目的探讨慢性胃炎胃粘膜肠化生CD44、CD44V6及cyclin D1、Cyclin E表达的意义。方法利用免疫细胞化学技术对39例伴有肠化生的慢性胃炎和5例正常人胃窦粘膜的活检组织进行检查。结果正常人胃窦粘膜上皮,腺上皮对CD44、CD44V6、Cyclin D1和Cyclin E均为阴性,但在有神经内分泌样细胞的粘膜,CD44V6和cyclin D1为阳性。慢性胃炎肠化生区和不典型增生区除CD44为阴性外,CD44V6、cyclin D1和cyclin E均呈现不同的阳性反应,但未见有阳性的神经内分泌样细胞。间质细胞大都呈阳性反应。结论CD44V6、cyclin D1和cyclin E可能是胃癌前状态的早期事件,而CD44可能为胃癌晚期的标志物。  相似文献

As biennial recretohalophytes, Limonium bicolor plants need 2 years to complete their life cycle. A growth habit mutant Vernalization Requirement Loss 15 (vrl15) was obtained by ion implantation. However, the biological characteristics of the mutant were unclear. In the current study, the related traits of vrl15 and some possible reasons for these traits were examined. Compared with wild type (WT), vrl15 can bolt and flower in approximately four months without vernalization. Moreover, vrl15 needed much less time to bolting and flowering than wild-type L. bicolor under different vernalization treatments. After 20 days’ vernalization, bolting vrl15 plants had 24 rosette leaves and bolting WT had 31 rosette leaves. Moreover, the pollen number per anther, the proportion of active pollen, the seed setting rate and the 1000 seed weight of vrl15 were all lower than those of WT. The soluble sugar content and soluble protein content in leaves of the vrl15 were much higher than those of WT sowed at the same time. In addition, the GA content in the leaves of bolting vrl15 was higher than that of the non-bolting WT sowed at the same time and non-bolting vrl15, whereas the contents of ABA and BR were much lower than that of the non-bolting WT. These results indicate that to some extent the increase of GA and decrease of ABA and BR content may be involved in the growth habit and male fertility alteration of mutant vrl15 of L. bicolor.

An electronic DNAzyme sensor for highly sensitive detection of Pb(2+) is demonstrated by coupling the significant signal enhancement of the layer-by-layer (LBL) assembled quantum dots (QDs) with Pb(2+) specific DNAzymes. The presence of Pb(2+) cleaves the DNAzymes and releases the biotin-modified fragments, which further hybridize with the complementary strands immobilized on the gold substrate. The streptavidin-coated, QD LBL assembled nanocomposites were captured on the gold substrate through biotin-streptavidin interactions. Subsequent electrochemical signals of the captured QDs upon acid dissolution provide quantitative information on the concentrations of Pb(2+) with a dynamic range from 1 to 1000 nM. Due to the dramatic signal amplification by the numerous QDs, subnanomolar level (0.6 nM) of Pb(2+) can be detected. The proposed sensor also shows good selectivity against other divalent metal ions and thus holds great potential for the construction of general DNAzyme-based sensing platform for the monitoring of other heavy metal ions.  相似文献

Limonium bicolor, a typical recretohalophyte, has a specialized salt-secreting structure in the epidermis called the salt gland and plays a significant role in improving saline land. Understanding the molecular mechanisms of salt secretion and salt gland development requires an efficient L. bicolor transformation system, which is described in this report. Leaf explants were incubated with Agrobacterium tumefaciens strain EHA105 harboring the plasmid pTCK303 containing the β-glucuronidase gene (GUS) as the transgene reporter and the hygromycin B resistance gene as a selectable marker. Up to 96.9% of leaves were induced to regenerate shoots on an Murashige and Skoog (MS) medium supplemented with 4.4 μM 6-benzyladenine and 1.1 μM α-naphthaleneacetic acid; roots were induced on the MS medium containing 2.5 μM indole-3-butyric acid. This tissue culture system was suitable for Agrobacterium-mediated transformation of L. bicolor. Pre-cultivated explants (2 d old) were incubated with Agrobacterium (0.6–0.7 at OD600) in a shaking culture for 20 min; the explants and bacterium were co-cultivated for 4 d in the dark before the explants were transferred to a selection medium containing 8 mg/L hygromycin B and 600 mg/L piperacillin sodium (added to prevent continued Agrobacterium growth). Histochemical assays and PCR to detect the GUS gene showed that transformation frequency was 4.43%. Quantitative PCR and Northern blotting further verified the integration and presence of the GUS gene in L. bicolor. This is the first report of an Agrobacterium-based transformation system for L. bicolor. The system will facilitate a research on the identity and function of genes involved in salt gland development and salt secretion.  相似文献

To improve the fermentation yield of xylanase by optimizing the fermentation conditions for strain Xw2, a Plackett-Burman design was used to evaluate the effects of eight variables on xylanase production by strain Xw2. The steepest ascent (descent) method was used to approach the optimal response surface experimental area. The optimal fermentation conditions were obtained by central composite design and response surface analysis. The results showed that the composition of the optimal fermentation medium was corn cob + 1.5% wheat bran (1:1), 0.04% MnSO4, 0.04% K2HPO4. 3H2O, and an inoculum size of 6% in 50 mL liquid volume (pH = 6.0). The optimal culture conditions were 28oc at 150 r/min for 54.23 h. The results of this study can serve as the basis for the industrial production and application of xylanase.  相似文献

A 2-mo experiment with the white Sprague-Dawley (SD) rats was conducted to investigate the effect of the water extracts of black tea (BTWE) and green tea (GTWE) and the black tea leaves (BTF) and the green tea leaves (GTF) on the metabolism of mineral elements. One hundred eight 12-mo-old white SD rats were randomly divided into 13 groups; 6 of these drank the BTWE or GTWE in which the water extracts concentrations of black tea or green tea were, respectively, 0.6%, 1.2%, and 2.4%, and 6 of these had black tea leaves (BTF) and green tea leaves (GTF) added in which the contents of BTF or GTF were, respectively, 0.5%, 1.0%, and 2.0%, one of these was control. The teas and their water extracts could promote the absorption of manganese and copper. In GTF, BTF, GTWE, and BTWE, the apparent absorption rates of manganese were significantly increased. The manganese contents in the tibia were also elevated, and the differences between GTWE and GTF were significant. The apparent absorption rates of copper and the copper contents in the tibia were increased, but not significantly. The teas and their water extracts inhibited the absorption of calcium (p > 0.05) and iron (p < 0.05). The cerebrum calcium contents were significantly decreased, but the contents of calcium and iron in tibia were not significantly changed. Compared with the control, although the apparent absorption rates of aluminum in all experimental groups were not observed to be different, the aluminum contents in the tibia (p > 0.05) and cerebrum (p < 0.05) were increased. GTF and GTWE decreased the apparent absorption rates of zinc, but BTF and BTWE increased them; the zinc contents in tibia were a little improved, whereas its contents in the cerebrum were gradually decreased with the increase of tea leaves dose and tea concentration.  相似文献
