Data on immuno- and biochemical identification, genetic control and phylogenesis of new allotype Lpm13 of the Lpm system in domestic mink are presented. This allotype is encountered in mink populations with the frequency 0.9 and higher. The availability of Lpm13 genetic marker permitted another haplotype to be revealed, in addition to the eight known Lpm haplotypes by means of genetic analysis. It was established that, alongside with the earlier described haplotype Lpm3,4,6,8,9,10,11 (abbreviation H3), there exists a similar haplotype, Lpm3,4,6,8,9,10,11,13 (abbreviation H3.13), containing the Lpm13 gene. Of the rest seven haplotypes, five have the Lpm13 gene and two do not. Taking into account this gene and corresponding antigenic marker, the differentiation of 28, instead of 25, phenotypes and 45, instead of 36, genotypes for the Lpm system became possible. Lpm13 antigenic specificity was found with no exception in all individual serum samples taken from ten species and interspecific hybrids of Mustelidae which are closely related to domestic mink. The data obtained give grounds to refer the newly identified Lpm13 gene to the first evolutionary conservative category of genes of the multigenic Lpm system which is also represented by the Lpm6, Lpm9, Lpm10 and Lpm11 genes. The hypotheses of instantaneous formation of polymorphism of the Lpm system in domestic mink are briefly regarded.  相似文献

By means of immunohistochemical methods a study of the liver intralobular localization of cytochromes P-450b(+e) and P-450c(+d) has been carried out after separate and consecutive treatment of rats with phenobarbital (PB) and 3-methylcholanthrene (MC). PB-treatment leads to localization of P-450b in the centrilobular region, whereas homogeneous distribution of P-450c in the lobule is observed after MC-treatment. The consecutive treatment with PB and MC is accompanied by the localization of P-450c only in cells of the periportal region of the lobule. PB-treatment of rats after preliminary MC-injection also results in the periportal localization of P-450c, and a small quantity of P-450b is localized in hepatocytes of the centrilobular region. Hence, the consecutive treatment with inducers of different molecular forms of cytochrome P-450 is accompanied by the redistribution of these isoenzymes in parenchymatous cells of the liver lobule. That is confirmed also by biochemical testing and immunochemical analysis of the microsomal fraction of hepatocytes.  相似文献

Using immunochemical and, subsequently, autoradiographic methods, the authors have shown that after the injection of the total fraction of polyribosomes isolated from the rat liver in the Acetabularia cytoplasm a protein interacting with the antiserum against rat thyrosine aminotransferase is synthesized. Prospects of using Acetabularia as a test-system for the translation of heterogenous mRNA and mRNP are discussed.  相似文献

Three new allotypes were discovered in mink sera by means of isoimmunization. Based on the results of identifications, they were referred to the Lpm system of serum alpha2-lipoprotein. They were designated as Lpm 6, Lpm 7 and Lpm 8. The allotyping of serum samples from 342 minks made possible to establish a relation between Lpm 7 and Lpm 8 using the chi2 method; it was also found that these two allotypes related to each of the first five Lpm as well as to their phenotypes, which was described earlier. There was a significant dependence of the expression of Lpm 6 on Lpm 5 indicating a genetic relation between Lpm 6 and other allotypes. The detection of Lpm 6, 7 and 8, which are the most likely under the control of the same gene (or a sustem of genes) as Lpm 1, 2, 3 and 5, is an evidence for the complex structure of the Lpm locus.  相似文献

The location of major quantitative trait loci (QTL) contributing to stem and leaf [Na+] and [K+] was previously reported in chromosome 7 using two connected populations of recombinant inbred lines (RILs) of tomato. HKT1;1 and HKT1;2, two tomato Na+‐selective class I‐HKT transporters, were found to be closely linked, where the maximum logarithm of odds (LOD) score for these QTLs located. When a chromosome 7 linkage map based on 278 single‐nucleotide polymorphisms (SNPs) was used, the maximum LOD score position was only 35 kb from HKT1;1 and HKT1;2. Their expression patterns and phenotypic effects were further investigated in two near‐isogenic lines (NILs): 157‐14 (double homozygote for the cheesmaniae alleles) and 157‐17 (double homozygote for the lycopersicum alleles). The expression pattern for the HKT1;1 and HKT1;2 alleles was complex, possibly because of differences in their promoter sequences. High salinity had very little effect on root dry and fresh weight and consequently on the plant dry weight of NIL 157‐14 in comparison with 157‐17. A significant difference between NILs was also found for [K+] and the [Na+]/[K+] ratio in leaf and stem but not for [Na+] arising a disagreement with the corresponding RIL population. Their association with leaf [Na+] and salt tolerance in tomato is also discussed.  相似文献
