The fatty acid composition of the total and polar lipid fractions of Choanephora cucurbitarum grown under different cultural conditions were analyzed by thin-layer and gas-liquid chromatography. It was observed that temperature, age, pH, and light influenced the degree of unsaturation, this being due mainly to changes in the gamma-linolenic acid concentration. The conditions used in this study did not alter the qualitative profile of fatty acids normally present in the organism. Neither did these conditions stimulate the production of further long-chain fatty acids (C20-C26) beyond gamma-linolenic acid (C18:3) as reported earlier using growth media containing glutamic acid. The fatty acid pattern of lipid fractions though the same qualitatively, differed quantitatively. The polar lipid fractions, phosphatidyl choline, phosphatidyl ethanolamine, and diphosphatidyl glycerol showed an appreciable variation in gamma-linolenic acid content under different cultural conditions. The degree of unsaturation of the various lipid fractions decreased with increases in temperature, light intensity, and pH, but within each treatment the same pattern of decreasing degree of unsaturation with increasing age was observed. Life cycle impact of emissions, energy requirements, and exergetic losses are calculated for a novel process for producing titanium dioxide nanoparticles from an ilmenite feedstock. The Altairnano hydrochloride process analyzed is tailored for the production of nanoscale particles, unlike established commercial processes. The life cycle energy requirements for the production of these particles is compared with that of traditional building materials on a per unit mass basis. The environmental impact assessment and energy analysis results both emphasize the use of nonrenewable fossil fuels in the upstream life cycle. Exergy analysis shows fuel losses to be secondary to material losses, particularly in the mining of ilmenite ore. These analyses are based on the same inventory data. The main contributions of this work are to provide life cycle inventory of a nanomanufacturing process and reveal potential insights from exergy analysis that are not available from other methods.  相似文献

Abstract: Obtaining reliable results from life-cycle assessment studies is often quite difficult because life-cycle inventory (LCI) data are usually erroneous, incomplete, and even physically meaningless. The real data must satisfy the laws of thermodynamics, so the quality of LCI data may be enhanced by adjusting them to satisfy these laws. This is not a new idea, but a formal thermodynamically sound and statistically rigorous approach for accomplishing this task is not yet available. This article proposes such an approach based on methods for data rectification developed in process systems engineering. This approach exploits redundancy in the available data and models and solves a constrained optimization problem to remove random errors and estimate some missing values. The quality of the results and presence of gross errors are determined by statistical tests on the constraints and measurements. The accuracy of the rectified data is strongly dependent on the accuracy and completeness of the available models, which should capture information such as the life-cycle network, stream compositions, and reactions. Such models are often not provided in LCI databases, so the proposed approach tackles many new challenges that are not encountered in process data rectification. An iterative approach is developed that relies on increasingly detailed information about the life-cycle processes from the user. A comprehensive application of the method to the chlor-alkali inventory being compiled by the National Renewable Energy Laboratory demonstrates the benefits and challenges of this approach.  相似文献

Synopsis The cerebellum of healthy and malnourished squirrel monkeys was studied histopathologically and cytochemically for a number of important enzymes such as phosphatases (acid and alkaline phosphatase, ATPase, thiamine pyrophosphatase), esterases (simple esterase and acetylcholinesterase), dehydrogenases (succinate, malate and isocitrate dehydrogenase, lactate dehydrogenase, 6-phosphogluconate dehydrogenase and glucose-6-phosphate dehydrogenase), monoamine oxidase and phosphorylase. The Purkinje cells, stellate and basket cells were found to be more sensitive to protein malnutrition compared to the other types of cells in the cerebellum. An increase in the number of dark cells with large amounts of ribonucleoprotein complex in the Purkinje cell layer of the extremely malnourished animals sacrificed after 15 weeks on a low protein diet may be significant and may reflect either an abnormal metabolic process or an interruption in the axonal transport of RNA complex. This may also be directly related to a significant reduction in the level of oxidative enzymes, especially those of the tricarboxylic acid cycle, these being the main source of energy stored in ATP. At the same time the level of lysosomal enzymes, which are responsible for the catalysis of the different degradation reactions, is greatly increased and indicates cellular catabolism. The present investigations point to the probability that the neurons adapt to the changed environment by beginning to utilize structural proteins for their basic metabolism.  相似文献

