The influence of phytohormones on chlorophyll and carotenoid formation during the greening of irradiated dark grown wheat leaves (Triticum aestivum L. cv. Starke II Weibull) was studied. Leaves were floated on solutions of abscisic acid, gibberellic acid and kinetin for 24 h. The chlorophyll and carotenoid contents were determined during a subsequent period of 48 h of continuous irradiation. Leaves treated with abscisic acid showed a longer lag phase and a lower rate of accumulation of chlorophyll as compared to the control than did leaves treated with gibberellic acid and kinetin. The carotenoid content was low both in leaves treated with abscisic acid and in those treated with gibberellic acid. Treatment with abscisic acid lowered the protochlorophyllide regeneration after a saturating light flash while gibberellic acid as well as kinetin had no effect. The influence of ABA was partly dependent on an increase of the wounded part of the cut leaf segments. The accumulation of protochlorophyllide in leaves treated with δ-aminolevulinic acid was not affected by the different hormonal treatments. These results suggest that the main effect of abscisic acid is probably outside the chloroplast, i.e. on the formation or transport of δ-aminolevulinic acid.  相似文献

ABSTRACT Traditional methods of monitoring gray wolves (Canis lupus) are expensive and invasive and require extensive efforts to capture individual animals. Noninvasive genetic sampling (NGS) is an alternative method that can provide data to answer management questions and complement already-existing methods. In a 2-year study, we tested this approach for Idaho gray wolves in areas of known high and low wolf density. To focus sampling efforts across a large study area and increase our chances of detecting reproductive packs, we visited 964 areas with landscape characteristics similar to known wolf rendezvous sites. We collected scat or hair samples from 20% of sites and identified 122 wolves, using 8–9 microsatellite loci. We used the minimum count of wolves to accurately detect known differences in wolf density. Maximum likelihood and Bayesian single-session population estimators performed similarly and accurately estimated the population size, compared with a radiotelemetry population estimate, in both years, and an average of 1.7 captures per individual were necessary for achieving accurate population estimates. Subsampling scenarios revealed that both scat and hair samples were important for achieving accurate population estimates, but visiting 75% and 50% of the sites still gave reasonable estimates and reduced costs. Our research provides managers with an efficient and accurate method for monitoring high-density and low-density wolf populations in remote areas.  相似文献

We developed and characterized primers for 11 variable microsatellite loci present in the genome of the flax rust, Melampsora lini. The microsatellite loci were identified by sequencing clones from a library of EcoRI DNA fragments enriched for four simple sequence repeat motifs (AAG, AAT, TC and TG). All 11 primer pairs successfully amplified DNA fragments from a sample of 102 M. lini isolates (98 isolated from Linum marginale and four from Linum usitatissimum), revealing a total of 32 alleles. Allelic diversity at the 11 loci ranged from 0.030 to 0.449.  相似文献

Young wheat plants (Triticum aestivum L. cv. Weibulls Starke II) were exposed to water stress for 1, 2 or 3 hours by cooling the roots. The plants were subjected to a constant water stress during the stress periods. By this treatment the leaf water potential was lowered from ?6.5 to ?11.5 bars. Leaf water potential, transpiration rate and abscisic acid content were determined during the stress periods and during the recovery. The water stressed plants showed an after-effect on transpiration rate lasting for between 10 and 24 hours depending on the duration of the stress. The amount of water stress in the stressed plants compared with the controls is defined as the difference in leaf water potential between the controls and the stressed plants during the stress period integrated over time. The amount of after-affect on transpiration is analogously defined as the difference in transpiration rate between the controls and the stressed plants during the recovery period integrated over time. There was a linear relationship between the amount of water stress and the amount of after-effect on transpiration of the leaves. The abscisic acid content of the leaves increased between 3.0 and 4.5 times the original content depending on the duration of the stress. However, during the recovery the abscisic acid content reattained the pre-stress level within 3 hours for all three stress periods. There was thus no direct relationship between the after-effect and the abscisic acid content of the leaf.  相似文献

SYNOPSIS. Besnoitia darlingi (Brumpt, 1913) Mandour, 1965 has been rediscovered in the common opossum Didelphis marsupialis in the Republic of Panama. A strain (D3) has been established in laboratory white mice. Proliferative crescents of the D3 strain are 6.1 (range 6.0-9.0) by 2.1 (range 1.7-4.0).
Mouse-adapted B. darlingi produced acute, fatal infections in white mice, hamsters, 14 marmosets ( Saguinus geoffroyi ), 2 squirrels ( Sciurus variegatoides and S. granatensis ), a woolly opossum ( Caluromys derbianus ) and a four-eyed opossum ( Philander opossum ) It probably produced chronic infections (cysts) in 2 wild-caughtopossums ( Didelphis marsupialis ) and one lizard ( Ameiva ameiva ). Animals in which induced infection could not be found after inoculation with trophozoites of mouse-adapted B. darlingi were: 2 adult and 2 new-born guinea pigs, laboratory rats, 1 night monkey ( Aotus trivirgatus ), 2 rhesus monkeys ( Macacca mulatta ), one iguana ( Iguaanaiguana ) and 2 baby caimans ( Caiman sclerops ).  相似文献

