Bacterial infections remain the leading killer worldwide which is worsened by the continuous emergence of antibiotic resistance. In particular, methicillin-sensitive (MSSA) and methicillin-resistant Staphylococcus aureus (MRSA) are prevalent and the latter can be difficult to treat. The traditional strategy of novel therapeutic drug development inevitably leads to emergence of resistant strains, rendering the new drugs ineffective. Therefore, rejuvenating the therapeutic potentials of existing antibiotics offers an attractive novel strategy. Plectasin, a defensin antimicrobial peptide, potentiates the activities of other antibiotics such as β-lactams, aminoglycosides and glycopeptides against MSSA and MRSA. We performed in vitro and in vivo investigations to test against genetically diverse clinical isolates of MSSA (n = 101) and MRSA (n = 115). Minimum inhibitory concentrations (MIC) were determined by the broth microdilution method. The effects of combining plectasin with β-lactams, aminoglycosides and glycopeptides were examined using the chequerboard method and time kill curves. A murine neutropenic thigh model and a murine peritoneal infection model were used to test the effect of combination in vivo. Determined by factional inhibitory concentration index (FICI), plectasin in combination with aminoglycosides (gentamicin, neomycin or amikacin) displayed synergistic effects in 76-78% of MSSA and MRSA. A similar synergistic response was observed when plectasin was combined with β-lactams (penicillin, amoxicillin or flucloxacillin) in 87–89% of MSSA and MRSA. Interestingly, no such interaction was observed when plectasin was paired with vancomycin. Time kill analysis also demonstrated significant synergistic activities when plectasin was combined with amoxicillin, gentamicin or neomycin. In the murine models, plectasin at doses as low as 8 mg/kg augmented the activities of amoxicillin and gentamicin in successful treatment of MSSA and MRSA infections. We demonstrated that plectasin strongly rejuvenates the therapeutic potencies of existing antibiotics in vitro and in vivo. This is a novel strategy that can have major clinical implications in our fight against bacterial infections.  相似文献

The cysteine peptidase cathepsin K is a major player in extracellular proteolysis. Here we describe the identification of the multifunctional extracellular chaperone clusterin as a cathepsin K-binding protein. Clusterin increases the stability of cathepsin K in dilute solution and in the presence of high protein concentration. It does not alter the activity of the enzyme but acts as a liberator by preventing substrate inhibition. Kinetic measurements show that clusterin binds cathepsin K with high affinity (K(d) = 0.5-0.6 nM). Altogether these results provide novel insights into the mechanisms involved in the fine-tuning of cysteine cathepsin activity in the extracellular space.  相似文献