Summary Squirrel monkeys were kept in 3 groups of 2 each maintained on one of the following light schedules: 12-12 hour light/dark, continuous light, and continuous dark. The animals (one from each group) were sacrificed after a period of 81/2 and 111/2 months and histochemical techniques for some oxidative and hydrolytic enzymes were employed in cryostat section of the brains of these animals. Lactic dehydrogenase (LDH), an enzyme involved in glycolytic metabolism, showed enhanced enzyme activity compared to the other two groups in a number of areas (e.g., corpus geniculatum laterale, nucleus n. oculomotorii and trochlearis, nucleus interpeduncularis, locus coeruleus, reticular formation, cerebellar cortex, etc.). LDH showed more pronounced enzyme changes in the neuropil, whereas the neurons showed somewhat more enhanced activity in the succinic dehydrogenase preparations. Butyrylcholinesterase and simple esterase, however, did not show any difference in enzyme reaction in the three groups under study. Similarly, certain areas (e.g., colliculus superior, griseum pontis, central gray, vestibular and cochlear nuclei, choroid plexus and ependyma) did not show any enzyme difference in all the eight enzyme preparations. The epiphysis showed a decrease in monoamine oxidase activity in the constant light-exposed animals compared to the other two groups. The significance of these findings is discussed.  相似文献

The detailed distribution of lactate dehydrogenase and monoamine oxide has been described at various levels through the pons and mesencephalon of the brain of the squirrel monkey. A comparison of the two enzymes has given an interesting picture of their selective localization in the different nuclei. Particular attention has been paid to select sections at comparative levels. Marked monoamine oxidase activity has been observed in the nucleus interpeduncularis, the nucleus centralis superior, the nucleus annularis, and in the neuropil and in the cytoplasm of neurons of some other nuclei. Particularly strong lactate dehydrogenase activity has, however, been observed in the corpus geniculatum laterale and mediale, the nucleus pulvinaris, the superior and inferior colliculi, the substantia grisea centralis and some of the nuclei of cranial nerves.  相似文献

Coronary artery disease (CAD) affects Indians 5–6 years earlier than in the west, is diffuse and malignant, and poses a heavy burden on India’s developing economy. Traditional risk factors have failed to explain this high incidence of premature CAD and hence this study investigated the association of two novel risk biomarkers, cystatin C and small dense LDL (sdLDL) with the presence and severity of CAD. Cystatin C and sdLDL were estimated in 204 CAD patients ≤45 years of age and compared with 161 age-matched healthy controls. The traditional lipid profile parameters, i.e., cholesterol, LDL, HDL, triglycerides, apolipoproteins A1 and B, and Lp(a) were also measured in both groups. Cystatin C was significantly raised and mean LDL particle size significantly reduced in CAD patients as compared to controls. 62.7 % of CAD patients showed pattern B while 37.3 % patients showed pattern A. Of the traditional lipid tests, only HDL and apolipoprotein A1 showed a significant decrease in the CAD group. sdLDL was significantly associated with the severity of CAD, while cystatin C was not. Both cystatin C and sdLDL emerged as independent risk factors, however, of the two, sdLDL was a more sensitive predictor of CAD events. Cystatin C and mean LDL particle size are significantly and independently associated with the presence of CAD events in patients ≤45 years with normal kidney function. Hence, these novel risk biomarkers can be useful tools in reducing the morbidity and mortality associated with CAD in the productive Indian workforce.  相似文献

The fatty acid composition of the total, neutral, sterol, free fatty acid, and polar-lipid fractions in the mycelium of Choanephora curcurbitarum was determined. The major fatty acids in all lipid fractions were palmitic, oleic, linoleic, and gamma-linolenic acid. Different lipid fractions did not show any particular preference for any individual fatty acid; however, the degree of unsaturation was different in different lipid fractions. Free fatty acid and polar lipid fractions contained a higher proportion of gamma-linolenic acid than did triglyceride and sterol fractions. Addition of glutamic acid to the malt-yeast extract and medium resulted in the biosynthesis of a number of long-chain fatty acids beyond the gamma-linolenic acid. These fatty acids, e.g., C22:1, C24:0, and C26:0, were never observed to be present in the fungus when grown on a malt-yeast extract medium without glutamic acid. Furthermore, thin-layer chromatographic analysis showed a larger and denser spot of diphosphatidyl glycerol from the mycelium grown on glutamic acid medium than from the control mycelium. The possible significance of this finding is discussed.  相似文献
