To investigate the mechanism of regulation of sarco-endoplasmic reticulum Ca(2+)-ATPase (SERCA) by phospholamban (PLB), we expressed Cerulean-SERCA and yellow fluorescent protein (YFP)-PLB in adult rabbit ventricular myocytes using adenovirus vectors. SERCA and PLB were localized in the sarcoplasmic reticulum and were mobile over multiple sarcomeres on a timescale of tens of seconds. We also observed robust fluorescence resonance energy transfer (FRET) from Cerulean-SERCA to YFP-PLB. Electrical pacing of cardiac myocytes elicited cytoplasmic Ca(2+) elevations, but these increases in Ca(2+) produced only modest changes in SERCA-PLB FRET. The data suggest that the regulatory complex is not disrupted by elevations of cytosolic calcium during cardiac contraction (systole). This conclusion was also supported by parallel experiments in heterologous cells, which showed that FRET was reduced but not abolished by calcium. Thapsigargin also elicited a small decrease in PLB-SERCA binding affinity. We propose that PLB is not displaced from SERCA by high calcium during systole, and relief of functional inhibition does not require dissociation of the regulatory complex. The observed modest reduction in the affinity of the PLB-SERCA complex with Ca(2+) or thapsigargin suggests that the binding interface is altered by SERCA conformational changes. Ditching is a common practice to dewater wetlands, including peatlands, and ditch blocking is a common method for restoring wetlands because substrate is often unavailable for filling the ditches. However, filling has many advantages compared to blocking ditches. Our goal was to test whether ditches could be filled in a Colorado sloping fen (Chattanooga Fen) using bales created from shredded aspen (Populus tremuloides) tree‐fiber. We monitored water table levels before and after we filled two ditches (combined length of approximately 165 m × 3 m wide) as well as an undisturbed reference portion of Chattanooga fen. The reference site had stable water tables that rarely dropped more than 20 cm below the soil surface. The ditches had been dewatering large areas of the fen for at least 100 years. Filling the ditches with fiber bales resulted in a water table increase between 2 and 22 cm in an area up to 150 m below the ditch. Native sedges now cover the area where we filled the ditches, with no erosion or compression/settling of the ground observed and no water backing up behind the filled ditches. Filling the ditches with shredded fiber bales is a good option for restoration in wilderness areas, or areas lacking peat or mineral soil fill because it is a natural material that is easily transported and placed in the ditches. 相似文献
This discussion focuses on discontinuities in both natural and technological systems caused by the introduction of exotic species into areas which they would not have been able to reach without human assistance or other alterations to native communities. The case histories of both the Asian clam and the zebra mussel are particularly instructive because of their recent introductions and the dramatic impact they have had on both natural and technological systems. Control of these mollusc species in technological systems (e.g. water intake systems) is effective to a degree but requires constant attention and utilization of resources. If control is neglected there could be extremely serious consequences. At present, no inexpensive means exist for controlling their effects, and further geographical distribution in North America is highly probable. The mollusc case histories do not appear to be linear and, in some instances, may not be continuous. Some importations of exotic species were deliberate (such as the gypsy moth, Porthetria dispar), but their escape into natural systems has often been accidental or, a less charitable person might say, careless. A viable strategy for coping with discontinuities is elusive. Discontinuities will, undoubtedly, have a major effect on the possibility of sustainable use of the planet or, to use the currently popular term, sustainable development. 相似文献
Cortical granules, which are specialized secretory organelles found in ova of many organisms, have been isolated from the eggs of the sea urchins Arbacia punctulata and Strongylocentrtus pupuratus by a simple, rapid procedure. Electron micropscope examination of cortical granules prepared by this procedure reveals that they are tightly attached to large segments of the plasma membrane and its associated vitelline layer. Further evidence that he cortical granules were associated with these cell surface layers was obtained by (125)I-labeling techniques. The cortical granule preparations were found to be rich in proteoesterase, which was purified 32-fold over that detected in a crude homogenate. Similarly, the specific radioactivity of a (125)I-labeled, surface glycoprotein was increased 40-fold. These facts, coupled with electron microscope observations, indicate the isolation procedure yields a preparation in which both the cortical granules and the plasma membrane-vitelline layer are purified to the same extent. Gel electrophoresis of the membrane-associated cortical granule preparation reveals the presence of at least eight polypeptides. The major polypeptide, which is a glycotprotein of apparent mol wt of 100,000, contains most of the radioactivity introduced by (125)I-labeling of the intact eggs. Lysis of the cortical granules is observed under hypotonic conditions, or under isotonic conditions if Ca(2+) ion is present. When lysis is under isotonic conditions is induced by addition of Ca(2+) ion, the electron-dense contents of the granules remain insoluble. In contrast, hypotonic lysis results in release of the contents of the granule in a soluble form. However, in both cases the (125)I-labeled glycoprotein remains insoluble, presumably because it is a component of either the plasma membrane or the vitelline layer. All these findings indicate that, using this purified preparation, it should be possible to carry out in vitro studies to better define some of the initial, surface-related events observed in vivo upon fertilization. 相似文献
Experiments have been done to confirm the previously reported effect of indoleacetic acid (IAA) on the rate of CO2 assimilation in bean leaves. It was shown that spraying the leaves of a variety of plants caused an increase in the rate of CO2 assimilation from 30% to 100% during the half-hour to 1 hour period following spraying. The only plant tested which did not show such an effect was corn.
The breaking of dormancy of axial buds in the bean plant was correlated with an increase in the rate of CO2 assimilation in adjacent leaves for a brief period of time. It has been shown that IAA solution sprayed on 1 leaflet of a leaf can cause an increase in the rate of CO2 assimilation in the other leaflets, and that IAA applied to the cut stem of a leaflet or a developing bud can be transported to adjacent leaves and cause an increase in the CO2 assimilation rate. The reaction caused by IAA is very similar to that caused by the breaking of dormancy of a bud. This indicates that the bud break response in CO2 assimilation in leaves is caused by auxin synthesized in a bud as it begins to grow, and exported into adjacent leaves.
The death of osteocytes, the terminally differentiated cells of the osteoblast lineage that are embedded in bone and regulate remodeling, is significant to both normal and pathological bone resorption. Apoptotic osteocytes putatively release a clarion signal that enhances the development of the bone-resorbing osteoclasts and targets their migration to the breach in the osteocyte network. This phenomenon is thought to underlie normal repair of bone microdamage and contribute to the etiologies of inflammatory bone loss. The chromatin protein high mobility group box 1 protein (HMGB1) has been identified as an \"alarmin\" in other tissues. An alarmin is an endogenous molecule released by dead and dying cells that alert the innate immune system to damage and the need for tissue repair. Wang and colleagues presented evidence in a landmark 1999 study showing that released HMGB1 is a lethal mediator of sepsis. Extracellular HMGB1 is a ligand for the toll-like receptors (TLRs) and for the receptor for advanced glycation end products (RAGE) all of which amplify inflammation. Recent studies by our lab and others have shown that HMGB1 is a bone-active cytokine. It is released by apoptotic osteoblasts in vitro, including the MLO-Y4 osteocyte-like cells. Extracellular HMGB1 enhances the expression of RANKL, TNFalpha, and IL6 in osteoblastogenic bone marrow stromal cell cultures, and it is chemotactic to osteoclasts. In this prospectus we will review HMGB1 activity at the immune-bone interface and propose a role for HMGB1 as an osteocyte alarmin and mediator of normal remodeling and inflammatory bone loss. 相似文献