Chlamydia psittaci is a zoonotic pathogen associated primarily with avian chlamydiosis. New chlamydial agents with suspected zoonotic potential were recently detected from domestic poultry in Germany and France indicating that the spectrum of Chlamydiaceae encountered in birds is not confined to a single chlamydial species. For further characterization, a specific real-time PCR targeting the conserved 16S rRNA gene was developed and validated for a specific detection of these atypical Chlamydiaceae. In order to address the epidemiological importance of the new chlamydial agents and their distribution, Chlamydiaceae-positive chicken samples collected from flocks from five different countries were examined. The results confirmed that C.psittaci is not the predominant chlamydial species among chickens examined and suggested that the new chlamydial agents could putatively be widespread in poultry flocks (France, Greece, Croatia, Slovenia and China at least) justifying their systematic investigation when poultry samples are submitted to laboratories for avian chlamydiosis diagnosis. Besides, 16S rRNA-based dendrogram, including sequences from both isolates of the new chlamydial agents or positive samples as well as representative sequences from species belonging to the order Chlamydiales, showed the new chlamydial agents to form a distinct line of descent separated from those of other chlamydial species, but clearly grouped within the family Chlamydiaceae. Finally, the phylogenetic tree inferred from the multi-locus sequence typing based on four housekeeping fragments (gatA, gidA, enoA and hflX) and the ompA-based dendrogram showed an almost identical topology of the new chlamydial agents with that recovered by 16S rRNA-based dendrogram. Deficiency in insulin secretion and function that characterize type 2 diabetes often requires administration of extraneous insulin, leading to injection‐site amyloidosis. Insulin aggregation at neutral pH is not well understood. Although oligomer formation is believed to play an important role, insulin oligomers have not been fully characterized yet. Here, we elucidate similarities and differences between in vitro insulin aggregation at acidic and neutral pH for a range of insulin concentrations (2.5–100 μM) by using kinetic thioflavin T fluorescence, circular dichroism, atomic force and electron microscopy imaging. Importantly, we characterize the size distribution of insulin oligomers at different assembly stages by the application of covalent cross‐linking and gel electrophoresis. Our results show that at the earliest assembly stage, oligomers comprise up to 40% and 70% of soluble insulin at acidic and neutral pH, respectively. While the highest oligomer order increases with insulin concentration at acidic pH, the opposite tendency is observed at neutral pH, where oligomers up to heptamers are formed in 10 μM insulin. These findings suggest that oligomers may be on‐ and off‐pathway assemblies for insulin at acidic and neutral pH, respectively. Agitation, which is required to induce insulin aggregation at neutral pH, is shown to increase fibril formation rate and fibrillar mass both by an order of magnitude. Insulin incubated under agitated conditions at neutral pH rapidly aggregates into large micrometer‐sized aggregates, which may be of physiological relevance and provides insight into injection‐site amyloidosis and toxic pulmonary aggregates induced by administration of extraneous insulin.  相似文献

The aim of the study was to investigate campylobacteriosis incidence in the County Primorsko-goranska (Croatia) between 2003 and 2007 and to find out possible connection with environmental factors (the average monthly temperature and total monthly precipitation). The data (number of stool samples examined, age and sex distribution of patients, monthly distribution of isolates and distribution of isolates according to the species) from the Laboratory for Diagnostics of Enteric Infections of the Teaching Institute of Public Health of the County Primorsko-goranska (Croatia) were analyzed retrospectively. During the observed period 30,164 stool samples were examined for Campylobacter spp. Campylobacters were identified in 1,242 (4.12%) samples. The overall annual campylobacter incidence rate was 81.3 +/- 21.9/100,000 population. Campylobacter jejuni was found in 1,093 (88%) and C. coli in 149 (12%) patients. Our findings showed age distribution of patients typical for developed countries. The patients were mostly children under 5 years (484.4 +/- 129.1/ 100,000, p < 0.001) and between 5 and 9 years of age (226.5 +/- 60.5/100,000, p < 0.05). Male consistently experienced higher rates, but the difference between genders was significant in the age groups from birth till late twentieth (p < 0.001). Campylobacter rates were significantly associated with monthly average temperatures (p < 0.05), but not with precipitation. Further investigations into the incidence of campylobacteriosis on the national level are necessary. The causes of the noticed monthly distribution, sources of infection and connection with the routes of transmission in humans need to be elucidated as well.  相似文献

A survey of large branchiopods in the Banat provincein Yugoslavia revealed a particularly diverse fauna in the vicinity of the village Melenci, 100 km north of Belgrade. The temporary ponds here include 10 species of branchiopods, 5 anostracans, 2 notostracans and 3conchostracans (all belonging to the Spinicaudata group). This diversity represents 53% of the species known in Yugoslavia. Community structure, spatial distribution and seasonal dynamics of the species were analyzed over the period 1991–1995. More than one species was found in about 67% of ponds. The maximum number of coexisting species was 7.Anostracans were the most common as a group, while the notostracan Triops cancriformis and the conchostracan Leptestheria saetosa were the most frequent species. A high frequency of coexistence was established. Regarding community structure, two significantly different periods during the season were distinguished: the first, from the end of February to mid-April, and the second, from mid-April to the end of July. This extensive pool longevity may underlie the high diversity in this region. This revised version was published online in August 2006 with corrections to the Cover Date.  相似文献

