Seven new loci, casein alpha-S1 (CSN1S1), casein alpha-S2 (CSN1S2), casein beta (CSN2), the Hardy-Zuckerman 4 feline sarcoma viral (v-kit) oncogene homolog (KIT), albumin (ALB), phosphodiesterase cyclic GMP (rod receptor) beta polypeptide (PDEB), and complement component 1 (IF), were assigned to sheep Chromosome (Chr) 6 by Southern hybridization to a panel of chromosomally characterized sheep x hamster cell hybrids. By isotopic in situ hybridization, CSN2 was regionally localized to sheep Chr (OOV) 6q22–q31, anchoring this syntenic group of markers on to OOV6 and confirming its homology at a molecular and cytological level with cattle Chr 6. The assignment of these loci, from PDEB (located on human Chr 4p16.3) to IF (on HSA4q24–q25), and the observation that interleukin 2 (IL2, on HSA 4q26–q27) and tryptophan 2,3-dioxygenase (TDO2, on HSA4q31) are not located on OOV6, is further evidence of the close evolutionary relationship of sheep and cattle and the conserved synteny in these species of this extensive region of human Chr 4. On the basis of this conserved synteny, and the similar G- and Q-banding patterns of this chromosome in cattle and sheep, we propose that this sheep chromosome be numbered as 6, not 4 as recommended by ISCNDA (1990).  相似文献

An experimental assessment of the temperature responses of two sympatric seagrasses, Amphibolis antarctica and Amphibolis griffithii, in relation to their biogeography.Seedlings of the viviparous seagrasses, Amphibolis antarctica (Labill.) Sonder & Aschers. and Amphibolis griffithii (Black) den Hartog, were grown in seawater cultures at temperatures of 10–30 °C. This temperature range exceeded the range of temperatures occurring in habitats where Amphibolis grows.All seedlings of both species survived at 15 °C, and all A. antarctica at 10 and 20 °C. There was some mortality at 25 °C, but more in A. griffithii than in A. antarctica. All seedlings showed marked senescence at 30 °C within 2 weeks, and all seedlings of both species were dead at this temperature in 6 weeks. Leaf production rates were different at different temperatures for each species, but were high across the 15–25 °C temperature range for both species. Given the time of release of seedlings from parent plants (winter), these results are consistent with the observed distribution of adult plants, and so the more restricted range of A. griffithii can be explained partially by its' response to temperature in culture. These results do not explain the absence of both Amphibolis species from the east coast of Australia, which may be a consequence of habitat availability.  相似文献

IntroductionIncreasing longevity means more people will be dying in very old age, but little is known about the preferences of the ‘oldest old’ regarding their care at the end of life.AimsTo understand very old people’s preferences regarding care towards the end of life and attitudes towards dying, to inform policy and practice.MethodsQualitative data collection for n = 42 population-based cohort study participants aged 95–101 (88% women, 42% in long-term-care): topic-guided interviews with n = 33 participants and n = 39 proxy informants, most with both (n = 30: 4 jointly + separate interviews for 26 dyads).ResultsDeath was a part of life: these very old people mainly live day-to-day. Most were ready to die, reflecting their concerns regarding quality of life, being a nuisance, having nothing to live for and having lived long enough. Contrasting views were rare exceptions but voiced firmly. Most were not worried about death itself, but concerned more about the dying process and impacts on those left behind; a peaceful and pain-free death was a common ideal. Attitudes ranged from not wanting to think about death, through accepting its inevitable approach to longing for its release. Preferring to be made comfortable rather than have life-saving treatment if seriously ill, and wishing to avoid hospital, were commonly expressed views. There was little or no future planning, some consciously choosing not to. Uncertainty hampered end-of-life planning even when death was expected soon. Some stressed circumstances, such as severe dependency and others’ likely decision-making roles, would influence choices. Carers found these issues harder to raise but felt they would know their older relatives’ preferences, usually palliative care, although we found two discrepant views.ConclusionsThis study’s rare data show ≥95-year-olds are willing to discuss dying and end-of-life care but seldom do. Formal documentation of wishes is extremely rare and may not be welcome. Although being “ready to die” and preferring a palliative approach predominated, these preferences cannot be assumed.  相似文献

South–west Australia contains extensive seagrass meadows along 2,500 km of coastline from the shallow subtidal to 50+ m water depths, and in many of the 51 bar-built estuaries along the coast. There are geomorphological differences between the south and west coasts that result in different patterns of swell exposure influencing the processes that structure seagrass habitats. In this paper, ‘sheltered’, ‘exposed’ and ‘estuarine’ seagrass habitat types are defined for south–west Australia to synthesize processes influencing seagrass communities. Sheltered habitats in south–west Australia are characterized by high light, low to moderate water motion and sporadic disturbance from storms, making them ideal habitats for a diversity of seagrass assemblages. Exposed seagrass habitats are characterized by the presence of strong and consistent ocean swells (3–8 m), predominantly from the south or south–west and seagrasses exhibit a suite of adaptive traits to survive the effects of exposure to ocean swell and associated sand movement. These include morphological features such as heavy fiber reinforcement to strengthen the aboveground stems or leaves, deep vertical rhizomes and life history traits such as rapid growth and high seed set. Within estuarine habitats highly dynamic seagrass communities are the result of fluctuating annual cycles in temperature, light and salinity. Compared to global seagrass meadows, coastal south–west Australian seagrass habitats experience high light, low nutrients and high water movement. Despite these differences, similarities with other regions do exist and here we place the habitats of south–west Australia into a global context using comparative data. The wide array of morphology and life history traits displayed among seagrass species of south–west Australia are presented in a conceptual model including habitat type, physical stressors and seagrass responses. The combination of adaptations to the habitats and processes that define them make south–west Australia a region where there is an unusually high number of co-occurring seagrass species, the highest in the world for a temperate environment (19 species), and approaching the species diversity of many tropical environments. Linking aspects of seagrass habitat, physical aspects of the environment and seagrass life history provides a context for applying knowledge gained from seagrasses in south–west Australia to other coastal ecosystems throughout the world.  相似文献

Background  

The influenza A virus is an important infectious cause of morbidity and mortality in humans and was responsible for 3 pandemics in the 20th century. As the replication of the influenza virus is based on its host's machinery, codon usage of its viral genes might be subject to host selection pressures, especially after interspecies transmission. The turnover of each protein in the mammalian proteome is a functionally important characteristic. Here, we employed high-resolution mass spectrometry to quantify protein dynamics in nondividing mammalian cells. The ratio of externally supplied versus endogenous amino acids to de novo protein synthesis was about 17:1. Using subsaturating SILAC labeling, we obtained accurate turnover rates of 4106 proteins in HeLa and 3528 proteins in C2C12 cells. Comparison of these human and mouse cell lines revealed a highly significant turnover correlation of protein orthologs and thus high species conservation. Functionally, we observed statistically significant trends for the turnover of phosphoproteins and gene ontology categories that showed extensive covariation between mouse and human. Likewise, the members of some protein complexes, such as the proteasome, have highly similar turnover rates. The high species conservation and the low complex variances thus imply great regulatory fine-tuning of protein turnover.  相似文献