Cut seedlings of wheat plants (Triticum aestivum L. cv. Starke II Weibull) between 6 and 7 days old were water stressed in darkness by exposing them to air of 35% relative humidity 2.5 to 20 h. This treatment resulted in a water potential of -11 bars in the leaves after 20 h. The leaves were then rewatered and irradiated. The chlorophyll formation that took place in fully turgid leaves during the greening was markedly decreased in the case of the water-stress pretreatmet. and especially the lag phase was prolonged. The longer the stress pretreatment the more evident was the subsequent effect on chlorophyll formation. However, no linear relationship was found between the amount of stress and the chlorophyll content. Protochlorophyllide regeneration from endogenously formed δ-aminolevulinic acid was markedly decreased even after the shortest water-stress period. However, protochlorophyllide accumulation from exogenously supplied δ-aminolevulinic acid was only slightly decreased following the water-stress pretreatment. Further more, the ratio of protochlorophyllide650 to protochlorophyllide628 was slightly reduced by the same conditions. During the stress period both abscisic acid and proline were accumulated in the leaves. The content of abscisic acid increased up to six times the normal level during water stress lasting for 20 h. The increase of proline was about three-fold for similar treatment. After rewatering the leaves the levels of both abscisic acid and proline rapidly declined and reached. 10 h later, the levels found in unstressed seedlings. The increase in abscisic acid during water stress associated with impaired chlorophyll metabolism suggested that the after-effect of water stress might be linked to chlorophyll metabolism through abscisic acid or some of its metabolites. The changes in proline content open the possibility that this substance could function as a reserve substance for the formation of chlorophyll after the discon tinuation of the stress.  相似文献

Effects of kinetin on transpiration rate and abscisic acid content were determined. Leaves from 9-day-old wheat plants (Triticum aestivum L. cv. Weibull's Starke II) were used. —Transpiration rate decreased in excised leaves put in water, but it was maintained at a higher rate when kinetin was supplied. When excised leaves were water stressed by air-drying for 1 h, addition of kinetin resulted in a considerable stimulation of transpiration rate. The effect reached its maximum after 15 h and this level remained relatively unchanged for at least 10 h. Intact seedlings which were stressed before leaf excision, showed only a slight stimulation of kinetin on transpiration rate. — Abscisic acid content slowly increased up to three-fold in 2 days in excised leaves put in water. In excised and water-stressed leaves the abscisic acid content was reduced during the first 24 h and then increased. As the leaves were fully turgid, the increase could not have been caused by water stress. However, both in stressed and unstressed leaves kinetin addition reduced the increase in abscisic acid content. — It is suggested that the stimulation by kinetin on transpiration rate in excised and water stressed leaves was mainly due to the combined effect of (1) a reduction in the activity of endogenous cytokinins, (2) kinetin acting as a ‘substitute’ for the inactivated cytokinins but exerting a stronger effect on transpiration than the endogenous cytokinins, and (3) the ‘extra’ reduction in abscisic acid content caused by the kinetin treatment. Furthermore, the results indicate that changes in cytokinins might be partly responsible for the aftereffect on transpiration.  相似文献

Parentage studies have shown that alternative reproductive strategies are widespread in many avian taxa that were once thought to be monogamous. Recent anthropogenically mediated habitat change may have disrupted ecological factors, such as breeding density, which have given rise to inter‐ and intraspecific variation in the frequency of extra‐pair fertilization (EPF) and intraspecific brood parasitism (IBP). We used genetic analyses to quantify the incidence of alternative reproductive strategies exhibited within clutches of Canada Geese Branta canadensis maxima nesting in high‐ and low‐density situations in and around urban areas in southern Michigan, USA. We tested the hypothesis that high nesting density would increase the frequency of EPF and IBP. There were no significant differences in rates of EPF and IBP clutches (14 and 26% of clutches, respectively) from nests in high‐density (21.7% EPF, 21.7% IBP) vs. low‐density (5.3% EPF, 31.6% IBP) areas, although high‐density sites had a fourfold higher rate of EPF. Rates of EPF and IBP in high‐density urban areas in Michigan were comparable to rates observed in other species nesting under different ecological conditions. Levels of relatedness between host and parasitic females were higher than expected by chance, suggesting that related females are more tolerant of one another and that host females could gain inclusive fitness benefits from rearing parasitic offspring. Our study highlights the importance of understanding the different costs and benefits associated with alternative behavioural repertoires that may vary as habitats and associated selection pressures are increasingly modified by human activities.  相似文献
