ABSTRACT

The southern part of the Tamar valley area in SW England is highly mineralised and mines in the region were the world's principal producers of tin, copper and arsenic during the mid nineteenth century. The Devon Great Consols Mine, covering 67.6 ha (167 acres) is situated in this area. Residues from the mining activity resulted in unvegetated spoil tips and local soils highly contaminated with As (range 120–52600 μg/g As). Sequential chemical extraction procedures were conducted on eight surface samples (0–15 cm) taken from a 2.0 km long transect from within the mine site to agricultural grassland. The proportion of water extractable As in agricultural top soils was lower (0.05–0.3%) than the values obtained for mine wastes (0.02–1.2%). Arsenic was found to be concentrated in the Fe-organic and residual fractions, which accounted for up 93 % of the total As in mine spoil and nearby soils.  相似文献

The effects of azalea lace bug, Stephanitis pyrioides (Scott), feeding injury on azalea growth and development were investigated using 'Girard's Rose' azaleas during a 2-yr field study in Georgia Low, medium, and high injury treatments, which corresponded to 6, 8, and 14% maximum canopy area injury, were compared with control azaleas that received no lace bug infestation. Flower number, whole-shrub leaf and stem dry mass, and dry mass and size of new growth tissues were unaffected by treatments. In contrast, growth index measurements, a general measure of variability frequently used for horticultural differentiation, showed significant reductions for all treatments in comparison to control azaleas after 20 wk. Though not directly quantified, this apparent discrepancy may be explained as an artifact of lace bug feeding-induced leaf abscission. Growth index measurements had considerable variability and may not be the most reliable measurement of size. In July 1998, plant canopy densities among azaleas maintained in the high injury treatments were approximately 15% less full than the canopies of control shrubs. Predaceous insects had a significant negative association with azalea lace bug number during the 2-yr study. Flower and new tissue production, measured destructively during two growing seasons, revealed azalea tolerance to 14% of maximum canopy area lace bug feeding-injury levels.  相似文献

Potential resistance to the twolined spittlebug, Prosapia bicincta (Say), was evaluated among 56 turfgrass genotypes. Greenhouse, laboratory, and field bioassays identified differences in spittlebug survival and development, host preference and damage levels, and turfgrass tolerance to and ability to recover from pest induced injury. All centipede grasses demonstrated high levels of susceptibility, followed by bermudagrasses, seashore paspalums, and zoysiagrasses. Average nymphal survival to the adult stage ranged from 1.5 to 78.1%. Development required 38.1-62.0 d under greenhouse conditions, depending on plant taxa. Among seashore paspalums, nymphal survival to the adult stage was lowest and duration of development was longest on HI-1, 'Sea Isle 2000', 561-79, and 'Mauna Kea'. Reduced spittlebug survival and increased developmental times were also observed on the bermudagrasses BERPC 91-15 and 'Tifway'. Although zoysiagrasses supported spittlebug development and survival to the adult stage, developmental times were extended on the zoysiagrass cultivars 'Emerald' and 'El Toro'. Spittlebug preference varied with generation evaluated. First-generation spittlebugs inflicted the greatest damage on TC201 (centipede grass), 'Primavera' (bermudagrass), and 'Emerald' (zoysiagrass) in choice tests. In the fall, second-generation spittlebugs damaged TC201 (centipedegrass) and 'Sea Isle 1' (paspalum) most severely, whereas 561-79 (paspalum) and 'Emerald'(zoysiagrass) were less severely affected. Among taxa included in field trials, HI-1, 'Mauna Kea', 'Sea Isle 2000',and AP-14 paspalums, 'Tifway' bermudagrass, and 'Emerald' zoysiagrass were most tolerant (demonstrated the best regrowth potential following twolined spittlebug feeding).  相似文献

Several restriction sites in the cytochrome oxidase I gene of fall armyworm, Spodoptera frugiperda (J.E. Smith), were identified by sequence analysis as potentially being specific to one of the two host strains. Strain specificity was demonstrated for populations in Florida, Texas, Mississippi, Georgia, and North Carolina, with an AciI and SacI site specific to the rice (Oryjza spp.)-strain and a BsmI and HinfI site joining an already characterized MspI site as diagnostic of the corn (Zea mays L.)-strain. All four of these sites can be detected by digestion of a single 568-bp polymerase chain reaction-amplified fragment, but the use of two enzymes in separate digests was found to provide accurate and rapid determination of strain identity. The effectiveness of this method was demonstrated by the analysis of almost 200 adult and larval specimens from the Mississippi delta region. The results indicated that the corn-strain is likely to be the primary strain infesting cotton (Gossypium spp.) and that an unexpected outbreak of fall armyworm on the ornamental tree Paulownia tomentosa (Thunb.) Sieb. & Zucc. ex Steud. was due almost entirely to the rice-strain.  相似文献

