An in vitro assay system that used erythrocytes was developed to assess the toxicity of delta endotoxin of Bacillus thuringiensis subsp. israelensis (Bti), a mosquito larvicidal agent. This endotoxin was activated at high alkaline pH without causing alkaline injury to the erythrocytes. The assay is carried out on blood agarose plates, prepared in MOPS-buffered saline (0·01 mol 1-1), pH 7·0. The threshold dose for toxicity of Bti HD-567 was 8 ng protein and that of Bti H-14 and IPS-82 standard were 6 ng and 4 ng protein respectively. The assay is rapid, sensitive, quantitative and applicable to different Bti formulations to assess the toxicity of the products. Only the activated toxin is detected by this method and has good correlation with insect bioassay. The percentage toxicity dose of different Bti preparations agrees well with percentages of LC50 values when the second instar larvae of Aedes aegypti were tested.  相似文献

Respirometric studies with mitochondrial, fractions and whole cells revealed the presence of a more actively functioning respiratory system inAspergillus sydowii grown under salinity conditions. Oxidation of substrate, i.e., succinate, by the mitochondrial fraction was inhibited by the addition of rotenone, antimycin A, and cyanide. Electron microscopic observations ofAsp. sydowii grown in the presence of 2M NaCl indicated a comparatively larger size of mitochondria than in the control grown culture. A relatively larger fraction of the total cytoplasmic volume was occupied by the mitochondria in theAsp. sydowii grown in the media containing 2M NaCl. Levels of respiratory enzymes like succinate dehydrogenase. NADH dehydrogenase, cytochrome oxidase, NADH oxidase, and succinoxidase were higher in the culture grown in the presence of 2 M NaCl than in that grown in the absence of NaCl.  相似文献

In the present study, a high chitinase producing strain Pantoea dispersa was isolated from the sea dumps at Bhavnagar, India. Chitin, urea, CaCl2 and MgSO4 x 7H2O were variables used in central composite design for chitinase production. Chitinase, biomass and pH were the responses used in different models to evaluate individually fit ones. Quadratic model was found to be fit for chitinase response whereas in the case of biomass and pH, linear model was found to be fit without the effect of others. Chitinase production was optimized with respect to other responses such as biomass and pH in multiresponse analysis of response surface design by using desirability approach. In multiresponse analysis, following medium formulation (g/l), chitin, 15; urea, 0.32; CaCl2, 0.10 and MgSO4 x 7H2O, 0.08 was found to predict optimum chitinase production of 482.77 units/ml with overall highest desirability of 0.854 as compared to other formulations. The selection of model was done on the basis of high Adjusted R-squared value and lowered p-value for each model in individual analysis of each response. In multiresponse experiment, it was found that for response chitinase quadratic model and for responses pH and biomass linear models were well fit. Through desirability analysis, it was found that in the chitinase production, pH was essential as compared to biomass in P. dispersa. Endochitinase and chitobiase actvities were also studied.  相似文献

AIMS: To develop a novel, rapid and effective screening method for chitinase producing bacteria. METHODS AND RESULTS: A simple and rapid technique for screening of potential chitinolytic bacteria has been developed using the chitin binding dye calcofluor white M2R in chitin agar. Microorganisms possessing high chitinolytic potential gave a clear zone under ultraviolet light after 24-48 h of incubation. This method was successfully applied for isolating the hyperchitinase mutant of Alcaligenes xylosoxydans. The mutant Alc. xylosoxydans EMS 33 was found to produce 3.4 times more chitinase than the wild type. CONCLUSIONS: In this study, the screening method for chitinase producing bacteria has been developed and it was applied to screen chitinase-overproducing mutant of Alc. xylosoxydans. SIGNIFICANCE AND IMPACT OF THE STUDY: The novel screening method for chitinase producer is more sensitive, rapid, user-friendly and reliable, which can also be used for screening of recombinants having chitinase gene.  相似文献

