Summary We report a patient (S.T.) with multiple congenital anomalies and developmental delay associated with an interstitial deletion of 1q23–1q25. Molecular analysis of the deletion was performed using DNA markers that map to 1q. Five DNA markers, MLAJ-1 (D1S61), CRI-L1054 (D1S42), HBI40 (D1S66), OS-6 (D1S75), and BH516 (D1S110), were demonstrated to be deleted. Informative polymorphisms demonstrated this to be a de novo deletion of the maternally derived chromosome. Deletion status was determined using restriction fragment length polymorphism (RFLP) analysis supplemented with densitometry in the experiments where RFLP analysis was not fully informative. Deletions were confirmed by Southern analysis using genomic DNA from a somatic cell hybrid retaining the del(1)(q23–q25) chromosome that was constructed from patient S.T. Flow karyotyping confirmed the deletion and estimated that the deletion encompassed 11,000–16,000 kb. The clinical and cytogenetic characteristics of S.T. are compared with those of ten previously described patients with monosomy 1q21–1q25.  相似文献

Membranes purified from castor bean endosperm glyoxysomes by washing with sodium carbonate exhibited integral NADH:ferricyanide and NADH:cytochrome c reductase activities. The enzyme activities could not be attributed to contamination by other endomembranes. Purified endoplasmic reticulum membranes also contained the redox activities; and marker enzyme analysis indicated minimum cross contamination between glyoxysomal and endoplasmic reticulum fractions. The glyoxysomal redox activities were optimally solubilized at detergent to protein ratios (weight to weight) of 10 (Triton X-100), 50 (3-[3-cholamidopropyl)-dimethylammonio]-1-propanesulfonate), and 100 (octylglucoside). Detergent in excess of the solubilization optimum was stimulatory to NADH:ferricyanide reductase and inhibitory to NADH:cytochrome c reductase. Endoplasmic reticulum redox activity solubilization profiles were similar to those obtained for glyoxysomal enzymes using Triton X-100. Purification of the glyoxysomal and endoplasmic reticulum NADH:ferricyanide reductases was accomplished using dye-ligand affinity chromatography on Cibacron blue 3GA agarose. Sodium dodecyl sulfate-polyacrylamide gel electrophoretic analysis of NADH:ferricyanide reductase preparations purified by rate-zonal density gradient centrifugation, affinity chromatography, and nondenaturing electrophoresis of detergent-solubilized glyoxysomal and endoplasmic reticulum membranes consistently displayed 32- and 33-kDa silver-stained polypeptide bands, respectively.  相似文献

Inadequate oxygenation of cardiac muscle leads to rapid loss of high energy compounds essential for contractile function. ATP can be regenerated by synthesis de novo, a route operating at a relatively slow rate in the heart. Myocytes isolated from mature rat heart have been used to measure the rate of ATP synthesis de novo from both [14C]glycine and [14C]ribose. Incorporation of glycine into ATP is accelerated 10-fold in the presence of 1 mM ribose. Myocytes also accumulate both precursors into IMP and four other metabolites on the de novo synthesis pathway. These metabolites represent 80% of the glycine entering the pathway. The potential of de novo synthesis for restoration of adenine nucleotides appears to be limited by the rates of early reactions, adenylosuccinate synthetase being only one of the enzymes operating at a sufficiently slow rate to make this pathway an inherently weak route for the restoration of normal energy status in post-ischemic myocardium. Interventions are being sought to alleviate these apparent metabolic delays.  相似文献

Summary The red, pelagic limestones of the Rosso Ammonitico Veronese (Upper Bajocian-Tithonian) of the Altopiano di Asiago area can be subdivided into eight facies. They differ from each other in structure (bedding style, presence and type of nodularity) and texture (nature of components, grain-vs mud-support, compactional features). Several discontinuities could be recognized, based on sedimentological or biostratigraphic evidence. In the context of a drowned platform, where sediments essentially consist of skeletal remains of both planktonic and benthic organism, the different facies are interpreted as reflecting a varying influence of currents on the winnowing of micrite and on triggering early cementation. Early cementation in turn, controlled the patterns of bioturbation and the response of sediments to later compaction and pressure-dissolution. At times, microbial mats, of unidentified nature, were important in trapping and binding sediment, giving rise to early lithified nodules and layers of stromatolitic aspect. The Rosso Ammonitico Veronese can be subdivided into three units: lower Rosso Ammonitico (RAI: Upper Bajocian-Lower Callovian), middle Rosso Ammonitico (RAM: Upper Callovian-Middle Oxfordian), and upper Rosso Ammonitico (RAS: Lower Kimmeridgian-Tithonian). Frequent ammonite moulds allow the precise dating of the base and top of each unit, and the documentation of facies heteropies and hiatusses in the more fossiliferous RAS. The lower unit (RAM) is massive and essentially nodular; the middle unit (RAM) is well bedded, non-nodular, and cherty; the upper unit (RAS) is richer in clay and typically nodular. The RAI and the RAS are present everywhere, though significant facies and thickness changes affect particularly the RAS; the RAM is much more variable, ranging from 0 to 10 metres. These variations, that may be gradual or abrupt, are inter-preted as the result of Middle-Late Callovian block-faulting which generated an irregular sea floor topography where the swells were more exposed to currents that continuously removed sediments, inducing long-lasting periods of nondeposition. Sediments preferentially accumulated in the adjacent lows. A confirmation of this hypothesis is provided by evidence of synsedimentary tectonics, described for the first time in the Rosso Ammonitico Veronese. Neptunian dykes, both vertical and horizontal, are developed at the top of the RAI and are filled with laminated sediments or collapse breccias. Glides of metre-size blocks and slumps are present at the top of the RAI and at the base of the RAM, respectively. Cm-thick layers of mud supported breccias are intercalated in the upper part of the RAI and within the RAM: they are interpreted as seismites. All these features document a tensional regime that generated fractures in more or less lithified sediments and failure along steep fault scarps or gently dipping slopes of tilted fault blocks. Recognition of this Callovian-Oxfordian tectonic activity shows that, after the Bajocian drowning, the Trento Plateau did not simply experience a uniform and general foundering: a small-scale block-faulting was still active and affected the pattern of facies distribution.  相似文献

