The nucleolus organizer locus of Xenopus laevis lampbrush chromosomes was identified by in situ hybridization of a 3H-labelled probe complementary to 18S + 28S rDNA. The nucleolus organizer is an axial granule on chromosome III that lies four-fifths the way down this chromosome reading from its larger (left) telomere, just within an exploded region that extends to its right end, where the lateral loops are exceptionally long. By in situ hybridization of 3H-labelled oocyte and somatic 5S spacer cRNA probes to similarly RNase-treated and denatured lampbrush chromosomes, the multiple sites of oocyte and somatic 5S gene families were identified. Oocyte 5S genes lie at the larger telomeres of the 15 chromosomes that possess these structures; that is, all but chromosomes X, XVII and XVIII. There are a further four sites, all peripheral, and in three of these, on chromosomes VII, X and XI, the sequences lie on lateral loops that are resolvable with the light microscope. By contrast all of the somatic 5S gene clusters occupy peripheral sites. There are two sites on chromosome III, one of which may be shared with oocyte 5S sequences; one on chromosome VII, which is very likely shared with oocyte 5S sequences; one terminal site on chromosome X; one site on chromosome XI that lies on a single pair of long loops which are inserted in a conspicuous and recognizable axial granule, loops which certainly carry oocyte 5S sequences too; two nearly terminal sites alongside the larger telomeres on chromosomes XII and XIV; and single interstitial sites on all three of the sphere-bearing chromosomes, VIII, IX and XVI. We suggest that 5S sequences on resolvable loops are transcribed by readthrough from upstream promoters, probably by polymerase II.  相似文献

Single batrachotoxin-activated sodium channels from rat brain were modified by trimethyloxonium (TMO) after incorporation in planar lipid bilayers. TMO modification eliminated saxitoxin (STX) sensitivity, reduced the single channel conductance by 37%, and reduced calcium block of inward sodium currents. These effects always occurred concomitantly, in an all-or-none fashion. Calcium and STX protected sodium channels from TMO modification with potencies similar to their affinities for block. Calcium inhibited STX binding to rat brain membrane vesicles and relieved toxin block of channels in bilayers, apparently by competing with STX for the toxin binding site. These results suggest that toxins, permeant cations, and blocking cations can interact with a common site on the sodium channel near the extracellular surface. It is likely that permeant cations transiently bind to this superficial site, as the first of several steps in passing inward through the channel.  相似文献

42 single women who want to be childless, 18 who desire a 1 child family, and 42 who want 2 children responded to questions about the quality of their present lives and nature of future marriages. The groups of Australian women were closely matched for age and education, and on current quality of life there were few differences. They had similar levels of positive and negative affect, and described life as enjoyable, interesting, and contented. Those wanting to be childless, however, had rated life as less optimistic and less loving, and also as currently somewhat less satisfying, but life satisfaction was still quite high. These early deciders of childlessness, and those wanting only 1 child, wanted to be financially and socially more independent in future marriage type relationships than women wanting at least 2 children. They also expected to follow interests and careers to their fullest, wanted more role innovative partners, and were somewhat less concerned about home ownership, living to a budget, and keeping contact with family and friends. All women placed considerable importance on the need for trust, self-disclosure, and open communication in a relationship, although the voluntary childless rated more highly the need for intellectual stimulation and for each partner to be happy about the success of their mate. Compared to women wanting 2 children, those wanting to be childless expected a partner to perform a wide range of non-traditional roles in the home.  相似文献

Background

Mycobacterium ulcerans (MU) is responsible for disfiguring skin lesions and is endemic on the Bellarine peninsula of southeastern Australia. Antibiotics have been shown to be highly effective in sterilizing lesions and preventing disease recurrences when used alone or in combination with surgery. Our practice has evolved to using primarily oral medical therapy.

Methods

From a prospective cohort of MU patients managed at Barwon Health, we describe those treated with primary medical therapy defined as treatment of a M. ulcerans lesion with antimicrobials either alone or in conjunction with limited surgical debridement.

Results

From 1/10/2010 through 31/12/11, 43 patients were treated with exclusive medical therapy, of which 5 (12%) also underwent limited surgical debridement. The median patient age was 50.2 years, and 86% had WHO category 1 and 91% ulcerative lesions. Rifampicin was combined with ciprofloxacin in 30 (70%) and clarithromycin in 12 (28%) patients. The median duration of antibiotic therapy was 56 days, with 7 (16%) receiving less than 56 days. Medication side effects requiring cessation of one or more antibiotics occurred in 7 (16%) patients. Forty-two (98%) patients healed without recurrence within 12 months, and 1 patient (2%) experienced a relapse 4 months after completion of 8 weeks of antimicrobial therapy.

Conclusion

Our experience demonstrates the efficacy and safety of primary oral medical management of MU infection with oral rifampicin-based regimens. Further research is required to determine the optimal and minimum durations of antibiotic therapy, and the most effective antibiotic dosages and formulations for young children.  相似文献

Epstein-Barr virus is a gammaherpes virus that establishes persistent infection in the majority of humans. Despite the potent oncogenic potential of the virus it is relatively rarely associated with malignant disease. We have analyzed the CD8(+) T cell response to EBV and find that a larger primary burst size is associated with proportionally greater decay during the development of memory. Consequently, immunodominance and clonal dominance are less marked in memory than primary responses. An intuitive interpretation of this finding is that there is a limit to the number of cell divisions a T cell clone can undergo, and that the progeny of clones that have expanded massively during a primary immune response are more prone to die as a result of senescence. To test this hypothesis, we have derived a mathematical model of the response of different T cell clones of varying avidity for Ag in the primary and persistent phases of viral infection. When cellular survival and replication are linked to T cell avidity for Ag and Ag dose, then high-avidity T cells dominate both the primary and secondary responses. We then incorporated a limit in the number of cell divisions of individual T cell clones to test whether such a constraint could reproduce the observed association between cell division number and alterations in the contribution of clones to the response to persistent infection. Comparison of the model output with the experimental results obtained from primary and persistent EBV infection suggests that there is indeed a role for cellular senescence in shaping the immune response to persistent infection.  相似文献
