increased beta diversity of Symbiodiniaceae and bacterial communities may be a reliable indicator of stress in the coral microbiome, and that there may be concordant responses to chronic disturbance between these communities at the whole‐ecosystem scale.  相似文献

Protecting and promoting recovery of species at risk of extinction is a critical component of biodiversity conservation. In Canada, the Committee on the Status of Endangered Wildlife in Canada (COSEWIC) determines whether species are at risk of extinction or extirpation, and has conducted these assessments since 1977. We examined trends in COSEWIC assessments to identify whether at-risk species that have been assessed more than once tended to improve, remain constant, or deteriorate in status, as a way of assessing the effectiveness of biodiversity conservation in Canada. Of 369 species that met our criteria for examination, 115 deteriorated, 202 remained unchanged, and 52 improved in status. Only 20 species (5.4%) improved to the point where they were ‘not at risk’, and five of those were due to increased sampling efforts rather than an increase in population size. Species outcomes were also dependent on the severity of their initial assessment; for example, 47% of species that were initially listed as special concern deteriorated between assessments. After receiving an at-risk assessment by COSEWIC, a species is considered for listing under the federal Species at Risk Act (SARA), which is the primary national tool that mandates protection for at-risk species. We examined whether SARA-listing was associated with improved COSEWIC assessment outcomes relative to unlisted species. Of 305 species that had multiple assessments and were SARA-listed, 221 were listed at a level that required identification and protection of critical habitat; however, critical habitat was fully identified for only 56 of these species. We suggest that the Canadian government should formally identify and protect critical habitat, as is required by existing legislation. In addition, our finding that at-risk species in Canada rarely recover leads us to recommend that every effort be made to actively prevent species from becoming at-risk in the first place.  相似文献

This paper seeks to understand why multinationals prefer to launch a label specific to their own product and examines how reliable these product-specific eco-labels are. A new methodology is applied to assess the extent to which eco-labels live up to claims about their contribution to conservation and the sustainable use of agricultural biodiversity. Product-specific eco-labels are considered as industry self-regulation and all three regulatory stages are studied: the planning, implementation and outcome stage. There are major differences between the product specific eco-labels in the degree in which agrobiodiversity management is part of the normative labeling schemes. Although there are some problems of reliability, such as transparency in the implementation stage and the monitoring in the outcome stage, the degree of reliability of product-specific labels is comparable with eco-labels of international labeling families. The conclusion is that only one of the product-specific eco-labels examined here is reliable when examined in the light of all three stages. The main reason why multinationals establish a product-specific eco-label instead of adopting one from an existing labeling family is that they want to profile themselves as distinct from other companies. The unique character of a product-specific label creates a market opportunity for them.  相似文献

Because dominant competitors can monopolize resources, any positive effects they have on other species can have large community impacts. The solitary tunicate Ascidia ceratodes is a dominant competitor in the fouling community in Bodega Harbor, CA. This tunicate preempts primary substratum from competitors, but its thick tunic also allows other species to grow on its surface. The net effect of Ascidia on the community as a whole therefore depends on the balance between competitive and facilitative effects. In this study we evaluate the facilitative effects of Ascidia on different life stages of common competing species. We quantified larval settlement onto Ascidia compared to unoccupied space; we quantified the growth rate on Ascidia of small colonies of two common species; and we measured whether established colonies could escape overgrowth by Ascidia by growing onto its tunic. We found evidence for high rates of settlement on Ascidia, with some species showing higher and others lower settlement relative to that observed on free space. The growth rate of settlers was generally lower on Ascidia compared to primary substratum. We also found that colonial species established on primary space commonly escape overgrowth from Ascidia by growing onto Ascidia, as this occurs in about half of all encounters. This study indicates that the total effect of Ascidia on the community will depend on species-specific and life stage-specific effects, with likely long-term consequences for the diversity and composition of the community.  相似文献

