Incubation of first trimester placental explants with [35S]O4 resulted in incorporation of radioactive sulfate into free and dimer forms of alpha subunit of human chorionic gonadotropin. Sulfate was not attached to N-linked oligosaccharides since it was not released by endoglycosidase F. Analysis of pronase digest revealed the presence of tyrosine-O-[35S]O4. Comparison of tryptic peptides of alpha subunit labeled with several amino acids identified the penultimate carboxyterminal peptide as the sulfation site. Since the C-terminal region of the hCG alpha plays a critical role in receptor binding of the hormone, modification in this region may regulate hormonal activity.  相似文献

The intracellular events that occur in T lymphoid cells after activation or after infection with HIV-1 are not well defined. In the case of HIV-1 infection, it is unknown whether the tat-I gene, an essential gene for viral replication, affects host cell nuclear factors. Using two-dimensional PAGE, we have identified a novel nuclear protein, designated nuclear protein-28,000 (NP-28), which is induced in Jurkat T cells by stimulation with PMA and/or PHA or ionomycin. This nuclear protein has an apparent molecular mass of 28,000 Da and an isoelectric point of 4.6. Interestingly, Jurkat cells transfected with tat-I express higher levels of NP-28 constitutively, without added stimulation. Incubation of Jurkat cells expressing tat-I with PMA and/or PHA or ionomycin causes superinduction of NP-28. We have therefore identified a novel lymphoid nuclear protein induced by T cell activation that occurs in tat-I expressing cells in the absence of activating agents.  相似文献

基于MODIS-EVI的西南地区植被覆盖时空变化及驱动因素研究   总被引:6,自引:0,他引:6  
基于MODIS-EVI和气象数据,利用最大值合成法、像元二分模型、趋势分析和相关分析等方法,探讨了西南地区2001-2015年植被覆盖时空变化特征及其对气候因子的响应,并分析了温度和降水对植被覆盖时空变化的驱动作用。结果表明:(1)2001-2015年,西南地区植被EVI以0.1%/a的变化率呈波动增加趋势,但空间异质性显著,呈现出从东南向西北逐渐递减的趋势;(2)西南地区以高和极高植被覆盖度为主,极低植被覆盖度区域约占研究区总面积的8.6%,植被覆盖度增加的区域集中分布在广西省北海-钦州、贵州省邵通-毕节-遵义、四川省广元-广安以及西藏那曲等地区,植被覆盖度呈减少趋势区域主要集中在西藏拉萨-阿里地区和四川成都-阿坝州-甘孜州等地区;(3)植被EVI与同期温度和降水相关性较好,均以正相关为主。在0.05显著水平下,受降水驱动的区域呈斑块状分布在西藏自治区和青海省交界处,以及云南和广西部分地区,约占研究区总面积的3.4%;受温度驱动的区域零星分布在各省、自治区,约占研究区总面积的1.6%;受温度和降水共同驱动的区域约占研究区总面积的7.2%,主要分布在西藏自治区的阿里地区北部,青海省的三江源地区以及四川和贵州两省交界处的小部分地区;西南地区大部分区域的植被EVI指数变化表现为非气候因素驱动。  相似文献

ABSTRACT

Dolphins possess a highly sophisticated auditory system and a keen capability for echolocation. Signals are emitted in the form of high intensity, short duration, broadband exponentially decaying pulses. The frequency spectra of echolocation signals used by many dolphins are dependent on the output intensity of the signals and not on any fine tuning by the animals. When the output intensity is low, the center frequency of the click tends to be low. As the output intensity increases, the center frequency also tends to increase. The pulses propagate from the dolphin's melon in a relatively narrow beam, and echoes are received via the lower jaw, with a slightly wider beam. Echo- locating dolphins can detect targets at ranges of approximately 100 plus meters, depending on the size of the targets. Target discrimination experiments have shown that dolphins can discriminate the shape, size, material composition and internal structure of targets from the echoes. The broadband short duration properties of the signal allow the echoes to have high temporal resolution, so that within the structure of the echoes a considerable amount of information on the properties of the target can be conveyed. A brief comparison between the bat and dolphin sonar system will also be made. Bats typically emit much longer signals and a wider variety of different types of signals than dolphins. Signals used by some bats are suited to detecting Doppler shift, whereas the dolphin signal is designed to be tolerant of Doppler effects.  相似文献

