Sequence variation in the internal transcribed spacer (ITS-1) ribosomal DNA subunit was examined for sorghum midge obtained from introduced and native hosts in south-eastern and central Queensland. No variation was observed relative to host plant or geographical distance for midges collected from two introduced hosts, grain sorghum ( Sorghum bicolor ) and Johnson grass ( S. halepense ); however, sequence differences were observed between midges from introduced and native hosts and among midges from a single native host, slender bluegrass ( Dichanthium affine ). No evidence was observed of introduced midges on native hosts, or vice versa. These results agree with previously hypothesised host distributions for native and introduced midges in Australia, and expand the sample of introduced hosts to include Johnson grass. They suggest that Stenodiplosis sorghicola , the principal midge infesting grain sorghum, is also the most common species on Johnson grass. This confirms that Johnson grass plays a role in the population dynamics of S. sorghicola and suggests that midges originating from Johnson grass may influence levels of infestation in grain sorghum. 相似文献

Nanoparticles have unique properties that make them attractive for use in industrial and medical technology industries but can also be harmful to living organisms, making an understanding of their molecular mechanisms of action essential. We examined the effect of three different sized poly(isobutyl‐cyanoacrylate) nanoparticles (iBCA‐NPs) on the unicellular green alga Chlamydomonas reinhardtii. We found that exposure to iBCA‐NPs immediately caused C. reinhardtii to display abnormal swimming behaviors. Furthermore, after one hour, most of the cells had stopped swimming and 10%–30% of cells were stained with trypan blue, suggesting that these cells had severely impaired plasma membranes. Observation of the cyto‐ultrastructure showed that the cell walls had been severely damaged and that many iBCA‐NPs were located in the space between the cell wall and plasma membrane, as well as inside the cytosol in some cases. A comparison of three strains of C. reinhardtii with different cell wall conditions further showed that the cell mortality ratio increased more rapidly in the absence of a cell wall. Interestingly, cell mortality over time was essentially identical regardless of iBCA‐NP size if the total surface area was the same. Furthermore, direct observation of the trails of iBCA‐NPs indicated that the first trigger was their contact with the cell wall, which is most likely accompanied by the inactivation or removal of adsorbed proteins from the cell wall surface. Cell mortality was accompanied by the overproduction of reactive oxygen species, which was detected more readily in cells grown under constant light rather than in the dark. 相似文献

Shallot (Allium cepa L. Aggregatum group) is an important Allium plant commonly grown in tropical countries, including Indonesia, and it is used as spice for daily cooking, medicine, cosmetics and dietary supplements. Since shallot is predominantly propagated vegetatively by bulbs, it augments pathogen accumulation which influences low seed bulb management as well as productivity. Hence, utilisation of seeds/True Seed Shallot (TSS) emerges an alternative propagation material. However, no information is available on the growth stages of shallot plants from TSS. The phenological growth stages of tropical shallot planted in lowland areas using seeds in accordance with the Biologische Bundesanstalt, Bundessortenamt nd CHemische Industrie (BBCH) scale are first described here. Five introduced TSS cultivars, that is, Tuk Tuk, Lokananta, Sanren, Maserati, Tropix and four landraces TSS cultivars, that is, Bima, Biru Lancor, Pancasona and Trisula, were used as planting materials to develop an extended numerical BBCH scale for the identification of different phenological growth stages of shallot from seed. The study was conducted in lowland areas under tropical conditions of Indonesia. Based on BBCH scale, phenology of tropical shallot from seed in lowland area exhibited five of the 10 principal stages (0–9): germination (stage 0), leaf development (stage 1), formation of lateral shoot/tiller of pseudostem (stage 2), development of bulb (harvestable vegetative part) (stage 4), and senescence (stage 9). The codification of the different growth stages is essential in determination of the correct timing for better crop management, particularly for transplanting, fertilisation, control of weeds, disease and pest, plant growth regulators application and harvest time. As an implication, these BBCH scales could be used in plant breeding activities to shorten the TSS life cycle. 相似文献