MORC1 and MORC2, two of the seven members of the Arabidopsis (Arabidopsis thaliana) Compromised Recognition of Turnip Crinkle Virus1 subfamily of microrchidia Gyrase, Heat Shock Protein90, Histidine Kinase, MutL (GHKL) ATPases, were previously shown to be required in multiple layers of plant immunity. Here, we show that the barley (Hordeum vulgare) MORCs also are involved in disease resistance. Genome-wide analyses identified five MORCs that are 37% to 48% identical on the protein level to AtMORC1. Unexpectedly, and in clear contrast to Arabidopsis, RNA interference-mediated knockdown of MORC in barley resulted in enhanced basal resistance and effector-triggered, powdery mildew resistance locus A12-mediated resistance against the biotrophic powdery mildew fungus (Blumeria graminis f. sp. hordei), while MORC overexpression decreased resistance. Moreover, barley knockdown mutants also showed higher resistance to Fusarium graminearum. Barley MORCs, like their Arabidopsis homologs, contain the highly conserved GHKL ATPase and S5 domains, which identify them as members of the MORC superfamily. Like AtMORC1, barley MORC1 (HvMORC1) binds DNA and has Mn2+-dependent endonuclease activities, suggesting that the contrasting function of MORC1 homologs in barley versus Arabidopsis is not due to differences in their enzyme activities. In contrast to AtMORCs, which are involved in silencing of transposons that are largely restricted to pericentromeric regions, barley MORC mutants did not show a loss-of-transposon silencing regardless of their genomic location. Reciprocal overexpression of MORC1 homologs in barley and Arabidopsis showed that AtMORC1 and HvMORC1 could not restore each other’s function. Together, these results suggest that MORC proteins function as modulators of immunity, which can act negatively (barley) or positively (Arabidopsis) dependent on the species.The evolution of a complex defense system has been the consequence of plants being constantly exposed to pathogenic microbes and pests. One of the first lines of active defense is based on a perception of pathogen-associated molecular patterns (PAMPs) by pattern recognition receptors located in the plant cell membrane. The defense response to PAMP recognition is called PAMP-triggered immunity (PTI). While PTI is sufficient to stop colonization by many microbes, some microorganisms overcome this immune response by releasing effectors (formerly called virulence factors). In a coevolutionary process, some plants have evolved resistance (R) proteins for direct or indirect recognition of microbial effectors (avirulence [Avr] factors) leading to effector-triggered immunity (ETI). ETI is frequently characterized by a rapid and locally restricted programmed cell death response (also known as hypersensitive reaction [HR]), which helps to limit pathogen proliferation and disease symptoms. On the contrary, the absence of an Avr-R protein interaction results in virulence of the pathogen. In addition, ETI is counteracted by some microbes by the release of additional virulence factors that block or overcome effector recognition and ensure pathogenicity. The mutual evolution of host and microbe leading to elicitation or suppression of ETI is summarized by the “zigzag” model proposed by Jones and Dangl (2006). PTI and ETI are associated with activation of various defense responses both at infection sites and in distal tissue, including production and accumulation of reactive oxygen species, salicylic acid, and pathogenesis-related proteins. Systemic activation of such responses, triggered in the uninfected tissue, leads to long-lasting, broad-based resistance to subsequent pathogen infections, termed systemic acquired resistance.A genetic screen in Arabidopsis (Arabidopsis thaliana) searching for mutants with compromised resistance mediated by the R protein HR to Turnip Crinkle Virus (HRT) against Turnip Crinkle Virus (TCV) led to the discovery of the Compromised Recognition of TCV1 (CRT1) subfamily of the microrchidia (MORC) subclade of the GHKL (for Gyrase, Heat Shock Protein90, Histidine Kinase, MutL) ATPase superfamily (Watson et al., 1998; Iyer et al., 2008; Kang et al., 2008). Genome analysis of Arabidopsis revealed that MORC1 (formerly named CRT1 in Kang et al., 2008, 2010, 2012) has two close (>70% sequence similarity on amino acid [aa] level) and four distant (<50% aa similarity) homologs. A double knockout mutant, morc1-2 morc2-1, lacking MORC1 and its closest homolog MORC2 also displayed compromised ETI to avirulent Pseudomonas syringae, suppressed basal resistance, systemic acquired resistance, and/or PTI to TCV and virulent P. syringae and compromised nonhost resistance to Phytophthora infestans (Kang et al., 2012). Arabidopsis MORC1 physically interacts with at least eleven R proteins belonging to three different structural classes (Martin et al., 2003), including HRT, the R protein involved in recognition of TCV. This interaction is a dynamic process, as MORC1 bound inactive R proteins, while little or no interaction was observed when the R proteins were activated (Kang et al., 2010). Taken together, these results argued that MORC1 protein family members in Arabidopsis are key components in multiple layers of resistance against a variety of pathogens. Recently, it was shown that a small fraction of AtMORC1 translocates to the plant nucleus after ETI and PTI activation (Kang et al., 2012). Because Arabidopsis MORC1 possesses DNA/RNA-binding capacity and endonuclease activity in vitro, these findings suggest a potential role in DNA recombination and repair (Kang et al., 2012). In addition, three recent independent studies identified Arabidopsis MORC1 and its homolog MORC6 (also named Defective in Meristem Silencing11) as novel factors involved in gene silencing and/or chromatin superstructure remodeling in response to epigenetic signals (Lorković et al., 2012; Moissiard et al., 2012; Brabbs et al., 2013).Given that the CRT1 subfamily of MORC ATPases is involved in multiple layers of disease resistance against various pathogens, these genes may have relevance for agronomic applications. To assess whether MORCs are involved in crop plant resistance and thus could be exploited in breeding strategies, MORC1 homologous genes were identified in the model cereal crop barley (Hordeum vulgare). We show here that all five barley MORCs, discovered in the not yet fully annotated barley genome, are involved in resistance to agronomically important diseases. Unexpectedly, however, and in clear contrast to Arabidopsis, barley plants silenced for MORC genes were more resistant, while overexpression compromised resistance to infections by both biotrophic and necrotrophic fungal pathogens. Moreover, reciprocal overexpression in Arabidopsis and barley showed that AtMORC1 and HvMORC1 homologs are not functionally interchangeable.  相似文献

