Studies have been made on the lipid composition of total lipids, triglycerides and their fatty acids, cholesterol and phospholipids in the vertebral column of young and adult rabbits. It was shown that the content of total lipids and triglycerides increases, whereas that of cholesterol and phospholipids decreases with age. The content of total lipids in the vertebral column is 10 times higher as compared to that in the bones of the extremities. Mid-thoracic part of the vertebral column exhibits higher lipid content than other thoracic parts of the column. Lipid content of the vertebral processes is lower than that of the vertebral bodies. These data indicate lipid specificity and heterogeneity of bone tissue of the vertebral column. The main fatty acids of vertebral triglycerides are presented by those with 14-18 carbon atoms (90%), no acids with 22 atoms were found. Higher content of the linoleic acid (19%) and higher total unsaturation of triglycerides were found in the bone tissue of rabbits in comparison with those of man.  相似文献

Dysbacteriosis of the large intestine is one of severe complications of long-term use of antituberculosis agents in the treatment of respiratory tract tuberculosis that results in a significant decrease of tolerability of antituberculosis agents, persistence of tuberculosis intoxication and slower involution of the tuberculosis process in the lungs. When the complex treatment with antituberculosis agents was accompanied by the use of cow's kumiss for correction of the large intestine dysbacteriosis, the intoxication signs disappeared in 12% of the patients in the main group, while in the patients of the control group the level of the intoxication syndrome increased twice. The rate of the tuberculosis lesions regression evident from the lung roentgenograms was 2.7-fold higher in the main group vs. the control (62 and 23% respectively). The indices of the lung functional capacity recovery in the patients of the main group vs. the control were also higher (41 and 33% respectively). Hepatic toxic reactions in the patients not given cow's kumiss for correction of dysbacteriosis were 8 times more frequent vs. the control. The results of the study made it possible to develop recommendations for phthisiologists in the use of cow's kumiss as one of the methods of pathogenetic therapy in complex treatment of patients with respiratory tract tuberculosis in sanatoria.  相似文献

Studies have been made on lipid composition of the gills in 19 species of fishes and of the lungs in 8 mammalian species. It was shown that the arbitrary "unit fragment" of the lipid component of the respiratory organs in all the investigated animals exhibits similar qualitative composition, but contains different quantity of lipid molecules, i. e. approximately 19 in fishes and 42 in mammals. Theoretical conformational analysis revealed predominant conformations of phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine, sphingomyelin and cholesterol; projections of these molecules on the surface which is parallel to membrane surface were composed. It was shown that projectional areas for the main phospholipid molecules differ only by 2-3%. These data indicate that the surface of a model of the lipid component in the membranes of respiratory organs in mammals is twice larger than in fishes. These differences presumably account for more effective oxygen transfer in mammals.  相似文献

The binding of pig skeletal muscle lactate dehydrogenase by F-actin has been studied using the sedimentation method in 10 mM Tris-acetate buffer, pH 6.0 at 20 degrees C. Adsorption capacity of F-actin is equal to (1 +/- 0.1) . 10(-5) moles of lactate dehydrogenase per 1 g of actin. NADH decreases the affinity of F-actin with respect to lactate dehydrogenase. The binding of lactate dehydrogenase by F-actin in diminishing the rate of enzymatic reduction of alpha-ketoglutarate. The microscopic dissociation constant for the complex of the enzyme with F-actin which is estimated from the dependence of the enzymatic reaction rate of F-actin concentration at saturating NADH concentrations is equal (3.0 +2- 0.5) . 10(-7) M. It has been shown that the bound enzyme is characterized by the greater value of Km and the lower value of Vmax in comparison to the free enzyme.  相似文献

Cyclase-associated proteins (CAPs) are among the most highly conserved regulators of actin dynamics, being present in organisms from mammals to apicomplexan parasites. Yeast, plant, and mammalian CAPs are large multidomain proteins, which catalyze nucleotide exchange on actin monomers from ADP to ATP and recycle actin monomers from actin-depolymerizing factor (ADF)/cofilin for new rounds of filament assembly. However, the mechanism by which CAPs promote nucleotide exchange is not known. Furthermore, how apicomplexan CAPs, which lack many domains present in yeast and mammalian CAPs, contribute to actin dynamics is not understood. We show that, like yeast Srv2/CAP, mouse CAP1 interacts with ADF/cofilin and ADP-G-actin through its N-terminal α-helical and C-terminal β-strand domains, respectively. However, in the variation to yeast Srv2/CAP, mouse CAP1 has two adjacent profilin-binding sites, and it interacts with ATP-actin monomers with high affinity through its WH2 domain. Importantly, we revealed that the C-terminal β-sheet domain of mouse CAP1 is essential and sufficient for catalyzing nucleotide exchange on actin monomers, although the adjacent WH2 domain is not required for this function. Supporting these data, we show that the malaria parasite Plasmodium falciparum CAP, which is entirely composed of the β-sheet domain, efficiently promotes nucleotide exchange on actin monomers. Collectively, this study provides evidence that catalyzing nucleotide exchange on actin monomers via the β-sheet domain is the most highly conserved function of CAPs from mammals to apicomplexan parasites. Other functions, including interactions with profilin and ADF/cofilin, evolved in more complex organisms to adjust the specific role of CAPs in actin dynamics.  相似文献

Separate and combined exposure of roach Rutilus rutilus embryos during the first 38 h after fertilization to magnetic field (500 Hz, 1.4–1.6 µT) and elevated temperature (23°?) causes morphological responses: decrease in mean values of length and weight of the body, increase in the number of vertebrae, and increase in the number of vertebral anomalies and morphological diversity of vertebral phenotypes. Size–weight characteristics and the number of vertebrae in underyearlings are related to the development of anomalies in its backbone. A method for counting of vertebrae in fish with backbone malformations is suggested.  相似文献

The thermal stability of rabbit skeletal muscle glycogen phosphorylase b was characterized using enzymological inactivation studies, differential scanning calorimetry, and analytical ultracentrifugation. The results suggest that denaturation proceeds by the dissociative mechanism, i.e., it includes the step of reversible dissociation of the active dimer into inactive monomers and the following step of irreversible denaturation of the monomer. It was shown that glucose 1-phosphate (substrate), glucose (competitive inhibitor), AMP (allosteric activator), FMN, and glucose 6-phosphate (allosteric inhibitors) had a protective effect. Calorimetric study demonstrates that the cofactor of glycogen phosphorylase-pyridoxal 5'-phosphate-stabilizes the enzyme molecule. Partial reactivation of glycogen phosphorylase b preheated at 53 degrees C occurs after cooling of the enzyme solution to 30 degrees C. The fact that the rate of reactivation decreases with dilution of the enzyme solution indicates association of inactive monomers into active dimers during renaturation. The allosteric inhibitor FMN enhances the rate of phosphorylase b reactivation.  相似文献

The oligomeric state and formation of supramolecular structures of glycogen phosphorylase b from rabbit skeletal muscle was studied in the system of aerosol OT (AOT) reversed micelles in octane. The sedimentation experiments have shown that the enzyme oligomeric state depends on the degree of micelle hydration. The enzyme monomer, dimer, trimer, tetramer, hexamer, and octamer were observed, depending on the degree of hydration.  相似文献
