Grassland desertification seriously threatens sustainable economic and social development. Much attention has been paid to the control of grassland desertification, and even to the restoration and reconstruction of the grassland. Vegetation restoration is considered to be a very effective solution. Soil sustains an immense diversity of microbes, and the characteristics of soil microbial communities are sensitive indicators of soil. It is important to understand the relationship between vegetation and soil microbial diversity during the restoration process. Soil microbial, which is the main index to evaluate soil quality, plays a significant role in ecosystem and soil microbial diversity is the important one of global diversity. Exploring the effects of different vegetation patterns on soil microbial diversity can provide scientific bases and technical support for systemic and impersonal assessment of the best vegetation restoration patterns, as well as the vegetation restoration and reconstruction of Hulunbeier sandy land. Based on PCR–DGGE technology, a case study was carried out to investigate the effects of five different vegetation restoration patterns on soil microbial functional diversity after 4 years in sandy land in Hulunbeier, China. The five vegetation restoration patterns included mono-cultivar planting of Agropyron cristatum (UA), mono-cultivar planting of Hedysarum fruticosum (UH), mono-cultivar planting of Caragana korshinskii (UC), mixed-cultivar planting of Agropyron cristatum and Hedysarum fruticosum (AC) and mixed-cultivar planting of Agropyron cristatum, Hedysarum fruticosum, Caragana korshinskii and Elymus nutans (ACHE). Completely degraded sandy land was used as control.The results indicated that the vegetation restoration increased the genetic diversity of soil bacterial community obviously, and the structure of soil bacterial community was changed. The results of phylogenetic analysis suggested that the bacterial community in Hulunbeier sandy land mainly attributed to Proteobacteria, Bacteroidetes, Firmicutes, Actinobacteria, and Acidobacteria. The dominant groups were Proteobacteria and Bacteroidetes. The effects of different vegetation type on soil bacterial community structures were different.  相似文献

本实验比较蔗糖或海藻糖作为非渗透性保护剂对水牛成熟卵母细胞冷冻效果的影响。将体外成熟的水牛卵母细胞随机分成对照组、蔗糖组、海藻糖组,研究玻璃化冷冻后的存活率、细胞骨架和孤雌激活后发育潜能的变化。结果表明:玻璃化冷冻中蔗糖组(89.68%)与海藻糖组(91.81%)的存活率差异不显著;冻融后的卵母细胞在细胞骨架方面的表现为:蔗糖组和海藻糖组的纺锤体结构、染色体形态与微丝分布正常率分别为34.69%、42.83%、39.13%和39.51%、49.43%、42.61%,两组间差异不显著(p0.05),但均明显低于对照组(66.40%,71.82%,76.18%,p0.01);在进一步研究发育潜能中发现,冻融后卵母细胞的卵裂率、囊胚率和囊胚细胞数在实验组中均无显著性差异(p0.05),但两组的卵裂率、囊胚率均显著低于对照组(p0.01)。综上所述,玻璃化冷冻时添加蔗糖或海藻糖作为非渗透性保护剂对水牛成熟卵母细胞的保护作用差异不明显,表明两者均可在冷冻时添加使用。  相似文献

采用改进的碱裂解法提取Gluconacetobacter hansenii ATCC23769自发不产膜突变体的内源隐蔽质粒。用不同的限制性内切酶对混合质粒直接进行酶切,酶切后的片段混合物与pUC18载体连接构建重组载体。重组载体回转入G.hansenii ATCC23769获得隐蔽质粒上具有复制能力的片段,序列结果分析表明:该片段上没有某些其他质粒所具有的Rep蛋白。利用该片段,构建了能同时在大肠杆菌和葡糖酸醋杆菌中复制的质粒载体,体内的抗生素抗性实验证明该载体具有良好的稳定性。  相似文献

Hereditary spastic paraplegia (HSP) is a neurodegenerative disorder characterized by retrograde axonal degeneration that primarily affects long spinal neurons. The gene encoding spastin has a well-established association with HSP, and protrudin is a known binding partner of spastin. Here, we demonstrate that the N-terminal domain of protrudin mediates the interaction with spastin, which is responsible for neurite outgrowth. We show that spastin promotes protrudin-dependent neurite outgrowth in PC12 cells. To further confirm these physiological functions in vivo, we microinjected zebrafish embryos with various protrudin/spastin mRNA and morpholinos. The results suggest that the spinal cord motor neuron axon outgrowth of zebrafish is regulated by the interaction between spastin and protrudin. In addition, the putative HSP-associated protrudinG191V mutation was shown to alter the subcellular distribution and impair the yolk sac extension of zebrafish, but without significant defects in neurite outgrowth both in PC12 cells and zebrafish. Taken together, our findings indicate that protrudin interacts with spastin and induces axon formation through its N-terminal domain. Moreover, protrudin and spastin may work together to play an indispensable role in motor axon outgrowth.  相似文献

采用寄主转接建立生命表的方法研究了棉花型和黄瓜型棉蚜对不同寄主植物的适应性,以及两寄主型棉蚜是否可通过中间桥梁寄主实现寄主互换的问题。结果表明,两寄主型棉蚜直接互换寄主后,其存活和繁殖力显著下降,表现为棉花型和黄瓜型棉蚜的净增殖率比在原寄主上分别下降980倍和12倍,平均世代寿命缩短5~12d。两寄主型棉蚜均能利用木槿植物,并且适应性没有显著差异。但是两寄主型棉蚜均不能在车前草和大叶黄杨上存活和繁殖后代。西葫芦作物对棉蚜在木槿、棉花和黄瓜寄主上的相互转移起到了重要的桥梁寄主作用。冬寄主木槿上棉蚜可通过甜瓜或西葫芦转移到黄瓜寄主上,棉花和黄瓜上棉蚜也可通西葫芦作物分别转移到黄瓜和棉花作物上,从而形成棉蚜在不同寄主植物间的相互转移通道,造成为害和病毒病的扩张。  相似文献

An accurate visual reporter system to assess homology-directed repair (HDR) is a key prerequisite for evaluating the efficiency of Cas9-mediated precise gene editing. Herein, we tested the utility of the widespread promoterless EGFP reporter to assess the efficiency of CRISPR/Cas9-mediated homologous recombination by fluorescence expression. We firstly established a promoterless EGFP reporter donor targeting the porcine GAPDH locus to study CRISPR/Cas9-mediated homologous recombination in porcine cells. Curiously, EGFP was expressed at unexpectedly high levels from the promoterless donor in porcine cells, with or without Cas9/sgRNA. Even higher EGFP expression was detected in human cells and those of other species when the porcine donor was transfected alone. Therefore, EGFP could be expressed at certain level in various cells transfected with the promoterless EGFP reporter alone, making it a low-resolution reporter for measuring Cas9-mediated HDR events. In summary, the widespread promoterless EGFP reporter could not be an ideal measurement for HDR screening and there is an urgent need to develop a more reliable, high-resolution HDR screening system to better explore strategies of increasing the efficiency of Cas9-mediated HDR in mammalian cells.  相似文献
