A species of Erwinia was found to produce no other pectolytic enzyme than the two transeliminases of exo-types, namely, an oligogalacturonide transeliminase and an exopectic acid transeliminase. Of the two enzymes, the exopectic acid transeliminase was isolated and its properties were investigated. The results are as follows: (1) Pectic acids having an unsaturated galacturonic acid residue at the non-reducing end of the molecule are susceptible but oxidized or reduced pectic acids resistant to the enzyme action. (2) The enzyme has no activity toward pectinic acid and polymethylpolygalacturonate methyl glycoside. The limit of the enzymatic degradation for citrus pectic acid is 43.8%. (3) The rate of the enzyme activity was maximal with tetragalacturonic acid and followed by acid-soluble pectic acid, acid-insoluble pectic acid, pectic acid and trigalacturonic acid. Unlike the oligogalacturonide transeliminases of Pseudomonas sp. (strain S2) and Erwinia aroideae, the present enzyme shows a considerably high activity toward pectic acids of high molecular weight. (4) The pH-activity curves vary with the buffer employed. (5) The enzyme is activated by Co2+ and Mn2+ but powerfully inhibited by Cu2+ and Hg2+. Acinetobacter sp. strain YAA has five genes (atdA1 to atdA5) involved in aniline oxidation as a part of the aniline degradation gene cluster. From sequence analysis, the five genes were expected to encode a glutamine synthetase (GS)-like protein (AtdA1), a glutamine amidotransferase-like protein (AtdA2), and an aromatic compound dioxygenase (AtdA3, AtdA4, and AtdA5) (M. Takeo, T. Fujii, and Y. Maeda, J. Ferment. Bioeng. 85:17-24, 1998). A recombinant Pseudomonas strain harboring these five genes quantitatively converted aniline into catechol, demonstrating that catechol is the major oxidation product from aniline. To elucidate the function of the GS-like protein AtdA1 in aniline oxidation, we purified it from recombinant Escherichia coli harboring atdA1. The purified AtdA1 protein produced gamma-glutamylanilide (γ-GA) quantitatively from aniline and l-glutamate in the presence of ATP and MgCl2. This reaction was identical to glutamine synthesis by GS, except for the use of aniline instead of ammonia as the substrate. Recombinant Pseudomonas strains harboring the dioxygenase genes (atdA3 to atdA5) were unable to degrade aniline but converted γ-GA into catechol, indicating that γ-GA is an intermediate to catechol and a direct substrate for the dioxygenase. Unexpectedly, a recombinant Pseudomonas strain harboring only atdA2 hydrolyzed γ-GA into aniline, reversing the γ-GA formation by AtdA1. Deletion of atdA2 from atdA1 to atdA5 caused γ-GA accumulation from aniline in recombinant Pseudomonas cells and inhibited the growth of a recombinant Acinetobacter strain on aniline, suggesting that AtdA2 prevents γ-GA accumulation that is harmful to the host cell.  相似文献

The three-dimensional solution structures of human lysozyme were determined at 35 and 4 degrees C using the heteronuclear multidimensional NMR spectroscopy, which were compared with each other to clarify the structural response of this enzyme to lowering of the temperature. Together with the data of the temperature dependence experiments of the lytic activity against Micrococcus luteus, we consider the implication of the observed structural change for the low-temperature-induced reduction of the activity of human lysozyme. The structures of human lysozyme determined at the two temperatures are found to be similar, both of which comprise four alpha-helices (A- to D-helices) and three antiparallel beta-strands (beta(1)-beta(3)), leading to the constructions of the alpha- and beta-domains as previously identified in the X-ray crystal structure. A significant structural change was observed for the "active site lobe" comprising the loop region connecting C- and D-helices and the following D-helix, which moves toward the active site cleft located between the alpha- and beta-domains so as to obstruct the cleft according to the temperature lowering. It further appeared that the total volume as well as the accessible surface area of human lysozyme decreases with lowering of the temperature, suggesting that the internal cavity of this enzyme shrinks under low temperature environment. Because in human lysozyme the region comprising the active site lobe is responsible for turnover of the enzymatic reaction against the substrate, the low-temperature-induced structural change of the active site lobe presumably controls the efficiency of the lytic activity under low temperatures.  相似文献

Diosgenin[(25R)-spirost-5-en-3 beta-ol] and cholesterol were detected as trimethylsilyl derivatives by gas chromatography-mass spectrometry in an alkaline lysate of a new hypotensive phospholipid from the lipid fraction of bovine brain. The ionic concentrations,conductivity and pH of water in the Lake Chany complex in West Siberian Russia change from the mouth to the interior of the lake. This difference is indicative of marked evaporation of lake water from the closed water body system in the dry climate of Western Siberia. The carbon isotope composition of particulate organic matter (POM, composed mainly of phytoplankton) clearly changes, along with the pH of the water, reflecting the concentration of dissolved CO2. Carbon and nitrogen isotope signatures of Chironomus plumosus larvae, a benthic invertebrate that may feed on bulk lake sediment, systematically increase, along with those of POM and sediment organic matter (SOM), through the lake chain. Both sulfate-sulfur and nitrogen isotope compositions of the POM and SOM increase with distance from the estuary into the Lake Chany complex. Heavier sulfur and nitrogen isotope recycling from the sediment, caused by microbial sulfate reduction and denitrification, respectively, may have led to the increased sulfate-sulfur and nitrogen isotope compositions of the POM and SOM.  相似文献

