39K nuclear magnetic resonance was used to measure the efflux of K+ from suspensions of human erythrocytes [red blood cells (RBCs)], that occurred in response to the calcium ionophore, A23187 and calcium ions; the latter activate the Gárdos channel. Signals from the intra- and extracellular populations of 39K+ were selected on the basis of their longitudinal relaxation times, T 1, by using an inversion- recovery pulse sequence with the mixing time, τ1, chosen to null one or other of the signals. Changes in RBC volume consequent upon efflux of the ions also changed the T 1 values so a new theory was implemented to obviate a potential artefact in the data analysis. The velocity of the K+ efflux mediated by the Gárdos channel was 1.19±0.40 mmol (L RBC)−1 min−1 at 37°C.  相似文献

The prayer book The Hours of the Blessed Virgin Mary, known as The Farnese Hours, was illustrated by the Croatian miniaturist, Julije Klović, between 1539 and 1546 in Rome. In this masterpiece of Renaissance miniature art, Klović drew in colour a bird of paradise. In 2001, the pictured bird was determined to be a Raggiana Bird of Paradise, Paradisaea raggiana P. L. Sclater 1873. Our later determination shows that it is actually a Greater Bird of Paradise Paradisaea apoda (Linnaeus 1758) and the first drawing of this species in colour in Europe. Klović’s model for the Greater Bird of Paradise was one of the first trade skins of birds of paradise that, from 1522, had been arriving in Europe. Revision of the early drawings of the birds of paradise species in Europe, relevant to the Klović drawing model, has also been carried out and is discussed.  相似文献

Adult tissue homeostasis and repair relies on prompt and appropriate intervention by tissue-specific adult stem cells (SCs). SCs have the ability to self-renew; upon appropriate stimulation, they proliferate and give rise to specialized cells. An array of environmental signals is important for maintenance of the SC pool and SC survival, behavior, and fate. Within this special microenvironment, commonly known as the stem cell niche (SCN), SC behavior and fate are regulated by soluble molecules and direct molecular contacts via adhesion molecules providing connections to local supporting cells and the extracellular matrix. Besides the extensively discussed array of soluble molecules, the expression of adhesion molecules and molecular contacts is another fundamental mechanism regulating niche occupancy and SC mobilization upon activation. Some adhesion molecules are differentially expressed and have tissue-specific consequences, likely reflecting the structural differences in niche composition and design, especially the presence or absence of a stromal counterpart. However, the distribution and identity of intercellular molecular contacts for adhesion and adhesion-mediated signaling within stromal and non-stromal SCN have not been thoroughly studied. This review highlights common details or significant differences in cell-to-cell contacts within representative stromal and non-stromal niches that could unveil new standpoints for stem cell biology and therapy.  相似文献

Previous studies from our laboratory and others presented evidence that oxidized 1-palmitoyl-2-arachidonyl-sn-glycero-3-phosphatidylcholine (OxPAPC) and oxidized 1-palmitoyl-2-arachidonyl-sn-glycero-3-phosphatidylethanolamine can inhibit lipopolysaccharide (LPS)-mediated induction of interleukin-8 (IL-8) in endothelial cells. Using synthetic derivatives of phosphatidylethanolamine, we now demonstrate that phospholipid oxidation products containing alpha,beta-unsaturated carboxylic acids are the most active inhibitors we examined. 5-Keto-6-octendioic acid ester of 2-phosphatidylcholine (KOdiA-PC) was 500-fold more inhibitory than OxPAPC, being active in the nanomolar range. Our studies in human aortic endothelial cells identify one important mechanism of the inhibitory response as involving the activation of neutral sphingomyelinase. There is evidence that Toll-like receptor-4 and other members of the LPS receptor complex must be colocalized to the caveolar/lipid raft region of the cell, where sphingomyelin is enriched, for effective LPS signaling. Previous work from our laboratory suggested that OxPAPC could disrupt this caveolar fraction. These studies present evidence that OxPAPC activates sphingomyelinase, increasing the levels of 16:0, 22:0, and 24:0 ceramide and that the neutral sphingomyelinase inhibitor GW4869 reduces the inhibitory effect of OxPAPC and KOdiA-PC. We also show that cell-permeant C6 ceramide, like OxPAPC, causes the inhibition of LPS-induced IL-8 synthesis and alters caveolin distribution similar to OxPAPC. Together, these data identify a new pathway by which oxidized phospholipids inhibit LPS action involving the activation of neutral sphingomyelinase, resulting in a change in caveolin distribution. Furthermore, we identify specific oxidized phospholipids responsible for this inhibition.  相似文献

Meat is an important source of animal protein but, at the same time, it includes saturated fatty acids, which makes it a potential cause of different cardiovascular diseases and still little is known about influence of age and sex on these parameters in roe deer muscles. The aim of this study was to determine the effect of age and sex on the CLA and other fatty acids content in the musculus longissimus lumborum in 67 roe deer (Capreolus capreolus L.). In the meat from the oldest individuals a higher content of CLA was noted (89.76 [mg/kg]) when compared to the fawns (42 [mg/kg]). In this research meat from roe deer does showed in general a higher percentage proportion of SFAs and MUFAs, but lower of PUFAs, than the meat from bucks. These results may provide an important source of information for consumers of roe deer meat because of differences between CLA and other fatty acids content depending on age and sex of the animals. Meat from roebucks is the most advantageous for dietary purposes.  相似文献

The functional state of the neurovascular unit (NVU), composed of the blood–brain barrier and the perivasculature that forms a dynamic interface between the blood and the central nervous system (CNS), plays a central role in the control of brain homeostasis and is strongly affected by CNS drugs. Human primary brain microvascular endothelium, astrocyte, pericyte, and neural cell cultures are often used to study NVU barrier functions as well as drug transport and efficacy; however, the proteomic and metabolomic responses of these different cell types are not well characterized. Culturing each cell type separately, using deep coverage proteomic analysis and characterization of the secreted metabolome, as well as measurements of mitochondrial activity, the responses of these cells under baseline conditions and when exposed to the NVU‐impairing stimulant methamphetamine (Meth) are analyzed. These studies define the previously unknown metabolic and proteomic profiles of human brain pericytes and lead to improved characterization of the phenotype of each of the NVU cell types as well as cell‐specific metabolic and proteomic responses to Meth.  相似文献

The asymmetric distribution of many components of the Xenopus oocyte, including RNA, proteins, and pigment, provides a framework for cellular specialization during development. During maturation, Xenopus oocytes also acquire metals needed for development, but apart from zinc, little is known about their distribution. Synchrotron X-ray fluorescence microprobe was used to map iron, copper, and zinc and the metalloid selenium in a whole oocyte. Iron, zinc, and copper were asymmetrically distributed in the cytoplasm, while selenium and copper were more abundant in the nucleus. A zone of high copper and zinc was seen in the animal pole cytoplasm. Iron was also concentrated in the animal pole but did not colocalize with zinc, copper, or pigment accumulations. This asymmetry of metal deposition may be important for normal development. Synchrotron X-ray fluorescence microprobe will be a useful tool to examine how metals accumulate and redistribute during fertilization and embryonic development.  相似文献
