Previous work has shown that agonistic Abs to CD40 (anti-CD40) can boost weak CD8 T cell responses as well as substitute for CD4 T cell function during chronic gammaherpes virus infection. Agonistic anti-CD40 treatment has, therefore, been suggested as a potential therapeutic strategy in immunocompromised patients. In this study, we investigated whether agonistic anti-CD40 could substitute for CD4 T cell help in generating a sustained CD8 T cell response and prevent viral recrudescence following infection with lymphocytic choriomeningitis virus (LCMV). Contrary to expectations, we found that anti-CD40 treatment of MHC class II-deficient mice infected with a moderate dose of LCMV resulted in severe suppression of the antiviral CD8 T cell response and uncontrolled virus spread, rather than improved CD8 T cell immune surveillance. In Ab-treated wild-type mice, the antiviral CD8 T cell response also collapsed prematurely, and virus clearance was delayed. Additional analysis revealed that, following anti-CD40 treatment, the virus-specific CD8 T cells initially proliferated normally, but an increased cell loss compared with that in untreated mice was observed. The anti-CD40-induced abortion of virus-specific CD8 T cells during LCMV infection was IL-12 independent, but depended partly on Fas expression. Notably, similar anti-CD40 treatment of vesicular stomatitis virus-infected mice resulted in an improved antiviral CD8 T cell response, demonstrating that the effect of anti-CD40 treatment varies with the virus infection studied. For this reason, we recommend further evaluation of the safety of this regimen before being applied to human patients. 相似文献

In 1931 a red-colored, sandy marker horizon was placed on the emergingSkallingen salt marsh. Sedimentation on top of the marker horizon sincethen shows two opposing tendencies. Coincident with salt marshdevelopment the sediments display up to 1964 a fining upward sequencewith an increasing content of organic matter. Since 1964 a nearby creekhas meandered towards the sampling plot. Consequently, the sedimentsbecome coarser with a decreasing organic matter content. Themorphological induced changes in sedimentary conditions strongly influencemetal content in the sediments and thereby hide anthropogenic inducedconcentration variations. Thus, an apparently diminishing Zn content (perkg dry weight) since 1964 could indicate lesser load to the area. However,corrected for grain size effects there is an increasing content of Zn. Othermetal concentrations (e.g. Cu) show a diminishing trend when corrected forgrain size effects and therefore indicate a reduced anthropogenic inducedload of these metals to the salt marsh. 相似文献

Interactions between cognition and emotion are important for survival, often occurring in the absence of awareness. These interactions have been proposed to involve competition between cognition and emotion for attentional resources. Emotional stimuli have been reported to impair performance on cognitive tasks of low, but not high, load if stimuli are consciously perceived. This study explored whether this load-dependent interference effect occurred in response to subliminal emotional stimuli. Masked emotional (appetitive and aversive), but not neutral, stimuli interfered with performance accuracy but not response time on a cognitive task (n-back) at low (1-back), but not high (2-back) load. These results show that a load-dependent interference effect applies to masked emotional stimuli and that the effect generalises across stimulus categories with high motivational value. This supports models of selective attention that propose that cognition and emotion compete for attentional resources. More specifically, interference from masked emotional stimuli at low load suggests that attention is biased towards salient stimuli, while dissipation of interference under high load involves top-down regulation of attention. Our data also indicate that top-down goal-directed regulation of attention occurs in the absence of awareness and does not require metacognitive monitoring or evaluation of bias over behaviour, i.e., some degree of self-regulation occurs at a non-conscious level. 相似文献

Bone marrow chimeras were used to determine the cellular target(s) for the antiviral activity of gamma interferon (IFN-gamma). By transfusing such mice with high numbers of naive virus-specific CD8(+) T cells, a system was created in which the majority of virus-specific CD8(+) T cells would be capable of responding to IFN-gamma, but expression of the relevant receptor on non-T cells could be experimentally controlled. Only when the IFN-gamma receptor is absent on both radioresistant parenchymal and bone marrow-derived cells will chimeric mice challenged with a highly invasive, noncytolytic virus completely lack the ability to control the infection and develop severe wasting disease. Further, the study shows that IFN-gamma receptor expression on parenchymal cells in the viscera is more important for virus control than IFN-gamma receptor expression on bone marrow-derived cells. 相似文献

Summary The Argyrotopos profile of the Oligocene Ayii Pantes Formation in the NW Greece was studied using litho- and biofacies-analysis.
The exposed sequence is mainly composed of monotonous marls and sandy marls. A rich fossil content, e.g. calcareous nannoplankton,
is recorded. Some sandstone beds and two graded limestone beds with abundant lepidocyclinids and globigerinids are intercalated.
The nannofossil assemblages from the measured profile of the Ayii Pantes Formation are attributed to theSphenolithus distentus Zone (NP 24) which is equivalent to theCyclicargolithus floridanus Subzone (CP 19a) of the Chattian, Upper Oligocene. The deposition took place in an euphotic subtidal environment situated
on the eastern margin of the Apulian Platform. The lepidocyclinids were derived from this platform and were transported basinwards.
The formation of the marls is influenced by sediment flux from the west. 相似文献

Objective
To assess the effects of one intra-articular corticosteroid injection two weeks prior to an exercise-based intervention program for reducing pain sensitivity in patients with knee osteoarthritis (OA).Design
Randomized, masked, parallel, placebo-controlled trial involving 100 participants with clinical and radiographic knee OA that were randomized to one intra-articular injection on the knee with either 1 ml of 40 mg/ml methylprednisolone (corticosteroid) dissolved in 4 ml lidocaine (10 mg/ml) or 1 ml isotonic saline (placebo) mixed with 4 ml lidocaine (10 mg/ml). Two weeks after the injections all participants undertook a 12-week supervised exercise program. Main outcomes were changes from baseline in pressure-pain sensitivity (pressure-pain threshold [PPT] and temporal summation [TS]) assessed using cuff pressure algometry on the calf. These were exploratory outcomes from a randomized controlled trial.Results
A total of 100 patients were randomized to receive either corticosteroid (n = 50) or placebo (n = 50); 45 and 44, respectively, completed the trial. Four participants had missing values for PPT and one for TS at baseline; thus modified intention-to-treat populations were analyzed. The mean group difference in changes from baseline at week 14 was 0.6 kPa (95% CI: -1.7 to 2.8; P = 0.626) for PPT and 384 mm×sec (95% CI: -2980 to 3750; P = 0.821) for TS.Conclusions
These results suggest that adding intra-articular corticosteroid injection 2 weeks prior to an exercise program does not provide additional benefits compared to placebo in reducing pain sensitivity in patients with knee OA.Trial Registration
