Context: Nickel oxide (NiO) nanoparticles (NPs) with appropriate surface chemistry have been widely used for their potential new applications in biomedical industry. Increased usage of these NPs enhances the chance of exposure of personnel involved in the work place.

Objective: This study was designed to assess the ability of NiO NPs to cause biochemical alterations post-acute oral exposure in female Wistar rats.

Materials and methods: Rats were administered with 125, 250, and 500?mg/kg doses of NiO NPs for haematological, biochemical, and histopathological studies. Biodistribution patterns of NiO NPs in female Wistar rats were also monitored.

Results: NiO NPs caused significant (p?Conclusions: This study revealed that exposure to nanosized NiO particles at acute doses may cause adverse changes in animal biochemical profiles. Further, the in vivo studies on toxicity evaluation help in biomonitoring of the potential contaminants.  相似文献

Both the epidermal growth factor receptor (EGFR) and the insulin-like growth factor receptor (IGFR) have been implicated in the tumorigenesis of a variety of cancers. Here we propose that simultaneous targeting of both receptors with a bispecific antibody would lead to enhanced antitumor activity. To this end, we produced a recombinant human IgG-like bispecific antibody, a Di-diabody, using the variable regions from two antagonistic antibodies: IMC-11F8 to EGFR and IMC-A12 to IGFR. The Di-diabody binds to both EGFR and IGFR and effectively blocked both EGF- and IGF-stimulated receptor activation and tumor cell proliferation. The Di-diabody also inherited the biological properties from both of its parent antibodies; it triggers rapid and significant IGFR internalization and degradation and mediates effective antibody-dependent cellular cytotoxicity in a variety of tumor cells. Finally, the Di-diabody strongly inhibited the growth of two different human tumor xenografts in vivo. Our results underscore the benefits of simultaneous targeting of two tumor targets with bispecific antibodies.  相似文献

A series of tungstate double perovskite Ca3WO6 doped with Tb3+ was prepared by a combustion process using urea as a flux. The crystal structure identification of Ca3WO6:Tb3+ phosphors was done using X-ray diffraction patterns, and a monoclinic structure was discovered. The Fourier transform infrared spectrum of Ca3WO6:Tb3+ displayed characteristic vibrations of tungstate bonds. Under 278 nm excitation, Ca3WO6:Tb3+ exhibited intense downconversion green emission, which corresponded to the 5D47FJ (J = 4,5) transitions of Tb3+. The phosphor exhibited the highest photoluminescence (PL) intensity when it was doped with 1 mol% of Tb3+; later intensity quenching appeared to be due to the multipolar interaction at higher dopant concentrations. Moreover, high-quality thermoluminescence (TL) was detected when phosphors were irradiated using beta rays. The effects of Tb3+ concentration and beta dose on TL intensity were the two major aspects studied in detail. The TL intensity demonstrated excellent linear response to the applied range of beta dose. The trap parameters of the studied phosphors were computed by the peak shape approach and glow curve deconvolution. The fading effect on TL intensity was studied by recording the TL glow curves after 1 month of beta irradiation. Obtained results from the PL and TL characterizations showed that the phosphors under study have the potential to be used in lighting displays and in thermoluminescence dosimetry.  相似文献

Objective: This study was designed to determine the genotoxic effect of exposure to a mixture of pesticides in 106 female agricultural workers employed in cotton fields from India.

Methods: Comet, micronucleus and chromosomal aberrations tests were carried out in peripheral blood lymphocytes. Micronucleus test was also performed in buccal epithelial cells. Levels of antioxidant enzymes, RBC acetylcholinesterase and hematological parameters were analyzed in the blood samples of the study subjects.

Results: The results indicated significant DNA damage, increased frequency of micronuclei and chromosomal aberrations in the exposed subjects (p?<?0.05). The levels of antioxidant enzymes were significantly lowered and the rate of lipid peroxidation was elevated in the exposed subjects.

Conclusion: The outcome of the study revealed an increased risk of genotoxicity and health implications in female agricultural workers.  相似文献

Platelet-derived growth factor (PDGF) and its receptors (PDGFR) play important roles in tumorigenesis through stimulating tumor growth and promoting angiogenesis via enhancing pericyte recruitment and vessel maturation. Here we produced a neutralizing antibody, 1B3, directed against mouse PDGFRbeta. 1B3 binds to PDGFRbeta with high affinity (9x10(-11)M) and blocks PDGF-BB from binding to the receptor with an IC(50) of approximately 1.2 nM. The antibody also blocks ligand-stimulated activation of PDGFRbeta and downstream signaling molecules, including Akt and MAPK p42/44, in tumor cells. In animal studies, 1B3 significantly enhanced the antitumor and the anti-angiogenic activities of DC101, an antibody directed against mouse vascular endothelial growth factor receptor 2, in a pancreatic (BxPC-3) and a non-small cell lung (NCI-H460) tumor xenograft models. Treatment with the combination of 1B3 and DC101 in BxPC-3 xenograft-bearing mice resulted in tumor regression in 58% of mice compared to that in 18% of mice treated with DC101 alone. Taken together, these results lend great support to use PDGFRbeta antagonists in combinations with other antitumor and/or anti-angiogenic agents in the treatment of a variety of cancers.  相似文献

A luminescent double perovskite phosphor Sr2YNbO6 doped with Eu3+ crystallized to the monoclinic phase and was synthesized successfully via a conventional high-temperature combustion method. The formation of the crystal structure, phase purity, and surface morphology were studied using X-ray diffraction patterns and scanning electron microscopy. The characteristic vibrations between the atoms of the functional groups present in phosphor were studied using Fourier transform infrared spectra analysis. The luminescence properties of the prepared phosphors were investigated in terms of photoluminescence (PL) and thermoluminescence (TL). PL excitation spectra exhibited charge transfer bands and the characteristic 4f6 transitions of Eu3+. A prominent PL emission was obtained for the phosphor doped with 4 mol% Eu3+ under the 396 nm excitation wavelength. PL emission quenching was observed for the higher doping concentrations due to a multipole–multipole interaction. A highly intense PL emission arose due to the hypersensitive 5D07F2 electric dipole transition of Eu3+ that dominated the emission spectra. The thermal stability of the phosphor was examined through temperature-dependent PL. The TL properties of the Sr2YNbO6 double perovskites irradiated with a 90Sr beta source at different doses were measured. The double perovskite phosphors under study showed a linear dose–response with increasing beta dose, ranging from 1 Gy to 10 Gy. Trapping parameters of the TL glow curves were determined using Chen's peak shape method and computerized glow curve deconvolution (CGCD). CGCD fitting of the TL glow curves revealed that it was consisted of three major peaks and followed second-order kinetics. The estimated activation energies were determined using different methods and were comparable and significant.  相似文献
