Telomeres are repetitive sequences (TTAGGG) located at the end of chromosomes. Telomeres progressively shorten with each cell replication cycle, ultimately leading to chromosomal instability and loss of cell viability. Telomere length anomaly appears to be one of the earliest and most prevalent genetic alterations in malignant transformation. Here we aim to estimate telomere length from whole-exome sequencing data in colon tumors and normal colonic mucosa, and to analyze the potential association of telomere length with clinical factors and gene expression in colon cancer.Reads containing at least five repetitions of the telomere sequence (TTAGGG) were extracted from the raw sequences of 42 adjacent normal-tumor paired samples. The number of reads from the tumor sample was normalized to build the Tumor Telomere Length Ratio (TTLR), considered an estimation of telomere length change in the tumor compared to the paired normal tissue. We evaluated the associations between TTLR and clinical factors, gene expression and copy number (CN) aberrations measured in the same tumor samples.Colon tumors showed significantly shorter telomeres than their paired normal samples. No significant association was observed between TTLR and gender, age, tumor location, prognosis, stromal infiltration or molecular subtypes. The functional gene set enrichment analysis showed pathways related to immune response significantly associated with TLLR.By extracting a relative measure of telomere length from whole-exome sequencing data, we have assessed that colon tumor cells predominantly shorten telomeres, and this alteration is associated with expression changes in genes related to immune response and inflammation in tumor cells.  相似文献

The contribution of heterotrophic plankton to nitrogen (N) regenerationin the water column, and its significance for the requirementsof phytoplankton, were studied at the seasonal scale in thecoastal upwelling ecosystem of A Coruña (Galicia, NWSpain). During 1995–1997, monthly measurements were takenof hydrographic conditions, dissolved nutrients, and abundanceand biomass of microplanktonic heterotrophs (bacteria, flagellatesand ciliates), phytoplankton and mesozooplankton (>200 µm).Additionally, series of experiments were conducted to quantifyN fluxes, including primary production (14C method), phytoplanktonuptake of nitrate, ammonium and urea (15N-labelling techniques),microheterotrophic regeneration of ammonium, mesozooplanktongrazing (chlorophyll gut-content method) and excretion of ammoniumby mesozooplankton. Two N budgets were built for the averagesituations of high (>100 mg C m-2 h-1) and low (<100 mgC m-2 h-1) primary production. The results revealed that phytoplanktonrelied strongly on regenerated ammonium all year round (33 and43% of total N uptake in high and low production situations,respectively). This demand for ammonium was closely matchedby regeneration rates of microplankton (0.14–0.25 mmolN m-2 h-1), whereas zooplankton contributed on average <10%to N regeneration. Likewise, zooplankton grazing had littledirect control on phytoplanktonic biomass. The results obtainedindicate that in the A Coruña upwelling system, N biomassof heterotrophic plankton is generally higher than phytoplanktonN biomass. The high rates of N regeneration measured also suggestthat a large proportion of the organic matter produced afteran upwelling pulse is recycled in the water column through themicrobial food web.  相似文献

Vibrio mimicus contamination of sand increased significantly during the arrival of the olive ridley sea turtles (Lepidochelys olivacea) at Ostional anidation beach, Costa Rica. Statistical analysis supports that eggs are contaminated with V. mimicus by contact with the sand nest. V. mimicus was isolated from eggs of all nests tested, and ctxA+ strains were found in 31% of the nests, all of which were near the estuary.  相似文献

Increasing global energy demands have led to the ongoing intensification of hydrocarbon extraction from marine areas. Hydrocarbon extractive activities pose threats to native marine biodiversity, such as noise, light, and chemical pollution, physical changes to the sea floor, invasive species, and greenhouse gas emissions. Here, we assessed at a global scale the spatial overlap between offshore hydrocarbon activities and marine biodiversity (>25,000 species, nine major ecosystems, and marine protected areas), and quantify the changes over time. We discovered that two‐thirds of global offshore hydrocarbon activities occur in areas within the top 10% for species richness, range rarity, and proportional range rarity values globally. Thus, while hydrocarbon activities are undertaken in less than one percent of the ocean's area, they overlap with approximately 85% of all assessed species. Of conservation concern, 4% of species with the largest proportion of their range overlapping hydrocarbon activities are range restricted, potentially increasing their vulnerability to localized threats such as oil spills. While hydrocarbon activities have extended to greater depths since the mid‐1990s, we found that the largest overlap is with coastal ecosystems, particularly estuaries, saltmarshes and mangroves. Furthermore, in most countries where offshore hydrocarbon exploration licensing blocks have been delineated, they do not overlap with marine protected areas (MPAs). Although this is positive in principle, many countries have far more licensing block areas than protected areas, and in some instances, MPA coverage is minimal. These findings suggest the need for marine spatial prioritization to help limit future spatial overlap between marine conservation priorities and hydrocarbon activities. Such prioritization can be informed by the spatial and quantitative baseline information provided here. In increasingly shared seascapes, prioritizing management actions that set both conservation and development targets could help minimize further declines of biodiversity and environmental changes at a global scale.  相似文献

