The expression of penicillin acylase (PA), cloned in the pPA102 plasmid under control of the wild-type lac promoter and using galactose as inducer in Escherichia coli JM101, JM103 and JM105 transformant cells, was analyzed. The E. coli JM101/pPA102 cultures attained the highest specific activity of PA. For large scale PA production based on E. coli JM101/pPA102 a culture media with galactose instead of isopropyl-thio-galactopyranoside as inducer would be as successful and less expensive.  相似文献

The Sierra de La Laguna, in the southern part of the state of Baja California Sur, Mexico, was decreed a biosphere reserve in 1994. It harbors relict plant communities with a high level of endemism. Floristic research has established that the two main communities, the oak–pine woodland (OPW) and the dry tropical forest (TDF), harbor 43 and 33 endemic species, respectively. We segregate the reserve surface into seven major habitats and each endemic species were assigned a hierarchic value into an abundance-distribution scale, useful as dominance ranking. Using such scale, we found that almost half of the species are in the lowest rank (low abundance and aggregate distribution). Using GIS, we obtain the surface for each habitat, where mountainsides represent the largest in the reserve area. We obtained an index of importance from the proportion of endemic species by habitat and its surface occupied, founding that, in spite of the small and fragmented nature of the upland riparian habitats, they have the greatest index value. Consequently, for future conservation plans in the reserve, these habitats should be considered remarkable sites, meriting better attention to ensure endemic plant preservation. Also, since 86% of the endemic species appear in one, two or three habitats, a high dependency on particular environmental conditions is suggested for them.  相似文献

The Cactaceae is considered one of the most threatened taxa in the world. However, the extent to which climate change could compromise the conservation status of this group has rarely been investigated. The present study advances this issue under three specific aims: (1) to assess the impact of climate change on the distribution of endemic cacti species in the Baja California Peninsula (n?=?40), (2) to study how the impact of climate change is distributed in this group according to the species’ conservation status, and (3) to analyze how these impacts are organized from a biogeographical and functional perspective. We addressed these objectives under three socioeconomic emission pathways (RCP 2.6, 4.5, and 8.5), and using two extreme migration scenarios: full climate change tracking and no migration. Altogether, all socioeconomic emission pathways under the two extreme migration scenarios show consistency regarding the identity of the species most vulnerable to climate change, and depict a discrepant future scenario that has, on one hand, species with large potential habitat gains/stability (winners); and on the other, species with large habitat reductions (losers). Our work indicates that winner species have a tropical affinity, globose growth, and includes most of the currently threatened species, whereas loser ones are in arid and Mediterranean systems and are mostly non-threatened. Thus, current and future threat factors do not overlap in the biogeographic and taxonomic space. That reveals a worrisome horizon at supraspecific levels in the study area, since the total number of threatened species in the future might largely increase.

Abstract. Our main objective was to use comparative floristic and structural criteria in order to understand the differences in assemblages brought about by the meeting of arid and dry‐tropical environments. We sampled 33 vegetation plots and recorded all perennial species. The data were subjected to multivariate analysis, including Principal Component Analysis (PCA). The floristic variation detected by the PCA was strongly related to altitude and rainfall. The gradient detected by the ordination procedure had Tropical deciduous forest (TDF) and Sonoran Desert (SD) at opposite extremes and Cape sarcocaulescent shrubland (CSCS) in an intermediate position. The numerical classification algorithm detected six distinct groups of species with clearly identifiable field distributions. Vegetation structure also differed significantly between the regions. SD had the lowest species‐richness (α‐diversity) and inter‐site similarity in the CSCS was highest, indicating that this environment is relatively uniform and has low species turnover (β‐diversity). It is concluded that the CSCS is distinctly different from the sarcocaulescent shrublands of the more northern plant communities of the peninsula, where Sonoran Desert floristic affinities prevail. The floristic composition of the CSCS is rich in dry‐tropical affinities, its species richness is higher, it is homogeneous in its species content and turnover and is more dense with a taller canopy than the northern desert scrubs. It is proposed that the boundary line separating the Cape sediments of granitic origin from the basalt‐derived sediments of the northern‐lying Sierra de la Giganta should be used as an easily identifiable landscape trait to delimitate this unique community.  相似文献
