Exopolysaccharides (EPSs) are high molecular weight carbohydrate polymers that make up a substantial component of the extracellular polymers surrounding most microbial cells in the marine environment. EPSs constitute a large fraction of the reduced carbon reservoir in the ocean and enhance the survival of marine bacteria by influencing the physicochemical environment around the bacterial cell. Microbial EPSs are abundant in the Antarctic marine environment, for example, in sea ice and ocean particles, where they may assist microbial communities to endure extremes of temperature, salinity, and nutrient availability. The microbial biodiversity of Antarctic ecosystems is relatively unexplored. Deep-sea hydrothermal vent environments are characterized by high pressure, extreme temperature, and heavy metals. The commercial value of microbial EPSs from these habitats has been established recently. Extreme environments offer novel microbial biodiversity that produces varied and promising EPSs. The biotechnological potential of these biopolymers from hydrothermal vent environments as well as from Antarctic marine ecosystems remains largely untapped.  相似文献

Studies were carried out to measure changes in the transmembrane potential of rat alveolar macrophages during exposure of the cells to zymosan particles or to the membrane perturbant, phorbol-12-myristate-13-acetate (PMA), and to determine if changes in membrane potential are related to superoxide anion release. Exposure of the cells to either zymosan or PMA leads to membrane depolarization, which precedes superoxide anion release. Furthermore, the magnitude of the depolarization is dependent upon the concentration of either zymosan or PMA. During exposure of the alveolar macrophages to increasing levels of zymosan, there is an increase in the amount of superoxide released as well as an increase in the magnitude of the depolarization. Incubation of the cells in medium containing 150 mM K+, a medium which causes membrane depolarization, leads to superoxide release from resting cells and a decrease in the amount of superoxide released from cells exposed to zymosan. These results indicate that release of superoxide anion from rat alveolar macrophages is related to membrane depolarization and suggest that the transmembrane potential change may act as a signal to initiate the phagocytotic responses of the cells.  相似文献

 Radiolytic reduction at 77 K of oxo-/hydroxo-bridged dinuclear iron(III) complexes in frozen solutions forms kinetically stabilized, mixed-valent species in high yields that model the mixed-valent sites of non-heme, diiron proteins. The mixed-valent species trapped at 77 K retain ligation geometry similar to the initial diferric clusters. The shapes of the mixed-valent EPR signals depend strongly on the bridging ligands. Spectra of the Fe(II)OFe(III) species reveal an S=1/2 ground state with small g-anisotropy as characterized by the uniaxial component (g z g av /2<0.03) observable at temperatures as high as ∼100 K. In contrast, hydroxo-bridged mixed-valent species are characterized by large g-anisotropy (g z g av /2>0.03) and are observable only below 30 K. Annealing at higher temperatures causes structural relaxation and changes in the EPR characteristics. EPR spectral properties allow the oxo- and hydroxo-bridged, mixed-valent diiron centers to be distinguished from each other and can help characterize the structure of mixed-valent centers in proteins. Vector-borne transmission of Chagas disease has become an urban problem in the city of Arequipa, Peru, yet the debilitating symptoms that can occur in the chronic stage of the disease are rarely seen in hospitals in the city. The lack of obvious clinical disease in Arequipa has led to speculation that the local strain of the etiologic agent, Trypanosoma cruzi, has low chronic pathogenicity. The long asymptomatic period of Chagas disease leads us to an alternative hypothesis for the absence of clinical cases in Arequipa: transmission in the city may be so recent that most infected individuals have yet to progress to late stage disease. Here we describe a new method, epicenter regression, that allows us to infer the spatial and temporal history of disease transmission from a snapshot of a population's infection status. We show that in a community of Arequipa, transmission of T. cruzi by the insect vector Triatoma infestans occurred as a series of focal micro-epidemics, the oldest of which began only around 20 years ago. These micro-epidemics infected nearly 5% of the community before transmission of the parasite was disrupted through insecticide application in 2004. Most extant human infections in our study community arose over a brief period of time immediately prior to vector control. According to our findings, the symptoms of chronic Chagas disease are expected to be absent, even if the strain is pathogenic in the chronic phase of disease, given the long asymptomatic period of the disease and short history of intense transmission. Plasma concentrations of 5alpha-androst-16-en-3-one (5alpha-androstenone), the steroid responsible for boar taint, were measured by radioimmuno-assay in eight sows during the estrous cycle. The steroid concentrations ranged from a low of approximately 500 pg/ml to a high of 2.2 ng/ml during the estrous cycle. Noticeable features in the hormone profile of sows sampled, were the decline in the period preceding estrus, comparatively low levels at estrus and an increase post-estrous. Changes in hormone concentrations over the estrous cycle were significant (P<.05). The timing of the changes in 5alpha-androstenone concentrations during the period preceding estrus among the sows studied was not consistent. The physiological significance of the changes in 5alpha-androstenone concentrations during the period preceding estrus among the sows studied was not consistent. The physiological significance of these changes in 5alpha-androstenone concentration seen during the estrous cycle is not clear. Any bearing these changes may have on the malefemale sexual relationship in pigs needs to be further investigated in view of the known pheromonal function of 5alpha-androstenone.  相似文献

The minimum inhibitory concentration values against Gram-negative and Gram-positive bacteria were determined and compared for a selected group of synthesized alpha-substituted 2-methyl-5-nitrofuran derivatives. In vitro oxidation of thiols to disulfides by 2-(iodomethyl)-5-nitrofuran indicated that oxidation of enzyme-thiol groups to disulfide bonds was a possible mode of action; but was discounted by noninhibition of thiol enzymes by these compounds. Electron-microscopic studies of the morphology of bacteria after treatment with these derivatives showed the formation of unusual elongation, branching and atypical rod shapes in E. coli, while S. aureus manifested multibud formation with some cytoplasmic protrusions. The possible mode of action of these compounds is discussed.  相似文献

The kinetic equations describing transport through a pore that has a binding site and that undergoes a conformational change are identical to those of a carrier model. Therefore, in order to distinguish between the two models it is necessary to test specific predictions based on detailed mechanistic models. A pore model is described in which the substrate (glucose) is able to reach the single binding site only from the outside when the pore is in conformation I and only from the inside when it is conformation II. On the basis of this model it is predicted that solutes which do not have any specific affinity for the binding site should still have a finite permeability via the glucose transport system if they are the same size or smaller than glucose. This permeability should be proportional to the volume of distribution of the solute in the pore and should therefore decrease with increasing molecular size. A geometric pore volume can be estimated from this size dependence. In order to test these predictions, the glucose-dependent permeability of a series of 4-carbon (erythritol), 5-carbon (d-arabitol, l-arabitol and xylitol) and 6-carbon (d-mannitol, d-sorbitol and myo-inositol) polyols was measured. The permeability of all the polyols is decreased by the presence of glucose and the KI of this “inhibitable” component is similar to that of d-sorbose, suggesting that this component is associated with the glucose transport system. Since these observations could be explained entirely in terms of a specific affinity for a carrier binding site, they do not exclude a carrier mechanism. However, as predicted for the pore model, this “inhibitable” permeability decreased with increasing molecular size and the calculated geometric pore volume was of a size that would be expected for a cell membrane pore.  相似文献
