Quantitative and qualitative distribution of surface phytoplankton,as related to hydrographic conditions, was studied in the Gulfof Naples in February 1979. Previous work has shown that the Gulf of Naples is a diversifiedecosystem, due to geographic and hydrographic features as wellas man made eutrophication, that can be subdivided into twomajor parts: a coastal subsystem and an open water one. Hydrographic analysis of the winter situation at the surfacefully confirms this picture, as it identifies two distinct watermasses corresponding respectively to surfaced Tyrrhenian IntermediateWater and to Coastal Surface Water. The structural analysisof phytoplankton reveals three assemblages of species characterizingdifferent water types: 1 - the Ischia and Procida channels affectedby the advection of Volturno river and Cuma outfall plumes;2 - the coastal area of the Gulf proper, namely the bays ofPozzuoli, Naples and Castellammare; 3 - the open waterhemislocated beyond the 100 m isobath. The channel area assemblage is dominated by diatoms, particularlyby fast growing species, such as Asterionella japonica, severalspecies of Chaetoceros and Rhizosolenia hebetata f. semispina.The coastal assemblage is identified, among others, by the diatomsCerataulina bergonii, Hemiaulus sinensis; the dinoflagellatesGlenodinium lenticula, Exuviaella compressa and Porella perforata.The open water assemblage is characterized by the diatoms Coscinodiscuscurvatulus and Hemidiscus cuneiformis, the dinoflagellate Amphidiniumacutissimum and the coccolithophore Coccolithus haeckelii.  相似文献

The occurrence and abundance of the toxic, chain-forming dinoflagellateGymnodinium catenatum in a Tyrrhenian coastal lagoon, the Fusaro,during an annual sampling cycle are reported. Peak abundanceswere observed from late spring until early autumn Although veryhigh cell numbers were recorded, up to 1 5 x 106 cells l–1,no monospecific bloom of this species occurred. The first observationof G.catenatum in the Mediterranean occurred in the Fusaro andthe appearance of this species in a traditional shellfish farmingarea, where no shellfish intoxication has been reported to date,is discussed in relation to human interventions in the basin.In particular, intensive dredging in recent years with resuspensionof bottom sediments may have seeded the water body with cysts.A Gymnodinium n d species, illustrated using scanning electronmicroscopy, caused a monospecific bloom in concomitance withmaximum abundances of G.catenatum, apparently outcompeting thislatter species  相似文献

Leishmania mexicana (Lm) causes localized (LCL) and diffuse (DCL) cutaneous leishmaniasis. DCL patients have a poor cellular immune response leading to chronicity. It has been proposed that CD8 T lymphocytes (CD8) play a crucial role in infection clearance, although the role of CD8 cytotoxicity in disease control has not been elucidated. Lesions of DCL patients have been shown to harbor low numbers of CD8, as compared to patients with LCL, and leishmanicidal treatment restores CD8 numbers. The marked response of CD8 towards Leishmania parasites led us to analyze possible functional differences between CD8 from patients with LCL and DCL. We compared IFNγ production, antigen-specific proliferation, and cytotoxicity of CD8 purified from PBMC against autologous macrophages (MO) infected with Leishmania mexicana (MOi). Additionally, we analyzed tissue biopsies from both groups of patients for evidence of cytotoxicity associated with apoptotic cells in the lesions. We found that CD8 cell of DCL patients exhibited low cytotoxicity, low antigen-specific proliferation and low IFNγ production when stimulated with MOi, as compared to LCL patients. Additionally, DCL patients had significantly less TUNEL+ cells in their lesions. These characteristics are similar to cellular "exhaustion" described in chronic infections. We intended to restore the functional capacity of CD8 cells of DCL patients by preincubating them with TLR2 agonists: Lm lipophosphoglycan (LPG) or Pam3Cys. Cytotoxicity against MOi, antigen-specific proliferation and IFNγ production were restored with both stimuli, whereas PD-1 (a molecule associated with cellular exhaustion) expression, was reduced. Our work suggests that CD8 response is associated with control of Lm infection in LCL patients and that chronic infection in DCL patients leads to a state of CD8 functional exhaustion, which could facilitate disease spread. This is the first report that shows the presence of functionally exhausted CD8 T lymphocytes in DCL patients and, additionally, that pre-stimulation with TLR2 ligands can restore the effector mechanisms of CD8 T lymphocytes from DCL patients against Leishmania mexicana-infected macrophages.  相似文献

