Absorption of iron has been followed after the intramuscular injection of 12 piglets with iron-dextrin and iron-sorbitol, labelled with Fe59. Disappearance from the site of injection was followed by a method developed for external radioactivity measurements. Blood levels of Fe59 were measured at intervals during a period of 51 days. The tissue distribution pattern of Fe59 was examined in six of the piglets after 30 days. Both iron-dextrin and iron-sorbitol were rapidly absorbed, iron-sorbitol more rapidly. After 20 minutes, 50 per cent of the iron-sorbitol had been absorbed and less than one per cent remained at the site of injection after 48 hours. The amount of Fe59 remaining at the site of injection of iron-dextrin had decreased by half by 60 minutes but some ten per cent had still not been absorbed after ten days. When given as iron-dextrin (94 mg Fe per piglet), the level of Fe59 in the blood remained fairly constant for 14 days. On piglets given iron-sorbitol, (100 mg Fe each) the blood level declined steadily. The amount of Fe59 in the liver, spleen and bone marrow was much greater 30 days after the injection of iron-dextrin than after iron-sorbitol. The relation was reversed in the kidneys, presumably because of the greater excretion of iron-sorbitol in the urine.  相似文献

This paper describes the improvement in the use of electrical impedance spectroscopy (EIS) for animal cell concentration monitoring of adherent cultures by using a four-electrode configuration instead of the commonly used two-electrode configuration. This four-electrode configuration prevents cell concentration measurements from external masking effects such as the electrode covering ratio, the degree of cellular adherence to the electrodes and the impedance of the measuring electrodes. Cell concentration was monitored using both four-electrode and two-electrode configurations in vero cell and human mesenchymal stem cell cultures in order to analyze the attained improvement in two cell lines with opposite growth characteristics. The experiments performed with vero cell cultures evidenced that the four-electrode configuration enables cell concentration measurements along all culture phases, even once the culture reached cell confluence (over 2×10(5) cells/cm(2)), confirming that this configuration is less effected by all the external influences. The experiments performed with human mesenchymal stem cells demonstrated good sensitivity of the measurement at very low cell concentrations, as well as a very good robustness all over the 12-days experiment. Finally, off-line cell measurements during cell cultures proved good accuracy of impedance measurements carried out with a four-electrode configuration along all cell growth phases, enabling determination of relevant cell growth parameters.  相似文献

Excitability in muscle cells manifests itself as contractility and may be evoked by electrical stimulation. Here we describe an electrical stimulator device applicable to cells seeded on standard multiwell plates and demonstrate how it effectively stimulates synchronous contraction of skeletal muscle C2C12 cells without damaging them. The electrical stimulator of cultured cells (ESCC) consists of two connection cards and a network of platinum electrodes positioned in such way that each well in a row is uniformly stimulated. The ESCC may produce a range of outputs based on the stimulation parameters it receives from a commercial pulse generator and can be placed in a standard cell incubator, allowing for long-term stimulation as required for biochemical and molecular biological assays. We show that a 90-min stimulation of C2C12 myotubes at 50 V, 30 ms of pulse duration, and 3 Hz of frequency enhances glucose metabolism and glycogen mobilization while oppositely modulating the activity ratio of glycogen metabolizing enzymes. Thus, we demonstrate that long-term electrical stimulation of C2C12 myotubes with the ESCC results in contractility and metabolic changes, as seen in exercising muscle.  相似文献

The application of impedance spectroscopy to estimate on-line cell concentration was studied. The estimation was based on the relative variation between electrical impedance measured at low (10 kHz) and high frequencies (10 MHz). Studies were carried out to characterise the influence of changes in physical and chemical parameters on the impedance measurement. Two different possibilities to perform on-line measurements were tested: a simple set-up, based on an in situ probe, gave good results but was not suitable for high agitation and aeration rates. An ex situ flow-through on-line measuring cell was used to overcome these problems, showing a better performance. The use of this set-up for the growth monitorisation of a Saccharomyces cerevisiae culture showed an efficient performance, having the correlation between estimated and measured S. cerevisiae a Pearson coefficient of 0.999.  相似文献

