Respiration and soluble sugar metabolism in sugar pine embryos   总被引:1,自引:0,他引:1  
Embroys excised from dormant seeds of sugar pine ( Pinus lambertiana Dougl.) incubated at 25°C (non-dormancy-breaking) or stratified at 5°C (dormancy-breaking) were analyzed to determine temperature effects on the relative activities of respiration and fermentative metabolism, the levels of soluble sugers and the activities of the hydrolytic enzymes, invertase and sucrose synthase, as related to the release of dormancy and germinatio. At 25°C, despite a sharp drop in embryo oxygen uptake after 48 h, a simultaneous decline in acetaldehyde and ethanol concentrations indicated that there was not a shift to fermentative metabolism. The concentrations of soluble sugars showed no treatment effects. Embryo invertase (EC 3.2.1.26) activity changed only slightly at either temperature, while stratification was accompanied by a 4-fold increase in sucrose synthase (EC 2.4.1.13) activity (cleavage direction). Upon transfer of stratified seeds to 25°C, embryo sucrose synthase activity rapidly increased almost 10-fold, with the increase beginning prior to germination, while mvertase activity increased 20-fold, concomitant with germination.  相似文献

The critical segment for the Langer-Giedion syndrome: 8q24.11----q24.12   总被引:2,自引:0,他引:2  
An 18-year-old intellectually normal male with characteristic features of the Langer-Giedion syndrome is reported. High resolution chromosome analysis showed a small deletion in the region of bands 8q24.11 and 8q24.12 in addition to an apparently balanced de novo translocation (2;9)(q21;q13). This finding provides additional information on the minimum deleted segment required to produce the Langer-Giedion syndrome and may indicate that deletions of this size or smaller are not necessarily associated with mental retardation.  相似文献

Characterization of the proteins and nucleic acid of the gypsy moth nuclear polyhedrosis virus isolated in Ithaca, N.Y. (LdNPV-IT) is presented. A total of 29 viral structural proteins were identified by sodium dodecyl sulfate-polyacrylamide gel electrophoresis when the virus was isolated in the absence of alkaline protease activity. Fourteen surface envelope viral proteins were identified by lactoperoxidase iodination. Eleven proteins were associated with nucleocapsids prepared by Nonidet P-40 detergent treatment. Distinct alterations of viral proteins were documented when virions were purified in the presence of occlusion body-associated alkaline protease(s). Restriction enzyme digests of viral DNA indicated that this isolate was composed of a large number of genetic variants. On the basis of the major molar fragments resulting from EcoRI, BamHI, BglII, and HindIII digests, the molecular weight of the LdNPV genome was approximately 88 × 106.  相似文献

An in vitro study of digestion by the subterranean larva of S. geminata revealed the presence of enzymes able to digest starch, trehalose, salicin, cellobiose, melibiose, maltose, sucrose, casein, and several forms of cellulose. The midgut is the major source of all enzymes except invertase and CMC-cellulase, for which hindgut preparations are more active. The pH values of the gut contents are: foregut 6.9, midgut 9.0 and hindgut 7.5. The midgut fluid is pumped forward into the foregut, and the carbohydrases in it are generally more active at the pH of the foregut than at the pH of the midgut. It is possible, therefore, that the foregut, which is itself insignificant as a source of enzymes, is the site of considerable carbohydrate digestion. The proteinase is inhibited 28% by trypsin inhibitors.
Zusammenfassung Eine in vitro-Untersuchung über die Verdauungsvorgänge in den unterirdisch lebenden Larven von S. geminata wies Enzyme nach, die Stärke, Trehalose, Salicin, Cellobiose, Melibiose, Maltose, Rohrzucker, Kasein und mehrere Celluloseformen abbauen können. Versuche mit Vorder-, Mittel- und Enddarmextrakten zeigten, daß der Abbau von Rohrzucker und CMC-Cellulose in Ansätzen mit Enddarmpräparaten prozentual am höchsten war. Maximale Verdauung aller anderen geprüften Substrate erfolgte dagegen mit Mitteldarmpräparaten.Die pH-Werte betrugen im Vorderdarm 6,9, im Mitteldarm 9,0 und im Enddarm 7,5. Im Mittel- und Enddarm lagen die jewciligen optimalen pH-Werte für Maltose bei 6,5–7,0 und 7,5, für Melibiose bei 6,0–7,0 und 7,0–7,5, für Cellobiose bei 6,0–7,0 und 4,5, für Rohrzucker bei 7,0 und 6,0, für CMC bei 5,2 und 5,6.Die Verdauung unlöslicher Celluloseformen war durchweg gering, die der CMC dagegen beträchtlich.Die Mitteldarmflüssigkeit wird nach vorn in den Vorderdarm gepumpt. Die darin enthaltenen Carbohydrasen sind beim pH-Wert des Vorderdarmes meistens wirksamer als in dem des Mitteldarmes. Es ist deshalb möglich, daß im Vorderdarm, welcher selbst wahrscheinlich keine Enzyme produziert, doch eine beträchtliche Kohlenhydratverdauung stattfindet.Die Wirksamkeit der Proteinase wird durch bekannte Tryptin-Hemmstoffe um 28% reduziert.

