Axial patterning of the aboral end of the hydra body column was examined using expression data from two genes. One, shin guard, is a novel receptor protein-tyrosine kinase gene expressed in the ectoderm of the peduncle, the end of the body column adjacent to the basal disk. The other gene, manacle, is a paired-like homeobox gene expressed in differentiating basal disk ectoderm. During regeneration of the aboral end, expression of manacle precedes that of shin guard. This result is consistent with a requirement for induction of peduncle tissue by basal disk tissue. Our data contrast with data on regeneration of the oral end. During oral end regeneration, markers for tissue of the tentacles, which lie below the extreme oral end (the hypostome), are detected first. Later, markers for the hypostome itself appear at the regenerating tip, with tentacle markers displaced to the region below. Additional evidence that tissue can form basal disk without passing through a stage as peduncle tissue comes from LiCl-induced formation of patches of ectopic basal disk tissue. While manacle is ectopically expressed during formation of basal disk patches, shin guard is not. The genes examined also provide new information on development of the aboral end in buds. Although adult hydra are radially symmetrical, expression of both genes in the bud's aboral end is initially asymmetrical, appearing first on the side of the bud closest to the parent's basal disk. The asymmetry can be explained by differences in positional information in the body column tissue that evaginates to form a bud. As predicted by this hypothesis, grafts reversing the orientation of evaginating body column tissue also reverse the orientation of asymmetrical gene expression. 相似文献

Systematic conservation planning requires spatial information on biodiversity. Such information is often unavailable, forcing spatial planning to rely on assumed relationships between species and environmental features. This problem is particularly acute in large, remote marine protected areas that are proliferating rapidly. Here, we use models to predict whether (a) macrobenthic biodiversity across four taxa (gorgonians, soft corals, hard corals, and sponges) with different life histories are congruent within seascape features through regional space; and (b) models generated in an intensively-sampled area in one region can predict the occurrence of habitat-forming macrobenthos in neighboring ones. All four taxa studied showed similar habitat preferences, but high variability in distributions among and within features suggesting factors other than simple geomorphology influence these regional biodiversity patterns. Nonetheless, models derived from one region accurately predicted the presence and absence of the same taxa hundreds of kilometers away. This transferability of models of species occurrences has the potential to deliver improved reserve design in data-deficient regions. 相似文献

The discovery of deleterious mutations in the breast and ovarian cancer susceptibility genes, BRCA1 and BRCA2, has facilitated the identification of individuals at particularly high risk of these diseases. There is a wide variation between populations in the prevalence and related risks of various types of BRCA1/2 mutations, so estimates cannot be extrapolated to Canadians, especially not founder populations such as French- Canadians. Polymerase chain reaction (PCR)-based methods were used to detect the majority of these mutations. These approaches usually failed to detect large DNA rearrangements, which have been claimed to be involved in other populations in 5% to up to 36% of BRCA1-positive families. There is very little information about the contribution of this type of mutation in BRCA2-positive families. To investigate if our available mutation spectrum of BRCA1 and BRCA2 in high-risk French-Canadian breast/ovarian cancer families has been biased by PCR-based direct sequencing methods, we first used Southern blot analysis to test DNA samples from 61 affected/obligate carrier individuals from 58 families in which no BRCA1/2 deleterious mutation was found. Finally, 154 individuals from 135 BRCA1/2 nonconclusive families, including all those tested previously by Southern blot analysis, were tested with the new multiplex ligation probe amplification (MLPA) technique. These approaches failed to detect any rearrangement. Moreover, if the frequency of MLPA-detectable rearrangements in our cohort of 135 BRCA1/2 nonconclusive families was 2.2% or higher, we would have had a 95% or greater chance of observing at least one such rearrangement. Abstract. Moisture and nutrient gradients consistently explain much of the variation in plant species composition and abundance, but these gradients are not spatially explicit and only reveal species responses to resource levels. This study links these abstract gradients to quantitative, spatial models of hill‐slope assembly. A gradient analysis in the mixed‐wood boreal forest demonstrates that patterns of upland vegetation distribution are correlated to soil moisture and nutrient gradients. Variation in species abundance with time since the last fire is removed from the gradient analysis in order to avoid confounding the physical environment gradients. The physical‐environment gradients are related to qualitative positions on the hill slope i.e. crest, mid‐slope, bottom‐slope. However, hill‐slope shape can be quantitatively described and compared by fitting allometric equations to the slope profiles. Using these equations, we show that hill‐slope profiles on similar surficial materials have similar parameters, despite coming from widely separated locations. We then quantitatively link the moisture and nutrient gradients to the equations. Moisture and nutrients significantly increase as distance down‐slope from the ridgeline increases. Corresponding vegetation composition changes too. These relationships characterize the general pattern of vegetation change down most hill slopes in the area. Since hill slopes are a universal feature of all landscapes, these principles may characterize landscape scale spatial patterns of vegetation in many environments. 相似文献

