Non-selected and Na2SO-, K2SO4- or KCl-selected callus culturesof Vaccinium corymbosum L. cv. Blue Crop were grown on mediasupplemented with 0, 25 and 50 mM Na2SO4 (non-selected and Na2SO(-selectedonly), 0, 25 and 50mMK2SO4 (non-selected and K2SO4-selectedonly) or 0, 50 and 100 mM KCl (non-selected and KCl-selectedonly). On all media, growth of selected callus (on a fresh-weightor dry-weight basis) was greater than that of non-selected callus,and selected callus grew optimally on the level and type ofsalt on which it was selected. Selected callus was friable andmaintained a higher f. wt:d. wt ratio. Tissue water potentialin selected callus was more negative than in non-selected callus. Flame photometry and chloridometry showed Na+, K+ and Claccumulated in callus to concentrations equal to or greaterthan the initial concentration in the medium. Turbidometry showedthat tissue SO42- concentration was lower than the concentrationin the medium. In most cases selected callus accumulated moreNa+, Ksup, SO42– or Cl than non-selected callus.Vacuolar ion concentration was measured by electronprobe X-raymicroanalysis, and on most media selected callus had highervacuolar ion concentrations than non-selected callus. SO42–and Cl were accumulated in the vacuoles at concentrationshigher than the external medium, but vacuolar Na+ concentrationdid not reach external concentration on Na2SO4 and on potassiumsalts was maintained between 12 and 17 mM. Vacuolar K+ concentration(approx. 142–191 mM on no salt) decreased on Na2SO4 andincreased on K2SO4 and KCl. There was no precise correlation between total or specific ionaccumulation (Na+, K+, SO42– and Cl and fresh-weightyield. Results suggest that selection results in adaptationin response to decreased water potential of the medium. ABSTRACT. Organisms in the genus Naegleria offer special opportunities for research in contemporary biology. the dramatic cell differentiation from amebae to flagellates is unique among eukaryotes in the rapidity, synchrony, reproducibility, homogeneity, and accessibility of a major phenotypic change. Environmental signals initiate a progressive signal transduction pathway in which genes are turned on, including those for several calcium-binding proteins, and newly synthesized proteins become localized in newly assembled organelles, including the centriole-like basal bodies, with the overall consequence that the cell changes its shape, motility, and behavior. This essay reviews research opportunities for which Naegleria excels, as well as interesting aspects of its biology that provide challenges for future investigations. Because these organisms alternate between two major eukaryotic motility forms, their phylogenetic position is also provocative. Although there are hints that Naegleria is capable of sexual reproduction in nature, mating has not yet been observed in the laboratory. In order to fully exploit the opportunities offered by this wonderful experimental system we are working to develop means to do genetic manipulation, in particular via DNA-mediated transformation.  相似文献

L1 retroposons are represented in mice by subfamilies of interspersed sequences of varied abundance. Previous analyses have indicated that subfamilies are generated by duplicative transposition of a small number of members of the L1 family, the progeny of which then become a major component of the murine L1 population, and are not due to any active processes generating homology within preexisting groups of elements in a particular species. In mice, more than a third of the L1 elements belong to a clade that became active approximately 5 Mya and whose elements are > or = 95% identical. We have collected sequence information from 13 L1 elements isolated from two species of voles (Rodentia: Microtinae: Microtus and Arvicola) and have found that divergence within the vole L1 population is quite different from that in mice, in that there is no abundant subfamily of homologous elements. Individual L1 elements from voles are very divergent from one another and belong to a clade that began a period of elevated duplicative transposition approximately 13 Mya. Sequence analyses of portions of these divergent L1 elements (approximately 250 bp each) gave no evidence for concerted evolution having acted on the vole L1 elements since the split of the two vole lineages approximately 3.5 Mya; that is, the observed interspecific divergence (6.7%-24.7%) is not larger than the intraspecific divergence (7.9%-27.2%), and phylogenetic analyses showed no clustering into Arvicola and Microtus clades.   相似文献

Some Observations on Infection of Arachis hypogaea L. by Rhizobium   总被引:2,自引:0,他引:2  
The infection process in Arachis hypogaea by rhizobia differsfrom that normally found in Trifolium spp. in that no infectionthreads are formed. The root hairs, which are long (up to 4mm), septate, and often with large basal cells, occur only atthe sites of emerging lateral roots. Infection occurs only wherethe root hairs have large basal cells. Rhizobia cause curlingand deformation of the root hairs (as in Trifolium spp.) butenter the root at the junction of the root hair and the epidermaland cortical cells. The bacteria are distributed intercellularlyvia the middle lamellae and enter the cortical cells throughthe structurally altered cell wall, often close to the hostcell nucleus. The root hairs and large basal cells become infectedin the same way. Within the cortical cells of the emerging lateralroot the rhizobia multiply rapidly and the invaded cells dividerepeatedly to form the nodule tissue. Bacteriod formation occursonly when the host cell ceases to divide.  相似文献

Maintenance of a high degree of biodiversity in homogeneous environments is poorly understood. A complex cheese starter culture with a long history of use was characterized as a model system to study simple microbial communities. Eight distinct genetic lineages were identified, encompassing two species: Lactococcus lactis and Leuconostoc mesenteroides. The genetic lineages were found to be collections of strains with variable plasmid content and phage sensitivities. Kill-the-winner hypothesis explaining the suppression of the fittest strains by density-dependent phage predation was operational at the strain level. This prevents the eradication of entire genetic lineages from the community during propagation regimes (back-slopping), stabilizing the genetic heterogeneity in the starter culture against environmental uncertainty.  相似文献

Irisin was first identified in muscle cells. We detected irisin immunoreactivity in various organs of the crested porcupine (Hystrix cristata). In the epidermis, irisin immunoreactivity was localized mainly in stratum basale, stratum spinosum and stratum granulosum layers; immunoreactivity was not observed in the stratum corneum. In the dermis, irisin was found in the external and internal root sheath, cortex and medulla of hair follicles, and in sebaceous glands. Irisin immunoreactivity was found in the neural retina and skeletal muscle fibers associated with the eye. The pineal and thyroid glands also exhibited irisin immunoreactivity.  相似文献