Trinexapacethyl (TriEt), an acylcyclohexanedionetype inhibitor of gibberellin (GA) biosynthesis, was applied to 3-year-old Eucalyptus globules saplings by localised injection near the base of each stem. The objective was to alter cambial region GA levels and to study the effects on secondary xylem fibre development. Seven weeks later wood samples, with bark and cambial region intact, were removed 10 and 30 cm above the point of injection. Fusiform cambial cell dimensions were compared with those of fibre-tracheids in the most recently formed 100 um of secondary xylem. Increasing TriEt applications from 5 to 5 000 mg active ingredient significantly reduced average fibre length, and to a lesser extent average fusiform cambial cell length. Also reduced was the number of cells in the cambial zone and the number of differentiating fibres with primary walls. However, no trends were evident for changes in fibre diameter, the proportion of vessel elements or the ratio of cambial ray cells to fusiform cambial cells. Two gibberellins (GA1 and GA20), indole-3-acetic acid (IAA) and abscisic acid (ABA) were quantified in cambial region tissues by gas chromatographymass spectrometry using stable isotope labelled internal standards. Increasing TriEt application reduced both GA1 and GA20 levels. Effects on IAA and ABA were not significant, although their levels tended to be lower at the highest TriEt application rate. The elongation of secondary xylem fibres was positively correlated with higher levels of endogenous GA1 (rs= 0.74, P < 0.01) and GA20 (rs= 0.72, P < 0.01). These results support a causal role for GA1 in cambial cell division. They are also consistent with the hypothesis that the elongation of differentiating secondary xylem fibres in woody an–giosperms is dependent on GA1 levels in the cambial region.  相似文献

No-salt flowthrough hydrophobic interaction chromatography (HIC) has been shown to effectively remove process and product-related impurities from bioprocess streams. In this publication, a panel of six antibodies has been used to demonstrate operating principles for the application of no-salt flowthrough HIC in antibody purification processes. The results indicate that no-salt flowthrough HIC provides robust aggregate clearance across operating conditions including flow rate, and variations in resin ligand density. Additionally, HMW reduction has an optimal pH range relative to the isoelectric point of each molecule and high molecular weight (HMW) reduction can be improved by altering the total protein load and/or HMW concentration to drive binding of high molecular weight species to the resin.  相似文献

On three separate occasions we investigated morphological and physiological aspects of bacterial associations with planktonic aggregates of the ubiquitous marine N2 fixing cyanobacterium Trichodesmium sp. Close associations generally characterized Trichodesmium blooms; associations were present during day- and night-time. Colonization by both rod-shaped and filamentous heterotrophic bacteria occurred on Trichodesmiun aggregates actively fixing N2 (acetylene reduction). Scanning electron and optical microscopy showed bacteria located both around and within aggregates. Microautoradiography demonstrated that associated bacteria largely mediated utilization of trace additions of 3H-labeled carbohydrates (fructose, glucose, mannitol) and amino acids, whereas Trichodesmium utilized amino acids only. Oxygen measurements using microelectrodes revealed high localized oxygen consumption among aggregates, with rapid (within a minute) changes from supersaturated to subsaturated oxygen following the transition from photosynthetic illuminated to dark periods. Stab culturing techniques confirmed the presence of heterotrophic N2 fixers among aggregate-associated bacteria. Parallel deployment of oxygen microelectrodes, the tetrazolium salt 2,3,5 triphenyl tetrazolium chloride (TTC) and acetylene reduction assays demonstrated microaerophilic requirements for expression of nitrogenase activity among cultured bacteria. Trichodesmium aggregates are characterized by dynamic nutrient and oxygen regimes, which promote and maintain simultaneous and contiguous oxygenic photosynthesis and N2 fixation. In part, the above-mentioned consortial interactions with a variety of heterotrophic bacteria facilitate Trichodesmium biomass production and bloom formation in nitrogen depleted, oligotrophic tropical/subtropical waters.  相似文献
