The Firmicutes represent a major component of the intestinal microflora. The intestinal Firmicutes are a large, diverse group of organisms, many of which are poorly characterized due to their anaerobic growth requirements. Although most Firmicutes are Gram positive, members of the class Negativicutes, including the genus Veillonella, stain Gram negative. Veillonella are among the most abundant organisms of the oral and intestinal microflora of animals and humans, in spite of being strict anaerobes. In this work, the genomes of 24 Negativicutes, including eight Veillonella spp., are compared to 20 other Firmicutes genomes; a further 101 prokaryotic genomes were included, covering 26 phyla. Thus a total of 145 prokaryotic genomes were analyzed by various methods to investigate the apparent conflict of the Veillonella Gram stain and their taxonomic position within the Firmicutes. Comparison of the genome sequences confirms that the Negativicutes are distantly related to Clostridium spp., based on 16S rRNA, complete genomic DNA sequences, and a consensus tree based on conserved proteins. The genus Veillonella is relatively homogeneous: inter-genus pair-wise comparison identifies at least 1,350 shared proteins, although less than half of these are found in any given Clostridium genome. Only 27 proteins are found conserved in all analyzed prokaryote genomes. Veillonella has distinct metabolic properties, and significant similarities to genomes of Proteobacteria are not detected, with the exception of a shared LPS biosynthesis pathway. The clade within the class Negativicutes to which the genus Veillonella belongs exhibits unique properties, most of which are in common with Gram-positives and some with Gram negatives. They are only distantly related to Clostridia, but are even less closely related to Gram-negative species. Though the Negativicutes stain Gram-negative and possess two membranes, the genome and proteome analysis presented here confirm their place within the (mainly) Gram positive phylum of the Firmicutes. Further studies are required to unveil the evolutionary history of the Veillonella and other Negativicutes.  相似文献

Agelasine and agelasimine derivatives with substantially less complicated terpenoid side chains compared to the naturally occurring compounds have been synthesized and their ability to inhibit growth of microorganisms and cancer cells has been studied. Compounds with excellent activity against cancer cell lines (MIC ca. 1 microM for the most potent compounds), including a drug resistant renal cell line, have been identified. Most compounds studied also exhibited broad spectrum antimicrobial activity including activity against Mycobacterium tuberculosis.  相似文献

Do all natural compounds have a distinct biological activity, or are most of them merely biosynthetic debris? Many natural compounds have important biological functions, and certainly many more of the ample 200,000 currently known will ultimately prove to be more than just 'secondary metabolites'. The question is how to select the most promising candidates for potential new drugs. 'Rediscovery' of known natural compounds is regarded as a nuisance or disappointment by scientists involved with the identification of novel compounds. The other side of the coin, however, is that the discovery that a particular compound occurs in unrelated species can be a valuable clue toward the identification of a novel receptor or enzyme. Here, we put forward the hypothesis that when a natural compound occurs in unrelated species, it must have an important biological function by interacting with a specific molecular target. This is because it is extremely improbable that in nature one particular compound is synthesized in totally unrelated species for no reason at all. For many compounds occurring in unrelated species, it is already known that they act on specific molecular targets. For others, it is just known that they occur in different species. In some cases, biological activities are known but not the underlying mechanisms of action. It is from this category of compounds that important discoveries are likely to be made. Some (around 70) of them were identified. They represent important clues from nature offering an alternative approach to the classical screening of large numbers of compounds.  相似文献