Herbivory by copepods was studied from the coast towards theocean, during a bloom in May 1994 off NW Spain.Ingestion rateswere estimated by the gut chlorophyll content method in threesize fractions. The chlorophyll content displayed significantdaily cycles. Three different water bodies were described: coastal,shelf break and oceanic; the latter two zones separated by athermo haline frontal structure. Marked differences in planktonspecies composition, vertical distribution and biological rateswere found between zones. Theiiighest phytoplankton biomass,dominated by chain- forming diatoms, occurred in the oceaniczone associated with low primary production rates. Copepod feedinghad a low effect on oceanic phytoplankton; up to 0.2% of carbonstock and<3% of carbon production was consumed daily. Incontrast, medium-sized and large copepods removed 3% of carbonstock and 12% of primary production daily near the coast, wherephytoplankton were dominated by small flagellates in activegrowth. The highest variability in both plankton compositionand ingestionrates was found in the shelf-break zone, probablydue to displacements of the front. Copepods exerted a moderatepredation pressure on phytoplankton in coastal waters. Meanwhile,the impact of copepods on the offshore bloom was negligibleand the fate of the accumulated particulate carbon would bemostly determined by sedimentationand water dynamics.  相似文献

Organochlorine pesticide residues were found in 82 samples of human adipose material from 82 surgical cases in 16 Costa Rica hospitals. Identification was made by gas-liquid chromatography. The highest pesticide concentration was that of DDT and its metabolites (33.16 micrograms/g). Residues of almost all commercial pesticides were also found. Concentrations of alpha-chlordane. Aldrin and Polychlorinated biphenyls were not significant.  相似文献

A number of biomarkers (e.g. Abeta, tau) has been identified in Alzheimer's disease CSF. However, none fulfils the criteria of sensitivity and specificity (> 80%) needed for the development of an accurate diagnostic test. The lack of a suitable marker has prompted the search for new CSF biomarkers. In this study, the glycosylation of CSF proteins was examined using lectin blotting. Lumbar CSF was collected ante mortem from 22 non-Alzheimer's disease and 12 probable Alzheimer's disease cases and ventricular CSF collected post mortem from 7 non-Alzheimer's disease and 16 Alzheimer's disease cases confirmed by pathologic examination. When CSF glycoproteins were stained with wheat germ agglutinin (WGA), the staining intensity was found to be significantly lower in the Alzheimer's disease group. No difference in staining was found using other lectins (Canavalia ensiformis agglutinin, Ricinus communis agglutinin, Lens culinaris agglutinin). The measurement of WGA-reactive glycoproteins in CSF may be a useful biomarker for diagnosis of Alzheimer's disease.  相似文献

We have reported the isolation of the tetranortriterpenoid 1-cinnamoyl-3,11-dihydroxymeliacarpin (CDM) from partially purified leaf extracts of Melia azedarach L. (MA) that reduced both, vesicular stomatitis virus (VSV) and Herpes simplex virus type 1 (HSV-1) multiplication. CDM blocks VSV entry and the intracellular transport of VSV-G protein, confining it to the Golgi apparatus, by pre- or post-treatment, respectively. Here, we report that HSV-1 glycoproteins were also confined to the Golgi apparatus independently of the nature of the host cell. Considering that MA could be acting as an immunomodulator preventing the development of herpetic stromal keratitis in mice, we also examined an eventual effect of CDM on NF-kappaB signaling pathway. CDM is able to impede NF-kappaB activation in HSV-1-infected conjunctival cells and leads to the accumulation of p65 NF-kappaB subunit in the cytoplasm of uninfected treated Vero cells. In conclusion, CDM is a pleiotropic agent that not only inhibits the multiplication of DNA and RNA viruses by the same mechanism of action but also modulates the NF-kappaB signaling pathway.  相似文献

Ammonium regeneration and dissolved organic nitrogen (DON) releasewere studied experimentally in the euphotic zone of shelf andoceanic waters of NW Spain in relation to coastal upwellingdynamics and the size-structure of phytoplankton communities.Incubations of plankton labelled with [15N]ammonium were madeduring four cruises, two of which also included size-fractionateddeterminations of chlorophyll a and primary production, andexperimental determinations of production rates of dissolvedorganic carbon (DOC) using 14C. Inorganic nitrogen concentrationswere mainly related to nitrate enrichment by upwelling pulses,while ammonium concentrations were generally low in all situations.Ammonium did not accumulate in the study area, suggesting adaily time scale coupling between regeneration and uptake. Incontrast, DON largely exceeded inorganic nitrogen in all situationsand generally increased from spring to autumn. Ammonium regenerationwas positively correlated with DON release and both rates showedthe largest variation in summer, with minimum values duringactive upwelling and maximum values when upwelling relaxed.Comparison of DON stocks and rates in different shelf areassuggests that DON release near the coast during summer was morepersistent in the water than DON release in off-shelf and oceanicareas. The carbon:nitrogen ratio of DOC and DON release rateswas highly variable, revealing a large excess of DOC comparedwith DON. This excess can be attributed to either an underestimateof total DON release (as only release from ammonium was measured)or to an enhanced production of carbon-rich organic substancesby diatoms in coastal areas. By considering a broad range oftrophic situations, this study reveals a fundamental differencebetween short term release of DOC and DON by plankton. Physiologicalprocesses (such as carbohydrate exudation by diatoms) seem tobe the cause of large DOC excess, whereas trophic processes(such as grazing) are more likely the cause of enhanced DONrelease.  相似文献