Immunological studies of Fe-containing superoxide dismutase (FeSOD) and glutamine synthetase (GS) have established a close relationship betweenOceanospirillum linum (the type species of the genus),O. beijerinckii, Alteromonas communis, A. vaga, and two unnamed species of marine bacteria (groups H-1 and I-1). The four latter species have, consequently, been assigned to the genusOceanospirillum asO. commune comb. nov.,O. vagum comb. nov.,O. kriegii sp. nov. (group H-1; type strain 197, ATCC 27133), andO. jannaschii sp. nov. (group I-1; type strain 207, ATCC 27135). The active site of tyrosinase is described with a view to depicting its interactions with substrates and inhibitors. Occurrence and mechanism(s) of tyrosinase-mediated browning of agrofood products are reviewed, with regard to both enzymic and chemical reactions, and their control, modulation, and inhibition. The contribution that oxidative damage to DNA and/or RNA makes to the aging process remains undefined. In this study, we used the hMTH1‐Tg mouse model to investigate how oxidative damage to nucleic acids affects aging. hMTH1‐Tg mice express high levels of the hMTH1 hydrolase that degrades 8‐oxodGTP and 8‐oxoGTP and excludes 8‐oxoguanine from both DNA and RNA. Compared to wild‐type animals, hMTH1‐overexpressing mice have significantly lower steady‐state levels of 8‐oxoguanine in both nuclear and mitochondrial DNA of several organs, including the brain. hMTH1 overexpression prevents the age‐dependent accumulation of DNA 8‐oxoguanine that occurs in wild‐type mice. These lower levels of oxidized guanines are associated with increased longevity and hMTH1‐Tg animals live significantly longer than their wild‐type littermates. Neither lipid oxidation nor overall antioxidant status is significantly affected by hMTH1 overexpression. At the cellular level, neurospheres derived from adult hMTH1‐Tg neural progenitor cells display increased proliferative capacity and primary fibroblasts from hMTH1‐Tg embryos do not undergo overt senescence in vitro. The significantly lower levels of oxidized DNA/RNA in transgenic animals are associated with behavioral changes. These mice show reduced anxiety and enhanced investigation of environmental and social cues. Longevity conferred by overexpression of a single nucleotide hydrolase in hMTH1‐Tg animals is an example of lifespan extension associated with healthy aging. It provides a link between aging and oxidative damage to nucleic acids.  相似文献

NMDA receptor‐mediated excitotoxicity is thought to play a pivotal role in the pathogenesis of Huntington's disease (HD). The neurotrophin brain‐derived neurotrophic factor (BDNF), which is also highly involved in HD and whose effects are modulated by adenosine A2ARs, influences the activity and expression of striatal NMDA receptors. In electrophysiology experiments, we investigated the role of BDNF toward NMDA‐induced effects in HD models, and the possible involvement of A2ARs. In corticostriatal slices from wild‐type mice and age‐matched symptomatic R6/2 mice (a model of HD), NMDA application (75 μM) induced a transient or a permanent (i.e., toxic) reduction of field potential amplitude, respectively. BDNF (10 ng/mL) potentiated NMDA effects in wild‐type, while it protected from NMDA toxicity in R6/2 mice. Both effects of BDNF were prevented by A2AR blockade. The protective effect of BDNF against NMDA‐induced toxicity was reproduced in a cellular model of HD. These findings may have very important implications for the neuroprotective potential of BDNF and A2AR ligands in HD.  相似文献

Carvedilol has beneficial effects on cardiac function in patients with heart failure but its effect on ovariectomy-induced myocardial contractile dysfunction remains unclear. Estrogen deficiency induces myocardial contractile dysfunction and increases cardiovascular disease risk in postmenopausal women. Our aim was to investigate whether carvedilol, a beta receptor blocker, would prevent ovariectomy-induced myocardial contractile dysfunction. Female rats (8 weeks old) that underwent bilateral ovariectomy were randomly assigned to receive daily treatment with carvedilol (OVX+CAR, 20 mg/kg), placebo (OVX) and SHAM for 58 days. Left ventricle papillary muscle was mounted for isometric tension recordings. The inotropic response to Ca2+ (0.62 to 3.75 mM) and isoproterenol (Iso 10−8 to 10−2 M) were assessed. Expression of calcium handling proteins was measured by western blot analysis. Carvedilol treatment in the OVX animals: prevented weight gain and slight hypertrophy, restored the reduced positive inotropic responses to Ca2+ and isoproterenol, prevented the reduction in SERCA2a expression, abolished the increase in superoxide anion production, normalized the increase in p22phox expression, and decreased serum angiotensin converting enzyme (ACE) activity. This study demonstrated that myocardial contractile dysfunction and SERCA2a down regulation were prevented by carvedilol treatment. Superoxide anion production and NADPH oxidase seem to be involved in this response.  相似文献
