Summary Tumor cells injected into Balb/c mice together with heat-killed 48-h P. acnes cells were rendered nontumorigenic as early as 12 h after injection, as determined by the inability of the tumor cells to give rise to tumors when transferred to a new host. Determination of tumor cell antigen levels by ELISA indicated that the tumor antigens had virtually disappeared by 24 h after injection of tumor cells and P. acnes. In contrast, in control animals injected with tumor cells only, there was an initial drop in tumor antigen levels at 12 h, after which the level rose steadily and tumors developed in 7–10 days. Since the cellular exudate at 12 h was almost entirely composed of polymorphonuclear leukocytes (PMN), we tested the ability of PMN, stimulated by phagocytosis of 48-h P. acnes cells, to produce substances toxic to tumor cells. Results indicated that the supernatant fluid from a phagocytosis mixture of PMN and P. acnes contained material toxic to tumor cells and also to Chinese hamster ovary cells. Tests with scavengers and inhibitors of oxygen-derived radicals suggested that the toxic material is either hydrogen peroxide (H2O2) or hydroxyl radicals (OH). Suspensions of 12-h P. acnes, P. acnes cells walls, P. freudenrichii, or latex beads were ineffective in preventing tumor growth, and induced little toxicity when phagocytosed. We conclude that in this test system 48-h P. acnes prevents tumor growth by stimulating the production of toxic oxygen metabolites during phagocytosis by PMN.  相似文献

Stereospecific analyses of glycerolipids from 7-, 14- and 21-day-old cultures of the yeast Lipomyces lipoferus revealed that each position of the glycerolipids had a unique distribution of fatty acids which changed to varying degrees with age, and that, in the triacylglycerols, age had a greater effect on fatty acid content at sn-3 that at sn-1 or sn-2. Age-related changes in unsaturation were, however, greater in the phospholipids than in the triacylglycerols. Among the major phospholipids of L. lipoferus (phosphatidylcholine, phosphatidylinositol and phosphatidylethanolamine), changes in the proportion of unsaturated to saturated fatty acids, and in the number of double bonds per mole, were greater at sn-2 than at sn-1, except for phosphatidylinositol between 14 and 21 days of age. The pattern of acylation of phosphatidylinositol between 14 and 21 days was thus different from that of phosphatidylcholine and phosphatidylethanolamine. Furthermore, at the three ages investigated, phosphatidylinositol had low and relatively constant levels of unsaturation compared with phosphatidylcholine and phosphatidylethanolamine. The net decrease in phospholipid double bonds per mole in aging cells of L. lipoferus, and previous data, suggest that aging in this yeast is accompanied by a decrease in membrane fluidity.  相似文献

The selection of an adequate exposure assessment approach is imperative for the quality of epidemiological studies. The use of personal exposimeters turned out to be a reasonable approach to determine exposure profiles, however, certain limitations regarding the absolute values delivered by the devices have to be considered. Apart from the limited dynamic range, it has to be taken into account that these devices give only an approximation of the exposure due to the influence of the body of the person carrying the exposimeter, the receiver characteristics of the exposimeter, as well as the dependence of the measured value on frequency band, channel, slot configuration, and communication traffic. In this study, the relationship between the field strength measured close to the human body at the location of the exposimeter and the exposure, that is, the field strength at the location of the human body without the human body present, is investigated by numerical means using the Visible Human model as an anatomical phantom. Two different scenarios were chosen: (1) For FM, GSM, and UMTS an urban outdoor scenario was examined that included a transmitting antenna mounted on the roof of one of four buildings at a street crossing, (2) For WLAN an indoor scenario was investigated. For GSM the average degree of underestimation by the exposimeter (relation of the average field levels at the location of the exposimeter to the field level averaged over the volume of the human body without the body present) was 0.76, and for UMTS 0.87; for FM no underestimation was found, the ratio was 1. In the case of WLAN the degree of underestimation was more pronounced, the ratio was 0.64. This study clearly suggests that a careful evaluation of correction factors for different scenarios is needed prior to the definition of the study protocol. The newly described toxic dinoflagellate Pfiesteria piscicida is a polymorphic and multiphasic species with flagellated, amoeboid, and cyst stages. The species is structurally a heterotroph; however, the flagellated stages can have cleptochloroplasts in large food vacuoles and can temporarily function as mixotrophs. The flagellated stage has a typical mesokaryotic nucleus, and the theca is composed of four membranes, two of which are vesicular and contain thin plates arranged in a Kofoidian series of Po, cp, X, 4′, 1a, 5″, 6c, 4s, 5″′, and 2″″. The plate tabulation is unlike that of any other armored dinoflagellate. Nodules often demark the suture lines underneath the outer membrane, but fixation protocols can influence the detection of plates. Amoeboid benthic stages can be filose to lobose, are thecate, and have a reticulate or spiculate appearance. Amoeboid stages have a eukaryotic nuclear profile and are phagocytic. Cyst stages include a small spherical stage with a honeycomb, reticulate surface and possibly another stage that is elongate and oval to spherical with chrysophyte-like scales that can have long bracts. The species is placed in a new family, Pfiesteriaceae, and the order Dinamoebales is emended.  相似文献

Desoxyribosenucleic acid molecules isolated from salmon sperm were studied with the electron microscope. The essential step in the technique which makes it possible to visualize the individual molecules consists in a preparative step wherein the materials are supported on the extremely smooth surface of cleaved mica where they are shadow-cast with platinum, which is then backed with a supporting film and stripped for observation in the usual manner. The DNA, which was originally about 8 million molecular weight, was also examined after fragmentation by sonic vibration. The fragments show a certain degree of rigidity and the ends generally terminate abruptly, indicating that the double helices of the Watson-Crick model both break close to the same place. DNA molecules heated to temperatures between 90 and 100°C, coil up into amorphous patches, although a few apparently unaltered molecules survive such heating.  相似文献