β‐Glucosidases (BGs) from Aspergillus fumigates, Aspergillus niger, Aspergillus oryzae, Chaetomium globosum, Emericella nidulans, Magnaporthe grisea, Neurospora crassa, and Penicillium brasilianum were purified to homogeneity, and analyzed by isothermal titration calorimetry with respect to their hydrolytic activity and its sensitivity to glucose (product) using cellobiose as substrate. Global non‐linear regression of several reactions, with or without added glucose, to a product inhibition equation enabled the concurrent derivation of the kinetic parameters kcat, Km, and the apparent product inhibition constant appKi for each of the enzymes. A more simple fit is not advisable to use as the determined appKi are in the same range as their Km for some of the tested BGs and produced glucose would in these cases interfere. The highest value for kcat was determined for A. fumigatus (768 s?1) and the lowest was a factor 9 less. Km varied by a factor of 3 with the lowest value determined for C. globosum (0.95 mM). The measured appKi varied a factor of 15; the hydrolytic activity of N. crassa being the most resistant to glucose with an apparent product inhibition constant of 10.1 mM. Determination of appKi using cellobiose as substrate is important as it reflects to what extent the different BGs are hydrolytically active under industrial conditions where natural substrates are hydrolyzed and the final glucose concentrations are high. The oldest early Mesolithic settlements found so far (i.e. 8600 b.p.) in the interior of northern Sweden, in the province of Norrbotten, have been discovered through the application of a model simulating glacio-isostatic land uplift. The objective of this study was to investigate, through pollen and charred particle analyses, vegetation composition and the influence of man on vegetation and fire pattern in the vicinity of two of these early settlements. Early Holocene vegetation was characterised by Betula, Hippophaë and Salix-species, but no initial impact by hunter-gatherers on vegetation was detected. Subsequently Betula and Pinus became dominant but abrupt changes in the tree layer followed, Poaceae, Hippophaë and Salix-species increased, and Humulus occurred. These changes, which were synchronous with dates of archaeological findings at the settlements, indicated local vegetation changes caused by man. The relationship between these changes and fire was, however, subtle. Eventually Pinus forests became dominant and human impact on vegetation became less apparent. It is concluded that the impact of early Mesolithic hunter-gatherers on vegetation was detectable, but that their effect on fire pattern was difficult to evaluate. Further, we show that the glacio-isostatic land uplift must be considered in the search for Mesolithic settlements in areas previously covered by the Weichselian Ice, and that the process has influenced the biological archives through re-deposition of material.  相似文献

Barettin (cyclo [(6-bromo-8-en-tryptophan) arginine]), a diketopiperazine isolated from the marine sponge Geodia barretti, is a potent inhibitor of barnacle larvae settlement with an EC50-value of 0.9 microM. In the present study, 14 analogs of barettin and its structural congener dipodazine were synthezised and tested for their ability to inhibit larval settlement. Two of the analogs have an intact barettin skeleton. The remaining analogs have a dipodazine skeleton (a diketopiperazine where arginine is replaced with glycine). Six of the tested synthetic analogs displayed significant settlement inhibition with the most potent inhibitor being benzo[g]dipodazine, which displayed even stronger activity than barettin (EC50-value 0.034 microM). The effect of benzo[g]dipodazine was also shown to be readily reversible, when cyprids were transferred to filtered seawater (FSW).  相似文献

Competition during the juvenile phase is a key process for regulating density in organisms with high fecundity. Juvenile density-dependent bottlenecks may become even more pronounced if several cohorts compete, but this has received relatively limited attention in previous literature. We performed a manipulation experiment in seven coastal streams to investigate the presence of inter-cohort competition, using habitat selection, body-size and density of newly emerged (age-0) brown trout (Salmo trutta) as response variables. The trout population (age ≥ 1 fish) was estimated using electro-fishing prior to the emergence of fry (April-May) and was either removed (manipulated sections) or maintained (control sections). Age-0 habitat selection was examined in June while density and body-size was evaluated in October (end of the growth season). We found that age-0 trout selected habitats that were located further from riffles (nursery habitats) in the absence of age ≥ 1 trout, suggesting a niche overlap between cohorts in the habitat dimension and, hence, that both inter-cohort competitive interactions and ontogenetic preference may influence habitat utilisation in the wild. Furthermore, we also found age-0 body-size to be significantly larger in manipulated sections and negatively related to its own density. We argue that competition from older cohorts influence the availability of age-0 feeding territories at the critical phase of emergence with secondary negative effects on age-0 growth. These results not only have implications for understanding the mechanisms of density dependence but can also provide valuable knowledge to the management of salmonid populations and their habitats in the wild.  相似文献
