Interactions have been studied between juvenile plants of green, brown, and red marine algae and 31 diatom clones isolated from a variety of marine eulittoral habitats. The interactions seemed to be of an individual nature for both juvenile plants and diatoms. Germlings of Ulva lactuca L. mostly showed enhanced growth, often with significant increases in population sizes of the accompanying diatoms. Fucus spiralis L. germlings were mainly unaffected by growth in the presence of the diatom clones, but growth of the diatoms was often stimulated. Ascophyllum nodosum (L.) Le Jol. germlings were little affected, whilst the accompanying diatoms were less noticeably affected than with Fucus spiralis. Germlings of F. vesiculosus L. often showed growth inhibitions in the presence of diatoms, with many diatom populations showing enhanced growth. Similarly, the discoid encrusting sporelings of Chondrus crispus Stackh. showed growth inhibitions where there were measurable interactions, although the accompanying diatoms usually failed to show growth stimulations. The discoid sporelings of Gigartina stellata (Stackh. in With.) Batt. showed high mortalities, usually with marked increases in population sizes of accompanying diatoms.  相似文献

Phytoplankton ecology of the Lake of Menteith,Scotland   总被引:1,自引:1,他引:0  
The results discussed in this paper represent the first seasonal ecological study carried out on the phytoplankton of the Lake of Menteith. All measured nutrients reached maximum levels during the winter, with silicate showing particularly high concentrations (up to 85 µg at Si l–1). During the summer period phosphate, nitrate and silicate showed almost complete exhaustion in surface waters. The lake water was consistently alkaline, never falling below pH 7, while the alkalinity ranged from 20 to 24 mg CaCO3 l–1. Generally, the nutrient status of the main inflow had a rapid effect on the water quality of the lake.The region of the lake under investigation showed no thermal stratification at any period of the year, although continuous thermal gradients were recorded in the winter. The continual circulation of the water mass probably prevented oxygen saturation from falling below 77% even following a large phytoplankton bloom and subsequent decomposition.From an examination of net phytoplankton samples the Lake of Menteith could be described as blue-green or blue-green/diatom in nature. From the quantitative study, large pulses of Melosira, Asterionella and Fragilaria were recorded in the spring. The disappearance of the species appears to be related to silicate limitation. The summer growth of Asterionella may have been promoted by a nitrogen source other than nitrate and nitrite, both of which were reduced to critical levels. This alternative source of combined nitrogen may have been contributed by nitrogen-fixing algae in the lake. Diurnal variation of phytoplankton in Loch Lomond   总被引:2,自引:2,他引:0  
A study of diurnal variation over a 48 hour period was undertaken in July 1973 to ascertain the extent and timing of some major chemical, physical and biological variables in Loch Lomond. The phytoplankton population was dominated by the diatom Tabellaria fenestrata, with a maximum abundance between 04.00 and 06.00 h in surface waters on both days. A distinct diurnal variation in cell numbers was also recorded. Chlorophyll a values also showed a regular pattern of variation with a single peak between 10.00 and 14.00 h each day. Some chemical changes appeared to be a direct consequence of phytoplankton multiplication. Nitrate-nitrogen showed a decrease in concentration coinciding with the period of cell multiplication, whereas dissolved silica concentrations only fell on the completion of this process. Other common diatoms displayed less distinct patterns of variation although Fragilaria crotonensis attained its maximum abundance in surface waters. Considerable variation was recorded in the number of organisms and chlorophyll a levels at 25 cm intervals in the upper metre of the water column, with large variations in standing-crop and chemical parameters in the space of one hour. Diurnal oscillations in the position of the thermocline were recorded, with the thermal discontinuity being at its greatest depth in the early hours of the morning. The hypolimnion and thermocline regions clearly acted as a source of nutrient supply to the epilimnion. Background

Bam32, a 32 kDa adaptor molecule, plays important role in B cell receptor signalling, T cell receptor signalling and antibody affinity maturation in germinal centres. Since antibodies against trypanosome variant surface glycoproteins (VSG) are critically important for control of parasitemia, we hypothesized that Bam32 deficient (Bam32-/-) mice would be susceptible to T. congolense infection.

Methodology/Principal Findings

We found that T. congolense-infected Bam32-/- mice successfully control the first wave of parasitemia but then fail to control subsequent waves and ultimately succumb to their infection unlike wild type (WT) C57BL6 mice which are relatively resistant. Although infected Bam32-/- mice had significantly higher hepatomegaly and splenomegaly, their serum AST and ALT levels were not different, suggesting that increased liver pathology may not be responsible for the increased susceptibility of Bam32-/- mice to T. congolense. Using direct ex vivo flow cytometry and ELISA, we show that CD4+ T cells from infected Bam32-/- mice produced significantly increased amounts of disease-exacerbating proinflammatory cytokines (including IFN-γ, TNF-α and IL-6). However, the percentages of regulatory T cells and IL-10-producing CD4+ cells were similar in infected WT and Bam32-/- mice. While serum levels of parasite-specific IgM antibodies were normal, the levels of parasite-specific IgG, (particularly IgG1 and IgG2a) were significantly lower in Bam32-/- mice throughout infection. This was associated with impaired germinal centre response in Bam32-/- mice despite increased numbers of T follicular helper (Tfh) cells. Adoptive transfer studies indicate that intrinsic B cell defect was responsible for the enhanced susceptibility of Bam32-/- mice to T. congolense infection.

