We have evaluated codon usage bias in Drosophila histone genes and have obtained the nucleotide sequence of a 5,161-bp D. hydei histone gene repeat unit. This repeat contains genes for all five histone proteins (H1, H2a, H2b, H3, and H4) and differs from the previously reported one by a second EcoRI site. These D. hydei repeats have been aligned to each other and to the 5.0-kb (i.e., long) and 4.8-kb (i.e., short) histone repeat types from D. melanogaster. In each species, base composition at synonymous sites is similar to the average genomic composition and approaches that in the small intergenic spacers of the histone gene repeats. Accumulation of synonymous changes at synonymous sites after the species diverged is quite high. Both of these features are consistent with the relatively low codon usage bias observed in these genes when compared with other Drosophila genes. Thus, the generalization that abundantly expressed genes in Drosophila have high codon bias and low rates of silent substitution does not hold for the histone genes.   相似文献

We provide an estimate of genetic differentiation within and among 11 populations of Cordia alliodora, an economically important timber tree Cordia alliodora is a widespread species that is distributed throughout Central and South America. Our survey of isozyme variation was conducted on material gathered for international provenance trials over approximately 1,000 km in Central America. Results from provenance trials indicate that there are significant differences between Atlantic and Pacific coast provenances for quantitative characters. Genetic data support some of these findings. Populations of C. alliodora show significant differences in allele frequency at various loci. Significant differences in multilocus allele frequencies occur at 13 of the 55 possible combinations. Eight of these 13 populations are situated on opposite coasts. This physical separation corresponds well with the results of provenance trials that indicate differentiation among the Atlantic and Pacific populations in quantitative morphological traits. We also found a significant negative correlation between levels of heterozygosity and the amount of rainfall, indicating that populations from the drier zone are genetically more heterogenous than populations from the wet zone. Our study indicates that in situ and ex situ conservation should accord high priority to the dry zone populations; furthermore, conservation of this widespread species would require preservation of multiple populations.  相似文献

A multilocus mixed mating model was used to evaluate the mating system of a natural population of Cordia alliodora (Boraginaceae), a neotropical tree. The population was highly outcrossed (tm = 0.966 ± 0.027), in agreement with results from controlled crosses. Departures from the mixed mating model were evident, suggesting some nonrandom, correlated mating. Pollen pool heterogeneity and variation in estimates of individual outcrossing rates indicated that the population may be genetically substructured. Individual outcrossing rates obtained for the samples taken from within different parts of the same tree indicated reduced levels of outcrossing due to limited sampling of the pollen pool. The incompatibility mechanism in C. alliodora, combined with variation in flowering and stand density, appears to lead to both temporal and spatial substructuring of the population.  相似文献

Prostaglandin H synthase (PGHS) activity within intrauterine tissues is considered to catalyze a critical step in prostaglandin (PG) biosynthesis at parturition. In sheep, the placenta is a major site of PG production throughout pregnancy, but little information is available concerning the cells that are responsible. Therefore we determined the distribution of immunoreactive (IR-) PGHS in ovine placental tissue obtained at different times of pregnancy using immunohistochemical techniques. In placentomes from early pregnancy (Days 30-54), IR-PGHS was present in maternal epithelial syncytium, but was not detectable in trophoblast cells. Between Day 54 and Day 100, the number of cells that stained positive for PGHS declined in the maternal epithelial layer in the body of the placenta, but IR-PGHS was present in maternal epithelial cells overlying the vascular cones of the placental hemophagous zone. It was also present in the chorionic fibroblasts, but remained undetectable from all classes of trophoblast cells. IR-PGHS was first detectable in the trophoblastic epithelium by Day 114. Between Day 119 and term the trophoblast mononuclear epithelial cells were intensely immunopositive for PGHS, although immunonegative binucleate cells were present. The maternal epithelium was immunonegative except during the last 7-10 days of pregnancy when PGHS immunostaining appeared in both basal and apical regions of the placenta. Thus, the cellular localization of IR-PGHS changes during ovine pregnancy, from predominantly maternal during the first half of gestation to undetectable and then to predominantly trophoblastic between Day 114 and term, suggesting a gestation-dependent change in sites of PG production during ovine pregnancy. Appearance of IR-PGHS in the trophoblast precedes activation of the fetal hypothalamic-pituitary-adrenal axis, generally considered to provide the trigger to the onset of parturition in sheep, and would therefore appear to be regulated through alternative pathways or mechanisms.  相似文献

With large tracts of once continuous forest now modified by human use to leave reduced and highly fragmented stands of trees, the determination of the genetic consequences of forest fragmentation is a priority for ascertaining the conservation value of resultant stands, and in formulating sustainable management strategies. The levels and distribution of genetic diversity over 10 microsatellite loci were investigated within a fragmented population of the neotropical tree Swietenia humilis Zucc. High levels of genetic variation, typical of a highly outcrossing species, were found in all fragments at all loci (mean HE = 0.548). The majority of the variation was within rather than between fragments (RST = 0.032), giving high indirect estimates of gene flow (Nm = 8.9), probably reflecting the genetic structure of the trees present under more continuous forest. A high proportion of loci also showed significant departures from Hardy-Weinberg equilibrium with associated significant levels of FIS. The initial effects of fragmentation were, however, seen in the fragments through the loss of low-frequency alleles present in the continuous 'control' stand. The percentage of this allelic loss increased with a decrease in fragment size.  相似文献

Background  

Tropical trees undergo severe stress through seasonal drought and flooding, and the ability of these species to respond may be a major factor in their survival in tropical ecosystems, particularly in relation to global climate change. Aquaporins are involved in the regulation of water flow and have been shown to be involved in drought response; they may therefore play a major adaptive role in these species. We describe genetic diversity in the PIP sub-family of the widespread gene family of Aquaporins in five Neotropical tree species covering four botanical families.  相似文献
