Five experimental grass paddocks were sequentially contaminated with fresh bovine faeces containing known numbers of eggs of predominantly Haemonchus and Cooperia spp. during the 1984/85 dry season (November to March). Faecal, herbage and soil samples were examined at regular intervals between November and June in order to determine the rate of development and mortality of infective larvae (L3) in faeces, the pattern of herbage infestation with L3 and the role of faeces and soil as reservoirs of L3 during the dry season and early rains. L3 first appeared in faeces approximately 4 days post contamination (PC) and peak counts were obtained 28, 14, 7 and 14 days PC in the paddocks contaminated in December, January, February and March, respectively. The counts initially declined linearly at the rate of approximately 7535 L3/week and 10,947 L3/week in P2 and P4 respectively, due primarily to mortality but later there was an accelerated fall in the counts as the surviving L3 moved out of the faecal pads onto herbage. The overall trend of faecal larval populations in each paddock was therefore distinctly curvilinear. Although large numbers of L3 were present inside dry faecal pads in most paddocks throughout the dry season, none migrated on to herbage at that time of the year. Translation of L3 to herbage was very rapid and occurred simultaneously in all the paddocks 24 hours following the first heavy rainfall in late March. Consequently peak herbage infestations in all paddocks were coincident and occurred a few days after commencement of larval migration. The closer to the end of the dry season the contaminations were carried out, the larger were the subsequent early rains rise and the peak herbage infestation and the longer this infestation survived on herbage. No L3 were recovered from soil throughout the study, which suggests that faecal pads were the sole reservoir of L3 during the dry season and hence the source of the early rains herbage infestation.  相似文献

The aim of this study was to determine the first effect of lead on microbial activity in soil. The study was carried out in the soil samples from four different radish (Raphanus sativus L. var. radicula, Brassicaceae) fields along the highway in a district (Kadirli, Osmaniye) of the Eastern Mediterranean Region, Turkey. After the calculation of Pb contents, the Pb amounts of the soil samples were brought up to 50 and 100 mg Pb kg?1 by treatment with Pb(NO 3 ) 2 , and the samples for the carbon and the nitrogen mineralization were incubated under controlled conditions (28°C, constant moist). The carbon mineralization was determined by a CO 2 respiration method for 30 days. The nitrogen mineralization was observed in vitro for 6 weeks. The untreated group was statistically different from the 50 and 100 mg Pb kg?1 treatments in the aspect of the C(CO 2 ) outlet during mineralization (P ≤ 0.05), but difference between the 50 and 100 mg Pb kg?1 treatments was not significant. NH 4 -N and NO 3 -N contents of each soil were shown differences between across treatments. Based on these results, it is possible to conclude that the addition of 50 and 100 mg Pb kg?1 provided a toxic effect threshold for the microbial activity into 30 days.  相似文献

Twelve fungi namelyAlternaria alternata, Aspergillus flavus, A niger, A ochraceus, Actinomucor repens, Capnodoium spp., Curvularia lunata, Fusarium pallidoroseum, F solani, F verticillioides, Penicillium citrinum and Rhizopus stolonifer were recorded from samples ofAegle marmelos, Aesculus indica, Buchanania lanzan andPinus gerardiana. In case ofPrunus amygdalus only Rstolonifer was recorded. A significant variation in pattern of mycoflora incidence was observed in terms of source and season. Fungal infestation in most of the substrates was found to be highest during monsoon. Aflatoxins were the most common mycotoxins elaborated by different isolates ofA flavus obtained fromA marmelos, B lanzan andP gerardiana. The amount of aflatoxins produced by the toxigenic isolates ofA flavus was in the range of traces to 0.9–26.0 μg/ml inA marmelos, 0.8–17.5 μg/ml inP gerardiana and 0.65–13.2 μg/ml inB lanzan. The percentage toxigenicity was comparatively lower in the isolates of other mycotoxigenic fungi. Aflatoxins were detected almost in all the samples analyzed for mycotoxin contamination. However, traces of zearalenone were detected inA marmelos. The concentration of aflatoxin B1 was in the range of 0.13–0.75 μg/g inA marmelos, 0.09–0.60 μg/g inP gerardiana and 0.01–0.20 ug/g inB lanzan. The West African Dwarf (WAD) goat from the humid zone of Nigeria is known for its trypanotolerance as well as for its resistance and resilience to Haemonchus contortus (haemonchotolerance). Another ecotype of WAD goat with a larger body size is found in the drier savanna zone of the country. We tested the hypothesis that the latter is less trypanotolerant, and less haemonchotolerant than the former ecotype because they have been less exposed to these infections and because of the likelihood of introgression of alleles for parasite susceptibility into the latter from neighbouring parasite-susceptible Sahelian genotypes. Two controlled experiments were carried out. In the first, we compared the responses in 8–9 month old kids of both ecotypes to subcutaneous infection with 5 × 106 Trypanosoma brucei. Infection in both ecotypes was characterised by (i) prepatent periods of 3 days; (ii) a modest peak parasitaemia 4–5 days post infection (pi), followed by rapid clearance of parasites from the blood to microscopically undetectable levels from D11 or D12 until the end of the experiment on D30 pi; (iii) a sharp but transient drop in PCV following peak parasitaemia, with no other clinical evidence of anaemia; and (iv) normal growth and a small but weakly significant change in body temperature. In a second experiment we infected groups of goats of both ecotypes with 6000 L3 of H. contortus. This infection also produced no significant changes in the PCV and body weight of the goats. Only a small percentage of the inoculum was recovered from both ecotypes at necropsy on D18 pi (Mean % recovery ± SE = 3.29 ± 0.61 for humid zone and 6.83 ± 2.72 for savanna goats) and there was no significant difference in their worm burdens. On the basis of these results we reject our hypothesis and conclude that the savanna WAD ecotype exhibits comparable, strong degrees of trypanotolerance and haemonchotolerance to its humid zone counterpart.  相似文献

Concurrent infection with Trypanosoma brucei (Tb) delays the normal protective responses of mice to the gastrointestinal parasite Nippostrongylus brasiliensis (Nb). The course of such infections was followed in mice genetically deficient in inducible nitric oxide synthase (INOS) to assess the role of nitric oxide (NO) in this effect. The time course of trypanosome infection in INOS deficient (INOS-/-) mice was similar to that in wild type (WT) and heterozygote (INOS+/-) mice but did not result in NO production. Although concurrent infection with Tb increased initial susceptibility to Nb in INOS-/- mice, the immune-mediated loss of N. brasiliensis and the associated decline in faecal egg output occurred more rapidly then in WT and INOS+/- littermates. Concurrent infection with trypanosomes markedly suppressed Concanavalin A (ConA)-induced in vitro proliferation of splenic lymphocytes in all groups, but had little effect on the responses of mesenteric node lymphocytes. Trypanosome infection was also associated with increased early release of interferon-gamma and reduced IL-5 from lymphocytes stimulated in vitro with ConA, but did not affect later release of IL-5. The overall similarity of proliferative and cytokine responses in WT, INOS+/- and INOS-/- mice suggest that the suppressive effects of T. brucei on N. brasiliensis infection do not simply reflect depressed lymphocyte responsiveness or altered cytokine profiles. NO appears to be involved in suppression only of the later phases of the host responses to Nb.  相似文献