Conclusions/Significance

Collectively, our data show that Bam32 is important for optimal anti-trypanosome IgG antibody response and suppression of disease-promoting proinflammatory cytokines and its deficiency leads to inability to control T. congolense infection in mice.  相似文献

IGF-I stimulates cell growth through interaction of the IGF receptor with multiprotein signaling complexes. However, the mechanisms of IGF-I receptor-mediated signaling are not completely understood. We have previously shown that IGF-I-stimulated 3T3-L1 cell proliferation is dependent on Src activation of the ERK-1/2 MAPK pathway. We hypothesized that IGF-I activation of the MAPK pathway is mediated through integrin activation of Src-containing signaling complexes. The disintegrin echistatin decreased IGF-I phosphorylation of Src and MAPK, and blocking antibodies to (alpha)v and beta3 integrin subunits inhibited IGF-I activation of MAPK, suggesting that (alpha)v(beta)3 integrins mediate IGF-I mitogenic signaling. IGF-I increased ligand binding to (alpha)v(beta)3 as detected by immunofluorescent staining of ligand-induced binding site antibody and stimulated phosphorylation of the beta3 subunit, consistent with inside-out activation of (alpha)v(beta)3 integrins. IGF-I increased tyrosine phosphorylation of the focal adhesion kinase (FAK) Pyk2 (calcium-dependent proline-rich tyrosine kinase-2) to a much greater extent than FAK, and increased association of Src with Pyk2 but not FAK. The intracellular calcium chelator BAPTA prevented IGF-I phosphorylation of Pyk2, Src, and MAPK, suggesting that IGF-I activation of Pyk2 is calcium dependent. Transient transfection with a dominant-negative Pyk2, which lacks the autophosphorylation and Src binding site, decreased IGF-I activation of MAPK, but no inhibition was seen with transfected wild-type Pyk2. These results indicate that IGF-I signaling to MAPK is dependent on inside-out activation of (alpha)v(beta)3 integrins and integrin-facilitated multiprotein complex formation involving Pyk2 activation and association with Src.  相似文献

A seasonal and ecological study of the phytoplankton of Loch Lomond   总被引:5,自引:4,他引:1  
The seasonal succession of phytoplankton in Loch Lomond has been studied both quantitatively and from observations on net plankton. From a qualitative investigation of net phytoplankton Loch Lomond may be described as diatom-desmid in nature although from the quantitative studies the general domination of the population by diatoms becomes clear. During the vernal phytoplankton increase diatoms contributed more than 90% of the population throughout the loch. The bulk of the phytoplankton production occurred in the southern region, where the standing crop was considerably greater throughout the year than in other regions of the loch.Patterns of seasonal succession are described for all species which make an important contribution to the productivity of the loch and the observed successions related to measured physical and chemical changes in the water. Explanations are put forward to account for the absence of a second (autumnal) pulse ofMelosira in Loch Lomond. The appearance ofAnabaena circinalis is a possible indication of the changing trophic status of the southern region of the loch.  相似文献

In autoimmune diseases, the accumulation of activated leukocytes correlates with inflammation and disease progression, and, therefore, the disruption of leukocyte trafficking is an active area of research. The serine/threonine protein kinase Tpl2 (MAP3K8) regulates leukocyte inflammatory responses and is also being investigated for therapeutic inhibition during autoimmunity. Here we addressed the contribution of Tpl2 to the regulation of macrophage chemokine receptor expression and migration in vivo using a mouse model of Tpl2 ablation. LPS stimulation of bone marrow-derived macrophages induced early CCR1 chemokine receptor expression but repressed CCR2 and CCR5 expression. Notably, early induction of CCR1 expression by LPS was dependent upon a signaling pathway involving Tpl2, PI3K, and ERK. On the contrary, Tpl2 was required to maintain the basal expression of CCR2 and CCR5 as well as to stabilize CCR5 mRNA expression. Consistent with impairments in chemokine receptor expression, tpl2−/− macrophages were defective in trafficking to the peritoneal cavity following thioglycollate-induced inflammation. Overall, this study demonstrates a Tpl2-dependent mechanism for macrophage expression of select chemokine receptors and provides further insight into how Tpl2 inhibition may be used therapeutically to disrupt inflammatory networks in vivo.  相似文献