为保护香港特有植物香港鹅耳枥,基于典型样地调查,对香港鹅耳枥所在群落进行了研究。结果表明,在225 m2的样地内,有维管植物37科66属68种。建群种为香港鹅耳枥,个体高度最高3 m,所在群落植株平均高度0.92 m,分层不明显。位于中国香港岛的香港鹅耳枥处于衰退状态,小个体极少。因此,建议采取就地保护和迁地保护措施,以重建种群。  相似文献

The murine visceral endoderm is an extraembryonic cell layer that appears prior to gastrulation and performs critical functions during embryogenesis. The traditional role ascribed to the visceral endoderm entails nutrient uptake and transport. Besides synthesizing a number of specialized proteins that facilitate uptake, digestion, and secretion of nutrients, the extraembryonic visceral endoderm coordinates blood cell differentiation and vessel formation in the adjoining mesoderm, thereby facilitating efficient exchange of nutrients and gases between the mother and embryo. Recent studies suggest that in addition to this nutrient exchange function the visceral endoderm overlying the egg cylinder stage embryo plays an active role in guiding early development. Cells in the anterior visceral endoderm function as an early organizer. Prior to formation of the primitive streak, these cells express specific gene products that specify the fate of underlying embryonic tissues. In this review we highlight recent investigations demonstrating this dual role for visceral endoderm as a provider of both nutrients and developmental cues for the early embryo.  相似文献

Two forms of the gonadotropin alpha subunit are synthesized in placenta and in human chorionic gonadotropin (hCG)-producing tumors: an uncombined (monomer) form and a combined (dimer) form. These forms show differences in their migration on sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The slower migration of the monomeric form on sodium dodecyl sulfate-polyacrylamide gel electrophoresis has been attributed to a different glycosylation pattern. Previous studies demonstrated different roles of each of the two alpha N-linked glycosylation sites (Asn-52 and Asn-78) in secretion of the uncombined subunit and the biologic activity of hCG dimer. To assess the influence of formation of dimer on the processing pattern at the individual sites, we characterized the N-linked oligosaccharides of monomer and dimer forms of recombinant human choriogonadotropin alpha subunit. Two approaches were employed. First, site-directed mutagenesis was used to alter the two N-linked oligosaccharide attachment sites, thus allowing the expression of alpha subunits containing only one glycosylation site. Second, tryptic glycopeptides of the wild-type subunits were examined. Concanavalin A (ConA) binding and sialic acid content indicated that the oligosaccharides at each glycosylation site of the uncombined alpha subunit are processed differently. Oligosaccharides present at Asn-52 are almost exclusively ConA-unbound and contain three sialic acid residues. The majority of Asn-78-linked oligosaccharides are ConA-bound and disialylated. Both sites are processed independently because no significant differences were observed between the oligosaccharides at the same sites in wild-type and mutant monomeric alpha subunits. By contrast, the majority of the oligosaccharides at both glycosylation sites of the dimer alpha are bound to ConA. Thus, combination primarily affects the processing pattern of the Asn-52-linked species. Because glycosylation at this site is essential for hCG assembly and signal transduction, these data imply a critical link between the site-specific processing and hormone function.  相似文献

Pulmonary surfactants enhance adenovirus-mediated gene transfer but inhibit cationic liposome-mediated transfection in lung epithelial cells in vitro. This study examines the effect of the synthetic lung surfactant Exosurf on dendrimer-mediated transfection in eukaryotic cells. Exosurf significantly enhanced dendrimer-luciferase plasmid transfection in a number of cell lines and was very effective in primary cells. Luciferase expression increased up to 40-fold in primary normal human bronchial/tracheal epithelial cells (NHBE). FACScan analysis demonstrated that the transfection rate of the human T cell leukemia Jurkat cell line has significantly improved from 10 to 90% of cells at 24 h after transfection. Analysis of the components of Exosurf revealed that the nonionic surfactant tyloxapol was responsible for the enhancement of dendrimer-mediated gene transfer. The tyloxapol effect was due to increased cell membrane porosity and DNA uptake. Our results demonstrate that Exosurf and its component, tyloxapol, constitute a powerful enhancer for dendrimer-mediated gene transfer in vitro.  相似文献