An early event in lung oncogenesis is loss of the tumour suppressor gene LIMD1 (LIM domains containing 1); this encodes a scaffold protein, which suppresses tumorigenesis via a number of different mechanisms. Approximately 45% of non-small cell lung cancers (NSCLC) are deficient in LIMD1, yet this subtype of NSCLC has been overlooked in preclinical and clinical investigations. Defining therapeutic targets in these LIMD1 loss-of-function patients is difficult due to a lack of ‘druggable’ targets, thus alternative approaches are required. To this end, we performed the first drug repurposing screen to identify compounds that confer synthetic lethality with LIMD1 loss in NSCLC cells. PF-477736 was shown to selectively target LIMD1-deficient cells in vitro through inhibition of multiple kinases, inducing cell death via apoptosis. Furthermore, PF-477736 was effective in treating LIMD1−/− tumours in subcutaneous xenograft models, with no significant effect in LIMD1+/+ cells. We have identified a novel drug tool with significant preclinical characterisation that serves as an excellent candidate to explore and define LIMD1-deficient cancers as a new therapeutic subgroup of critical unmet need.Subject terms: Targeted therapies, Non-small-cell lung cancer 相似文献

Abstract Opportunistic sightings and strandings of Caperea marginata (n=196) from the vicinity of Australia and New Zealand (1884 to early 2007) were used to relate geographic and temporal patterns to oceanographic and broad-scale climatic variability. Records were not uniformly distributed along the coast and more (69%) were from Australia than New Zealand. Seven coastal whale ‘hotspots’ were identified which accounted for 61% of records with locality data. Half of the hotspot records were from southeast (37) and northwest (20) Tasmania—others each had 9–15 events. Upwelling and/or high zooplankton abundance has been documented near all whale hotspots. Records of C. marginata occurred in all months, with 75% in spring and summer. Inter-annual variability showed broad agreement between increased whale records (usually in spring/summer) and strongly positive ‘Niño 3.4’ during 1980–1995 but not thereafter. Coastal upwelling and productivity increase during climatic phenomena such as El Niño and are likely to be quickly beneficial to plankton-feeding whales such as C. marginata. 相似文献

Allosteric Activation of Na-Ca Exchange by L-Type Ca Current Augments the Trigger Flux for SR Ca Release in Ventricular Myocytes

The possible contribution of Na+-Ca2+ exchange to the triggering of Ca2+ release from the sarcoplasmic reticulum in ventricular cells remains unresolved. To gain insight into this issue, we measured the “trigger flux” of Ca2+ crossing the cell membrane in rabbit ventricular myocytes with Ca2+ release disabled pharmacologically. Under conditions that promote Ca2+ entry via Na+-Ca2+ exchange, internal [Na+] (10 mM), and positive membrane potential, the Ca2+ trigger flux (measured using a fluorescent Ca2+ indicator) was much greater than the Ca2+ flux through the L-type Ca2+ channel, indicating a significant contribution from Na+-Ca2+ exchange to the trigger flux. The difference between total trigger flux and flux through L-type Ca2+ channels was assessed by whole-cell patch-clamp recordings of Ca2+ current and complementary experiments in which internal [Na+] was reduced. However, Ca2+ entry via Na+-Ca2+ exchange measured in the absence of L-type Ca2+ current was considerably smaller than the amount inferred from the trigger flux measurements. From these results, we surmise that openings of L-type Ca2+ channels increase [Ca2+] near Na+-Ca2+ exchanger molecules and activate this protein. These results help to resolve seemingly contradictory results obtained previously and have implications for our understanding of the triggering of Ca2+ release in heart cells under various conditions. 相似文献

Parasitic worms survive within their immunocompetent hosts by modulating their immune system and by inhibiting inflammatory responses directed against the parasites. This immunomodulation has a spill over effect and also inhibits inflammatory responses originating from other causes. For this reason, persons who are infected with certain species of worms show a lower rate of allergic diseases as compared to persons who are free of parasites. In the same line, studies in mouse models revealed that many inflammatory diseases can be treated by worm infections. This effect is among others owing to specific proteins that are released by the worms. Such secreted immunomodulators, shaped by co‐evolution between parasites and their hosts, could become lead compounds for the development of new therapies against allergic and inflammatory diseases. 相似文献

Electrospinning is a highly adaptable method producing porous 3D fibrous scaffolds that can be exploited in in vitro cell culture. Alterations to intrinsic parameters within the process allow a high degree of control over scaffold characteristics including fiber diameter, alignment and porosity. By developing scaffolds with similar dimensions and topographies to organ- or tissue-specific extracellular matrices (ECM), micro-environments representative to those that cells are exposed to in situ can be created. The airway bronchiole wall, comprised of three main micro-environments, was selected as a model tissue. Using decellularized airway ECM as a guide, we electrospun the non-degradable polymer, polyethylene terephthalate (PET), by three different protocols to produce three individual electrospun scaffolds optimized for epithelial, fibroblast or smooth muscle cell-culture. Using a commercially available bioreactor system, we stably co-cultured the three cell-types to provide an in vitro model of the airway wall over an extended time period.This model highlights the potential for such methods being employed in in vitro diagnostic studies investigating important inter-cellular cross-talk mechanisms or assessing novel pharmaceutical targets, by providing a relevant platform to allow the culture of fully differentiated adult cells within 3D, tissue-specific environments. 相似文献