This article explores the multifactorial relationship between mastication and cognition, with a focus on dementia. Older persons, especially those with dementia, are at great risk of suffering from oral health problems such as orofacial pain and loss of natural teeth. A possible explanation could be that the cognitive and motor impairments resulting from dementia cause a decrease in self‐care and as such, a worsening of oral health. An alternative explanation is that cognition and oral health influence each other. Animal studies show that a decrease in masticatory activity, for example, due to a soft diet or loss of teeth, causes memory loss and neuronal degeneration. The relationship between mastication and cognition has also been researched in human studies, but a cause‐effect relationship has not been proven. It is likely that multiple factors play a role in this relationship, such as self‐care, nutrition, stress and pain.  相似文献

A mark‐recapture study based on the Petersen method was implemented in 1998 to estimate the abundance of the invasive common carp, Cyprinus carpio L., in Lake Crescent, Tasmania. Multiple gear types were employed to minimise capture bias, with multiple capture and recapture events providing an opportunity to compute and compare Petersen and Schnabel estimates. A single Petersen estimate on recapture data and two Schnabel estimates – one each on mark (forward‐Schnabel estimate) and recapture (reverse‐Schnabel estimate) data – were conducted. An independent long‐term double tag study facilitated estimation of the annual natural mortality. Subsequent fish‐down of the population suggests that, in all likelihood, the carp have been eradicated from the lake, providing an unprecedented opportunity to verify the forward population estimates carried out in 1998. Results suggest that all three estimates were close to the true population size, with the reverse‐Schnabel estimate being the most accurate and within 1% of the true population in this relatively large lake (~2365 ha). Greater accuracy of the reverse‐Schnabel approach can be attributed to either minimised fish behavioural (i.e. gear susceptibility or avoidance) or computational bias associated with the forward‐Schnabel and Petersen approaches, respectively. While the original estimates served as a guide in eradication of carp from the lake, the ultimate validation provides a reliable framework for abundance estimation of this invasive fish in relatively large water bodies elsewhere.  相似文献