Microbiological leaching of synthetic cobaltous sulfide (CoS) was investigated with a pure strain of Thiobacillus ferroxidans. The strain could not grow on CoS-salts medium in the absence of ferrous ions (Fe2+). However, in CoS-salts medium supplemented with 18 mM Fe2+, the strain utilized both Fe2+ and the sulfur moiety in CoS for growth, resulting in an enhanced solubilization of Co2+. Cell growth on sulfur-salts medium was strongly inhibited by Co2+, and this inhibition was completely protected by Fe2+. Cobalt-resistant cells, obtained by subculturing the strain in medium supplemented with both Fe2+ and Co2+, brought a marked decrease in the amount of Fe2+ absolutely required for cell growth on CoS-salts medium. As one mechanism of protection by Fe2+, it is proposed that the strain utilizes one part of Fe2+ externally added to CoS-salts medium to synthesize the cobalt-resistant system. Since a similar protective effect by Fe2+ was also observed for cell inhibition by stannous, nickel, zinc, silver, and mercuric ions, a new role of Fe2+ in bacterial leaching in T. ferrooxidans is proposed.  相似文献

Senescence-accelerated mouse prone 6 (SAMP6) is a model of senile osteoporosis. From 10 to 22 wk of age, SAMP6 mice were heavier than age-matched AKR/J and SAMR1 mice. Body mass indices of 10- and 25-wk-old SAMP6 mice were higher than those of age-matched AKR/J and SAMR1 mice, indicating obesity in the SAMP6 animals. We compared the blood biochemical values among SAMP6, SAMR1, and AKR/J mice to assess whether the SAMP6 strain has abnormal obesity-related parameters. Plasma glucose, triglyceride, insulin, and leptin levels were higher in 10-wk-old SAMP6 mice than in age-matched SAMR1 and AKR/J mice, whereas plasma glucagon and adiponectin levels in 25-wk-old SAMP6 were lower compared with those in age-matched SAMR1 and AKR/J. Total cholesterol levels in SAMR1 and SAMP6 mice at 10 and 25 wk of age were higher than those in AKR/J mice. Hepatic lipid levels were higher in 10- and 25-wk-old SAMP6 mice compared with age-matched AKR/J and SAMR1 animals. These results indicate that SAMP6 mice exhibit obesity and hyperlipidemia, suggesting that the SAMP6 strain is a potential tool for the study of hyperlipidemia.Abbreviations: BMI, body mass indexThe senescence-accelerated mouse strains were developed through selective breeding of AKR/J mice based on graded scores for senescence and pathologic phenotypes.44 The 9 senescence-prone (SAMP) strains all have a shortened lifespan and display an early onset of senescence after normal development and maturation, whereas the 3 senescence-resistant (SAMR) strains are resistant to early senescence and serve as controls. Among the SAMP strains, SAMP8 and SAMP10 exhibit deficits in learning and memory at a relatively early stage in their lifespan.6,30 In contrast, SAMP6 mice are considered to be a model of senile osteoporosis, with their low bone mass and slow bone loss;24 the bone mineral density of SAMP6 mice decreases after 4 mo of age.14,17Our regular measurement of body weight revealed that SAMP6 mice were significantly higher between 10 and 22 wk of age than were age-matched SAMR1 and AKR/J. Based on this observation, we decided to compare body mass indices (BMIs), blood biochemical values, and liver sections among mice of these strains at 10 and 25 wk of age, which respectively correspond to the beginning and end of a period of significant body weight gain in SAMP6 mice compared with age-matched SAMR1 and AKR/J. Increased BMIs of SAMP6 mice at 10- and 25 wk compared with those of age-matched AKR/J and SAMR1 animals would indicate obesity in the SAMP6. In addition, because osteoblasts and adipocytes are thought to share a common precursor cell, osteoporosis and enhanced adipogenesis may be related. For example, adipogenesis in the bone marrow increases with aging and during osteoporosis,15,33,34 and increased bone turnover occurs in hypercholesterolemic or dyslipidemic patients.22 Therefore obesity in SAMP6 mice might be due at least in part to enhanced adipogenesis. We measured and compared blood biochemical values among SAMP6, SAMR1, and AKR/J (the founder for the SAM strains) mice to assess whether the SAMP6 strain has abnormalities in blood biochemical markers, such as triglycerides or cholesterol.  相似文献

Erwinia exopolygalacturonate lyase is strongly activated by Na+, but very weakly by divalent cations such as Ca2+, in contrast to most of the known pectic lyases; nevertheless, this enzyme is completely inhibited by 1 mM EDTA. In this work, six polyamino carboxylates such as EDTA and a polyamine were examined for their effects on the enzyme activity.

EDTA, trans-1,2-cyclohexanediaminetetraacetate, and diethylenetriaminepentaacetate inhibited the enzyme strongly, but ethylenediaminediacetate showed little inhibition. Only tri-ethylenetetramine stimulated the enzyme. The removal of EDTA from the enzyme solution resulted in an almost complete restoration of the activity. The EDTA-treated enzyme as well as the untreated one revealed no requirement for any divalent cations. The inactivation by hydroxyethylenediaminetriacetate, a mild inhibitor, was the mixed type. It seems most likely that the inactivation by the polyamino carboxylates is caused not by sequestering any metals but by directly forming an enzyme–chelator complex.  相似文献