Long-term records of environmental history at decadal to millennial time-scales enable an assessment of ecosystem variability and responses to past anthropogenic disturbances and are fundamental for the development of environmental management strategies. This study examines the local variability of land-use history in the South Swedish Uplands over the last 200 years based on pollen records from three lake-sediment successions. Temporal changes in the proportional cover of 14 plant taxa were quantified as percentages using the landscape reconstruction algorithm (LRA). The LRA-based estimates of the extent of four land-use categories (cropland, meadows/grassland, wetland, outland/woodland) were compared to corresponding estimates based on historical maps and aerial photographs from ad 1769–1823, 1837–1895, 1946 and 2005. Although the LRA approach tends to overestimate grassland cover by 10–30 % for the two earliest time periods, the reconstructed vegetation composition is generally in good agreement with estimates based on the historical records. Subsequently, the LRA approach was used to reconstruct the 200-year history of local land-use dynamics at 20-year intervals around two small lakes. The qualitative assessment of difference approach, which requires fewer assumptions and parameters than LRA for objective evaluation of between-site differences in plant abundances, provides consistent results in general. Significant differences exist in the land-use history between the sites. Local catchment characteristics, such as soil conditions and wetland cover, appear important for the development of human impact on the landscape. Quantifications of past vegetation dynamics provide information on the amplitude, frequency and duration of the land-use changes and their effects on terrestrial and aquatic ecosystems, and should be taken into account when nature conservation strategies are developed.  相似文献

In situ electroporation of adherent cells provides significant advantages with respect to electroporation systems for suspension cells, such as causing minimal stress to cultured cells and simplifying and saving several steps within the process. In this study, a new electrode assembly design is shown and applied to in situ electroporate adherent cell lines growing in standard multiwell plates. We designed an interdigitated array of electrodes patterned on copper with printed circuit board technology and covered with nickel/gold. Small interelectrode distances were used to achieve effective electroporation with low voltages. Epoxy-based microseparators were constructed to avoid direct contact with the cells and to create more uniform electric fields. The device was successful in the electropermeabilization of two different adherent cell lines, C2C12 and HEK 293, as assessed by the intracellular delivery of the fluorescent dextran FD20S. Additionally, as a collateral effect, we observed cell electrofusion in HEK 293 cells, thus making this device also useful for performing cell fusion. In summary, we show the effectiveness of this minimally invasive device for electroporation of adherent cells cultured in standard multiwell plates. The cheap technologies used in the fabrication process of the electrode assembly indicate potential use as a low-cost, disposable device.  相似文献

A simple disposable six minibioreactor system has been developed in order to perform multiple cell culture experiments in parallel, as a tool to accelerate experimentation in cell culture optimization. The system consists of a fixed part containing all instrumentation, sensors and actuators, and a disposable part, a compact unit with six minibioreactors with 10–15 mL of working volume each. This single-use unit is made of transparent biocompatible plastic material (polystyrene). Each one of the minibioreactors is equipped with agitation, headspace aeration supply and two optical probes, one for total cells measurement and pH, and another for dissolved oxygen measurement (and consequently the evaluation of Oxygen Uptake Rate, OUR). As an example of application, the performance of the system is successfully demonstrated for the culture of hybridoma cells growing in suspension under different conditions. The results allowed confirming the reproducibility of the system, and the feasibility to follow-up continuously the differences in cell growth, pH, pO2 and OUR evolution when hybridoma cells are cultured in different experimental conditions. For this, three different sets of experiments are considered. First, the use of the same culture medium DMEM supplemented with 10% fetal calf serum in all six minibioreactors. Second, the use of DMEM supplemented with three different FCS percentages (0, 1 and 10%) in two minibioreactors each. Third, the use of two different media (DMEM supplemented with 10% FCS and a chemically defined medium) in three minibioreactors each.  相似文献

A pollen record obtained from a 2.2-m sediment succession deposited in a small lake in the province of Västerbotten, north-eastern Sweden, reveals the presence of continuous forest cover since 8,500 calendar years before present (cal b.p.). Forest with abundant Pinus (pine) and Betula (birch) initially colonized the area, followed by a dominance of deciduous trees, primarily Betula, from ca. 8,000 to ca. 3,200 cal b.p. Pollen accumulation rates of Quercus (oak), Ulmus (elm) and Tilia (linden) suggest the possible local presence of these thermophilous tree species during this period. The climate gradually became colder and moister around 3,500 cal b.p. and an increased abundance of Sphagnum spores indicates paludification. Picea (spruce) became established around 3,200 cal b.p. and less than 500 years later this was the dominant tree species around the lake. The fire frequency as inferred from charcoal particles exhibits a general increase from ca. 3,000 cal b.p. with subsequent charcoal accumulation maxima at around 2,800 cal b.p., 1,700 cal b.p. and in recent time. The human influence on vegetation was significant during the last 200–300 years. Soil erosion increased substantially and fern spores amount to ca. 55% of the total pollen assemblage in the uppermost samples. These results suggest an extensive anthropogenic impact on the local forest ecosystem, with abundant logging, burning and ditching in the vicinity of the lake. Independent evidence of sub-recent human-induced environmental change is provided by historical accounts. Complementary information on catchment soil development and aquatic nutrient status was provided by records of magnetic susceptibility and elemental carbon, and nitrogen contents obtained from the same sediment core.  相似文献