The investigations of Anostraca, Notostraca and Conchostraca in Yugoslavia began after a great delay compared with other European countries. Intensive systematic investigations date back only to the 1970s.To date, 19 species have been recorded. All species, with the exception of Artemia salina, are faunal elements of the Pannonian region. Seven species, including Artemia salina, Branchinecta orientalis, Branchipus serbicus, Leptestheria dahalacensis, Limnadia lenticularis, Imnadia cristata and Imnadia panonica are known from single localities. Six species including Branchinecta ferox, Chirocephalus brevipalpis, Chirocephalus carnuntanus, Lepidurus apus, Eoleptestheria spinosa and Imnadia banatica have restricted distributions. Streptocephalus torvicornis and Cyzicus tetracerus are known from several localities, while Branchipus schaefferi, Chirocephalus diaphanus, Triops cancriformis and Leptestheria saetosaare common. Large branchiopods are mainly confined to anthropogenic landscapes, especially in the northern part of the country. Rare species inhabiting a single pond, or those with rather restricted distributions, are the most endangered and such species account for about 70% of the fauna. Limnadia lenticularis, Imnadia cristata and I. panonica have not been found for more than 20 years and they are probably extinct in Yugoslavia. Branchipus serbicus, Imnadia cristata and panonica have not been documented since being originally described. Eoleptestheria spinosa is currently the most endangered species since it only appears irregularly in small populations. At present, Branchinecta ferox, Chirocephalus carnuntanus and Imnadia banatica are the safest of the rare species as they regularly appear in large numbers. An antimicrobial finishing for cotton fabric was prepared from commercial (iSys AG, Germany) silver chloride (Ag) dispersed at different concentrations in a reactive organic–inorganic binder (RB) (iSys MTX (CHT, Germany). Pad-dry-cure and exhaustion methods were used for the sols application, giving Ag-RB coating with Ag concentration from ca. 48 to ca. 290 ppm on the cotton fabric. The presence of silver on the cotton finishes was confirmed by measuring its concentration in the fabrics with the help of inductively coupled plasma mass spectroscopy (ICP-MS). The morphology of the finished fabrics was investigated by SEM, while their composition was established from EDXS measurements combined with the results of FT-IR spectral analysis. The antimicrobial activity of variously treated cotton fabrics was assessed before and after repetitive (up to 10×) washing by the application of standard tests: for the fungi Aspergillus niger (ATCC 6275) and Chaetomium globosum (ATCC 6205) by the modified DIN 53931 standard method, while the presence of Gram-negative bacterium Escherichia coli (ATCC 25922) was followed by using ISO 20645:2004 (E) and AATCC 100-1999 standard methods. Results revealed that the antimicrobial activity of the coatings strongly depended on the concentration of Ag in the corresponding Ag-RB dispersions, indirectly depending on the preparation method (pad-dry-cure vs. exhaustion) and that the Ag-RB coatings were more effective for bacteria than for fungi. The Ag concentrations on the cotton fabrics achieved by the pad-dry-cure method (48 and 52 ppm) were not sufficient to impart satisfactory antifungal activity to the cotton fabrics, though they assured excellent reduction of the bacterium E. coli (98–100%). A minimal inhibitory concentration of Ag in the coating providing a sufficient bacterial reduction of 60% was ca. 24 ppm. Effective antifungal activity was achieved only by applying the exhaustion method, enabling high initial Ag concentration in the Ag-RB coating (>100 ppm). The antibacterial activity depended on the washing treatment. No antifungal activity was noted for washed cotton fabric, even those with highly concentrated Ag (290 ppm) in the Ag-RB coating, but a 94% bacterial reduction was obtained for the corresponding cotton fabric, after 10 repetitive washings, corroborated by the Ag concentration on washed fabric of about 65 ppm.  相似文献