Studies on the chemical and biological properties of annual pack ice at a coastal station in Terra Nova Bay (74°41.72′S, 164°11.63′E) were carried out during austral spring at 3-day intervals from 5 November to 1 December 1997. Temporal changes of nutrient concentrations, algal biomasses, taxonomic composition, photosynthetic pigment spectra and P–E relationships were studied. Quantity, composition and degradation rates of organic matter in the intact sea ice were also investigated. In addition, microcosm experiments were carried out to evaluate photosynthetic and photo-acclimation processes of the sympagic flora in relation to different light regimes. High concentrations of ammonia were measured in four ice-cores (weighted mean values of the cores ranged from 4.3 ± 1.9 μM to 7.2 ± 3.4 μM), whereas nitrate and phosphate displayed high concentrations (up to 35.9 μM and 7.6 μM, respectively) only in the bottom layer (135–145 cm depth). Particulate carbohydrate and protein concentrations in the intact sea ice ranged from 0.5 to 2.3 mg l−1 and 0.2 to 2.0 mg l−1, respectively, displaying a notable accumulation of organic matter in the bottom colored layer, where bacterial enzymatic activities also reached the highest values. Aminopeptidase activity was extremely high (up to 19.7 μM l−1 h−1 ± 0.05 in the bottom layer), suggesting a rapid turnover rate of nitrogen–enriched organic compounds (e.g. proteins). By contrast, bacterial secondary production was low, suggesting that only a very small fraction of mobilized organic matter was converted into bacterial biomass (<0.01‰). The sympagic autotrophic biomass (in terms of chlorophaeopigments) of the bottom layer was high, increasing during the sampling period from 680 to 2480 μg l−1. Analyses of pigments performed by HPLC, as well as microscope observations, indicated that diatoms dominated bottom communities. The most important species were Amphiprora sp. and Nitschia cfr. stellata. Bottom sympagic communities showed an average P B max of 0.12 mgC mg Chl−1 and low photoadaptation index (E k=18 μE m−2 s−1, E m=65 μE m−2 s−1). Results of the microcosm experiment also indicated that communities were photo-oxidized when irradiance exceeded 100 μE m−2 s−1. This result suggests that micro- autotrophs inhabiting sea ice might have a minor role in the pelagic algal blooms. Accepted: 4 August 1999  相似文献

The aim of the study was to investigate the capacity of microalgae from the extremely low light habitat of bottom ice to acclimate to different light conditions. During austral spring 1997 the bottom layer of land-fast ice in Terra Nova Bay displayed high values of microalgal biomass up to 2,400 μg Chla L−1 concentrated in a few centimetres ice layer. The algal assemblage was dominated by benthic pennate diatoms. Photoacclimation of the microalgae was addressed in terms of pigment spectra and photosynthetic parameters. Immediate and long term (minutes to days) changes in the photoprotective pigments (DD-cycle) were analysed. Severe photodamage occurred in microalgal assemblages exposed to high light. However, part of the bottom ice algal community showed a notable ability to acclimate to high irradiance levels. Changes in photosynthetic parameters preceded the sudden abrupt changes in pigment synthesis and the rapid increase in biomass and growth rates. Phytoplankton populations were investigated weekly at a central station in the Fusaro lagoon (Mediterranean Sea) from 27 November 1989 to 18 June 1990 to assess species composition, temporal succession and standing stock of the different species. Chlorophyll concentrations varied from 1.2 to 73.2 µg 1–1 in surface waters, and from 1.3 to 53.5 µg 1–1 at the 4.5 m depth. Phytoplankton communities were dominated by Prorocentrum micans Ehrenberg in December and January, and by small-sized diatoms in the rest of the sampling period. In surface waters, a maximum biomass of 9.5 mg C 1–1 was measured in January, in correspondence with high concentrations (8.1 × 106 cells 1–1) of P. micans, whereas an abundance peak of 159.9 × 106 cells 1–1 was registered on the last sampling date due to a massive bloom of a very small diatom, Minutocellus polymorphus (Hargraves & Guillard) Hasle, von Stosch & Syvertsen. On the whole, phytoplankton populations of the Fusaro lagoon showed distinct characters as compared to those of southern Tyrrhenian coastal waters and of other lagoons.  相似文献

The surface distribution of chlorophyll a (chl a) from size-fractionated phytoplankton and of particulate organic matter was studied along the Strait of Magellan during late austral summer (February 20th to March 2nd, 1991), in order to contribute an outline of the ecological characteristics of its pelagic compartment. Sampling of surface water was carried out at 2.5 mile intervals, yielding 152 sampling points for chl a and 104 for particulate organic carbon (POC). The Strait appeared as a system strongly controlled by land forcing. Its phytoplankton community was dominated by the picoplanktonic fraction along its entire length, with mean chl a concentrations of 0.74 and 1.17 g dm–3 for pico- and total phytoplankton, respectively. The microphytoplankton never exceeded 0.02 g dm–3. POC concentrations, with a maximum of 242.5 and a mean of 144.8 g dm–3, were mainly of autotrophic origin, as indicated by a mean POC:chl a ratio of 138.4.  相似文献
