Cloning and functional characterization of LCR-F1: a bZIP transcription factor that activates erythroid-specific,human globin gene expression. 下载免费PDF全文
DNase I hypersensitive site 2 (HS 2) of the human beta-globin Locus Control Region (LCR) directs high level expression of the beta-globin gene located 50 kilobases downstream. Experiments in cultured cells and in transgenic mice demonstrate that duplicated AP1-like sites in HS 2 are required for this powerful enhancer activity. A cDNA clone encoding a basic, leucine-zipper protein that binds to these sites was isolated and designated Locus Control Region-Factor 1 (LCR-F1). This protein is a member of a new family of regulatory factors that contain a 63 amino acid ''CNC domain'' overlapping the basic region. This domain is approximately 70% identical in the Drosophila Cap N Collar (CNC) protein, NF-E2 and LCR-F1. LCR-F1 transactivates an HS 2/gamma-globin reporter gene over 170-fold in transient transfection experiments specifically in erythroid cells. These results suggest that LCR-F1 may be a critical factor involved in LCR-mediated, human globin gene expression. 相似文献

3-O-Methylfunicone (OMF) is a secondary metabolite produced by the soil fungus Penicillium pinophilum which has cytostatic properties. The aim of this study was to investigate the mechanisms by which such properties are exerted, with special reference to any anti-proliferative and apoptotic potential, on HeLa cells. OMF treatment caused about 44% inhibition of cell growth after 24 h, and modifications in the tubulin fibre organization. In addition, a significant increase in p21 mRNA expression and a decrease in cyclin D1 and Cdk4 mRNA expression resulted at the same time. Apoptosis induction was demonstrated by the annexin V assay, cytofluorimetric analysis of the DNA content of the sub-G1 fraction and DNA laddering. Taken together, our data showed that the compound inhibits proliferation of HeLa cells by several mechanisms, such as disruption of tubulin fibres, cell cycle arrest and apoptosis induction. The capacity of the compound to affect the cell cycle and to modulate apoptosis is indicative of a potential for the development of a new agent for cancer chemotherapy. 相似文献

Humic acids (HAs) from soil and compost at the beginning (S0) and at the end of the stabilization process after 130 days (S130) have been fractionated by coupling size exclusion chromatography (SEC) and polyacrylamide gel electrophoresis (PAGE). Preparative quantities of HA fractions (HAFs) with different molecular sizes (MSs) and exactly defined electrophoretic mobility (EMs) have been obtained from all samples and the HAFs weight content has been studied. A high degree of similarity in HAFs weight content between soil HA and a stabilized compost HAs130 has been observed. Such data seem to be reliable for monitoring the evolution of the compost organic matter to humic substances for their agricultural uses. 相似文献

The agronomic performance and the mineral composition and trace element content in Begonia semperflorens "Bellavista F1", Mimulus "Magic x hybridus", Salvia splendens "maestro", and Tagete patula xerecta "Zenith Lemon Yellow", were tested by growing the plants on substrates of white peat and 25-50-75-100% green waste and sewage sludge (80%+20%v/v) compost (CP). A commercial peat medium of black and white peat (2:1v/v) was used as control. At flowering, the agronomic parameters were compared by ANOVA and plant nutritional status was compared by vector analysis. Substrate-species interactions (P<0.001) were evident for all measured parameters. In the 25% CP medium all the species showed an increase or preservation of the studied agronomic parameters. Begonia grown in 25% CP, showed the highest dry weight (DW) and number of flowers. Other treatments were comparable to the control. Mimulus and Salvia showed the highest DW in the 25-50% CP. Mimulus, after a DW increase up to 50% CP, showed the steepest reduction as the CP increased further. Tagete showed no differences in DW up to 50% CP, or in flower number up to 25% CP, compared to the control. The additional increases of CP in the medium showed a DW decrease similar to that of Salvia. Vector analysis showed the use of compost mainly induced a decrease of P concentration in tissues, except for Begonia which remained unchanged. Plant tissues showed a general P reduction due to a dilution effect in the low compost mixtures (25-50%) and a deficiency in the higher CP mixtures. In contrast, an increase of Mg in the aboveground tissues of all species was detectable as compost usage increased, with the exception of Salvia which suffered a Mg deficiency. Vector analysis also highlighted a Ni and partial Fe deficiency in Tagete and Salvia. 相似文献

Objectives: Cancer stem cells make up a subpopulation of cells within tumours that drive tumour initiation, growth and recurrence. They are resistant to many current types of cancer treatment, causing failure of such therapeutic approaches, including chemotherapy and radiotherapy. In the study described here, anti‐proliferative effects of 3‐O‐methylfunicone (OMF), a metabolite from Penicillium pinophilum, were investigated on human breast cancer MCF‐7 cells and cancer stem cells selected as mammospheres derived from MCF‐7s. Materials and methods: Stemness markers were analysed on isolated mammospheres showing positive expression of CD24, CD29, CD44, CD133, CD184 and CD338. Cell proliferation and apoptosis were analysed by flow cytometry and RT‐PCR. Cell colony formation assays were performed to evaluate colony formation of mammospheres. Results and conclusion: OMF treatment affected both MCF‐7 and mammosphere growth, inducing apoptosis. In addition, OMF strongly reduced stemness markers and survivin, hTERT and Nanog‐1 gene expression. Growth of colonies in soft‐agar was significantly affected by OMF treatment, too. Lastly, we tested ability of MCF‐7 cells to form mammospheres after treatment with OMF or cisplatin, demonstrating that OMF treatment resulted in drastic reduction in number of mammospheres. These results introduce OMF as an effective molecule in suppressing breast cancer stem cells. 相似文献

The lipophilic extracts of two marine aeolid nudibranch molluscs of the genus Spurilla collected in distinct geographical areas have been chemically analyzed. The Et2O extracts of the nudibranchs were dominated by the presence of usual fatty acids and sterols and contained terpenoid compounds 1 – 3 as minor metabolites. Spurillin A ( 1 ) and spurillin B ( 3 ) were new molecules whereas cis‐γ‐monocyclofarnesol ( 2 ) was already reported in the literature as a synthesis product. Interestingly, bursatellin ( 4 ), previously isolated from anaspidean molluscs of the genus Bursatella, was found in the butanol extract of both Spurilla species. Compounds 1 – 4 were not detected in the extracts of the sea‐anemone preys collected together with the molluscs. 相似文献