Studies with whole cells and mitochondrial fractions revealed increased respiratory activity inAspergillus repens grown under salt stress conditions. The state 3 and state 4 respiration rates, PO ratios, and Mg2+-dependent ATPase were higher in mitochondria of stressed cells than in control cells.A. repens respires via an antimycin A-and cyanide-sensitive pathway. Oligomycin, dicyclohexylcarbodiimide (DCCD) and rotenone inhibited respiration rates less in mitochondria of stressed cells than in controls. Though 2,4-dinitrophenol (DNP), carbonyl cyanide-m-chlorophenylhydrazone (m-Cl-CCP), and carbonyl cyanide-p-trifluoromethylhydrazone (p-F3-CCP) did not stimulate Mg2+-ATPase activity, DNP enhanced the respiration rates, whereasm-Cl-CCP andp-F3-CCP decreased the respiration rates in either condition; mitochondria of stressed cells exhibited a lower degree of inhibition than controls. Addition of DNP, oligomycin, and DCCD inhibited the basal Mg2+-ATPase (ATPase activity without Mg2+ addition). Oligomycin inhibited the Mg2+-ATPase. DCCD showed less inhibition in mitochondria under stress than did the controls. Levels of some respiratory enzymes were higher in the culture grown under stress than in the controls.  相似文献

Cladosporium sphaerospermum, isolated from salt pans was halotolerant. When grown in the presence of salt, the activities of invertase, isocitrate lyase, fructose-1,6 diphosphate aldolase and malate dehydrogenase were found to be increased and that of amylase decreased. Both, enzyme activation as well as an increase inde novo synthesis of enzymes were found to be some of the mechanisms of salt mediated changes. This may be one of the adaptive mechanisms, in halotolerantCladosporium sphaerospermum.  相似文献

Aspergillus repens, a salt-pan isolate, was halotolerant. When grown for 72 h (log phase) and 144 h (beginning of stationary phase) in a medium containing 2m sodium chloride, the activities of invertase, malate dehydrogenase (MDH), glucose-6-phosphate dehydrogenase (G6PDH), and glutamate dehydrogenase (GDH) were found to have increased. Control cultures grown in a medium devoid of 2m NaCl failed to show such changes. The activities of MDH, G6PDH, and GDH increased with rising concentrations of Na+ (as NaCl) when added up to 100mm in vitro. At higher concentrations they decreased. Changes in kinetic constants, Km and Vmax of these enzymes, as well as their de novo synthesis, were found to be some of the responses to NaCl stress-mediated changes.  相似文献

Summary Optimization of medium composition and pH for chitinase production by the Alcaligenes xylosoxydans mutant EMS33 was carried out in the present study and the optimized medium composition and conditions were evaluated in a fermenter. The medium components screened initially using Plackett–Burman design were (NH4)2SO4, MgSO4 7H2O, KH2PO4, yeast extract, Tween 20 and chitin in shake flask experiments. The significant medium components identified by the Plackett–Burman method were MgSO4 7H2O, Tween 20 and chitin. Central composite response surface methodology was applied to further optimize chitinase production. The optimized values of MgSO4 7H2O, Tween 20, chitin and pH were found to be 0.6 g/l, 0.05 g/l, 11.5 g/l and 8.0, respectively. Chitinase and biomass production of Alcaligenes xylosoxydans EMS33, was studied in a 2-l fermenter containing (g/l): chitin, 11.5; yeast extract, 0.5; (NH4)2SO4, 1; MgSO4 7H2O, 0.6; KH2PO4, 1.36 and Tween 20, 0.05. The highest chitinase production was 54 units/ml at 60 h and pH 8.0 when the dissolved O2 concentration was 60%, whereas the highest biomass production was achieved at 36 h and pH 7.5 without any dissolved O2 control.  相似文献

Alcaligenes xylosoxydans protected pigeonpea from Fusarium wilt in a pot experiment and field trials. When seeds of pigeonpea (C. cajan) were treated with A. xylosoxydans and sown in soil infested with Fusarium, the incidence of wilt was reduced by 43.5% and resulted in 58% higher grain yield. The antifungal activity of A. xylosoxydans was based on chitinase production and was comparable in efficacy to commercial antifungal agents such as benlate, monitor WP, thiram and bavistin.  相似文献
