The kinetics of the 130 kDa monomer obtained by treatment of duck salt gland Na,K-ATPase with C12E8 was compared with that of the membrane-bound enzyme. The shapes of the substrate-velocity curves for the membrane-bound and solubilized forms were quite different: a hyperbolic one for the monomeric Na,K-ATPase and a nonhyperbolic one for the native enzyme. A reaction scheme for ATP hydrolysis based on a comparative analysis of kinetic properties of these two forms is proposed. Experimental evidence in favour of this hypothesis is presented. 相似文献

Gangliosides are significant participants in suppression of immune system during tumor processes. It was shown that they can
induce apoptosis of T-lymphocytes in a raft-dependent manner. Fluorescence confocal microscopy was used to study distribution
and influence of ganglioside GM1 on raft properties in giant unilamellar vesicles. Both raft and non-raft phase markers were
utilized. No visible phase separation was observed without GM1 unless lateral tension was applied to the membrane. At 2 mol
% of GM1 large domains appeared indicating macroscopic phase separation. Increase of GM1 content to 5 mol % resulted in shape
transformation of the domains consistent with growth of line tension at the domain boundary. At 10 mol % of GM1 almost all
domains were pinched out from vesicles, forming their own homogeneous liposomes. Estimations showed that the change of the
GM1 content from 2 to 5–10 mol % resulted in a several-fold increase of line tension. This finding provides a possible mechanism
of apoptosis induction by GM1. Incorporation of GM1 into a membrane leads to an increase of the line tension. This results
in a growth of the average size of rafts due to coalescence or merger of small domains. Thus, necessary proteins can find
themselves in one common raft and start the corresponding cascade of reactions.

We have found that neutrophils begin to express NMDA receptors on their membranes after in vivo activation. These receptors are the target for action of homocysteine (HC). After incubation of activated neutrophils with

We have studied the influence of hydrogenation on the relative stability of the low-lying isomers of the anionic B7− cluster, computationally. It is known that the pure-boron B7− cluster has a doubly (σ- and π-) aromatic C6v (3A1) quasi-planar wheel-type triplet global minimum (structure 1), a low-lying σ-aromatic and π-antiaromatic quasi-planar singlet C2v (1A1) isomer 2 (0.7 kcal mol−1 above the global minimum), and a planar doubly (σ- and π-) antiaromatic C2v (1A1) isomer 3 (7.8 kcal mol−1 above the global minimum). However, upon hydrogenation, an inversion in the stability of the species occurs. The planar B7H2− (C2v, 1A1) isomer 4, originated from the addition of two hydrogen atoms to the doubly antiaromatic B7− isomer 3, becomes the global minimum structure. The second most stable B7H2− isomer 5, originated from the quasi-planar triplet wheel isomer 1 of B7−, was found to be 27 kcal mol−1 higher in energy. The inversion in stability occurs due to the loss of the doubly aromatic character in the wheel-type global minimum isomer (C6v, 3A1) of B7− upon H2−addition. In contrast, the planar isomer of B7− (C2v, 1A1) gains aromatic character upon addition of two hydrogen atoms, which makes it more stable.

The results of research of camosine as an antioxidative system corrector in conditions of oxidative stress caused by the action of damaging factors (y-rays, overcooling, hypobaric hypoxia, brain ischemia, neurotoxin impact) are summarized in the present review. The effects of carnosine are characterized not only at the level of the whole organism but also in "in vitro" models with use of a whole series of enzymatic systems. The results of the experiments conducted displayed the ability of carnosine to protect animals from oxidative stress based on the combination of direct antioxidative effects and a modulation of enzymes' activities which participate in controlling of reactive oxygen species level in tissues. 相似文献

For a series of 1,10-phenantroline tris-beta-diketonate europium complexes (EuC), cytotoxic activity on the HBL-100 human breast carcinoma cells was determined. Liposomal preparation of the most active EuC, V12, was also tested for cytotoxicity.
