Since their introduction, artificial lipid bilayer membranes were used in a wide array of applications, such as sensors, biocompatible materials and study-models of the cell's outer boundary. Here, we present a nanohybrid membrane using an inorganic host and amphiphilic organic molecules with lipid bilayer-like properties. The stability of the presented mimetic membrane is significantly improved when compared to existing methods. The nanohybrid membrane exhibited two thermotropic phases corresponding to the L(alpha) and L(beta) phases that lipid bilayer membranes are known to adopt. Integration of cholesterol molecules into the nanohybrid membrane lead to the same qualitative effects as in lipid bilayers, including expansion of the bilayer spacing and decrease of the L(alpha) to L(beta) transition enthalpy. To further illustrate the similarities of the synthesized membrane with a lipid bilayer, the ability of the nanohybrid membrane to function as saccharin conductimetric sensor was evaluated. The lower limit of detection of the sensor was 6 microM and the linear range of response was from 20 to 400 microM.  相似文献

The intestine is highly sensitive to ischemia/reperfusion (I/R) injury. Intestinal I/R may cause local tissue injury and disruption of the intestinal mucosal barrier, allowing the passage of viable bacteria and endotoxins from the gastrointestinal lumen to distant organs. This phenomenon, known as bacterial translocation (BT), may lead to systemic disorders with high morbidity and mortality. Oxidative stress mediators such as reactive oxygen species, polymorphonuclear neutrophils and nitric oxide are believed to contribute to the intestinal I/R injury. Many antioxidants have shown protective effects against I/R injury of various organs. The present article provides an overview of studies investigating the effect of antioxidant supplementation on BT after intestinal I/R.  相似文献

Atherosclerosis is intimately coupled to blood flow by the presence of predilection sites. The coupling is through mechanotransduction of endothelial cells and approximately 2000 gene are associated with this process. This paper describes a new platform to study and identify new signalling pathways in endothelial cells covering an atherosclerotic plaque. The identified networks are synthesized in primary cells to study their reaction to flow. This synthetic approach might lead to new insights and drug targets.  相似文献

The number of functional teats is an important selection criterion in pig breeding. Inherited defects of the udder, such as the inverted teat, do have a considerable negative impact on the nursing ability of the sow. To investigate the genetic background of this defect and the number of functional teats in Swedish maternal lines, samples from 230 Yorkshire pigs were selected for genotyping using the PorcineSNP60K BeadChip (Illumina Inc.), each pig with at least one inverted teat was matched with one non-affected pig (fullsib or pairs with matching herd and gender). A genome-wide association study on these 230 pigs was performed using the two-step approach implemented in GenABEL using 46,652 single nucleotide polymorphisms across all autosomes and the X chromosome. A number of significant regions were identified for the inverted teat defect on chromosomes 2, 10, and 18. Many of the regions associated with the number of functional teats were located in the same or close regions, except two associated markers on the X chromosome and one on chromosome 3. We identified some of the regions on chromosomes previously reported in one linkage and one gene expression study. We conclude, despite being able to suggest new candidate genes, that further studies are needed to better understand the biologic background of the teat development. Despite the in-depth comparison of identified regions for the inverted teat defect done here, more studies are required to allow a clear identification of genetic regions relevant for this defect across many pig populations.  相似文献