Prosapia bicincta (Say) (Hemiptera: Cercopidae), the twolined spittlebug, is an economic pest of turfgrass in the southeastern United States. No data concerning natural enemies of P. bicincta in turfgrass have been reported previously. We compared predation of spittlebug eggs, nymphs, and adults in the laboratory by potential generalist predators commonly found in turfgrass: bigeyed bugs Geocoris uliginosus Say and Geocoris punctipes Say; red imported fire ant, Solenopsis invicta Buren; wolf spiders (Lycosa sp. Walckenaer); carabid beetles Harpalaus pensylvanicus DeGeer and Calosoma sayi Dejean; and tiger beetles Megacephala carolina carolina L. Eggs were readily consumed by generalist predators. S. invicta consumed 100% of the eggs offered. H. pensylvanicus and C. sayi were also significant predators of P. bicincta eggs. Nymphs live in spittlemasses that protect them from attack by predators, but exposed nymphs were susceptible to attack when mechanically removed from their spittlemasses. S. invicta and M. carolina carolina caused significant mortality of exposed nymphs. P. bicincta adults are aposematic and have the ability to reflex bleed; however, reflex bleeding did not prevent attack by predators. S. invicta and M. carolina carolina killed 100% of the adult spittlebugs offered in laboratory bioassays. Lycosa sp. are less voracious predators of adults. Sound background knowledge about P. bicincta and its potential natural enemy complex is important for the development and implementation of a detailed, site-specific, biologically based pest management program in turfgrass.  相似文献

An improved radioactive assay has been developed for serine transhydroxymethylase. This assay involves the direct measurement of the [14C]HCH0 which is generated when [3-14C]-serine is employed as the substrate. The new assay eliminates 14 the need for a solvent extraction of a [C]HCHO-dimedon adduct which is the basis of the assay devised by Taylor and Weissbach.

The enzyme has been purified employing Affi-Gel Blue. The purified enzyme retains full activity when bound to this affinity chromatography matrix and can be stored in this state at 4° indefinitely.  相似文献

Healthy hemlock trees, Tsuga canadensis (L.) Carrière, and hemlock woolly adelgid, Adelges tsugae Annand (Hemiptera: Adelgidae), populations should favor retention and population growth of adelgid predators such as Laricobius nigrinus Fender (Coleoptera: Derodontidae) and Sasajiscymnus tsugae (Sasaji & McClure) (Coleoptera: Coccinellidae). Eastern hemlock trees between 15 and 38 cm diameter at breast height (dbh) were treated with 0, 10, or 25% of 1.5 g imidacloprid (Merit 75 WP) per 2.5 cm dbh and were either fertilized or not, in a 3 by 2 factorial design. After 2 yr, imidacloprid reduced the numbers of ovisacs and eggs found on trees in a dosage-dependent manner, while enhancing tree growth parameters such as new shoots or needles and the length of new shoots. Fertilized trees had greater adelgid fecundity, which was positively correlated with total foliar N in both winter generations. In February 2009 (27 mo after imidacloprid treatment), higher imidacloprid dosages to unfertilized trees resulted in reduced adelgid fecundity. Concentrations of N, P, and K were higher in the foliage of trees treated with insecticide, whereas foliar aluminum concentrations were consistently lower in trees with higher insecticide dosages. Trees treated with low rates of imidacloprid were healthier than untreated trees, but only trees treated with the 0.1 x dosage had sufficient adelgids to possibly sustain predators over extended periods.  相似文献

As part of our genome-mapping effort, we undertook a large-scale screening study to identify RFLPs useful as genetic markers. Some 1,664 single-copy or repeat-containing phage clones from a Charon 4A genomic library were tested for polymorphism against a panel of DNAs, from five unrelated individuals, digested with eight restriction enzymes. Approximately 30% (515) of the clones revealed polymorphism by Southern hybridization; 67 loci detected had PIC values greater than .5. Restriction enzymes MspI, TaqI, and RsaI were most efficient in detecting polymorphism within the 1-20-kb-fragment size range resolved. With only one exception each of the clones detected polymorphism originating from a single locus.  相似文献

Ten cultivars of seashore paspalum, Paspalum vaginatum Swartz, were compared for their response to Japanese beetle, Popillia japonica Newman, larval root feeding. Cultivars of Bermuda grass, Cynodon sp., and zoysiagrass, Zoysia sp., also were included for comparison. Turf grown in pots in the greenhouse was infested with second and third instars in this 2-yr study. Grub survival and weight gain, foliar growth, and root loss were compared among turfgrass species and cultivars. Few species-related differences were identified. Differences in grub tolerance were, however, observed to be a function of turfgrass cultivar. Some turf types demonstrating tolerance to grub feeding had rapid root growth and high root mass in control pots, but this was not consistent for all cultivars showing enhanced ability to maintain foliar growth despite grub feeding. The paspalum cultivars that seemed most tolerant of grub feeding were '561-79', 'Sea Isle 2000', 'Durban', 'HI-10', 'Kim-1', 'Sea Dwarf', and 'Sea Spray'.  相似文献
