A simple and efficient garlic in vitro shoot regeneration protocol has been developed. This system uses axenic root tips cultivated from the beginning in the presence of light and does not require any change or refreshing of the original medium during the entire process. The application of light from the beginning of the culture process did not affect the callus formation rate but did significantly improve the explant regeneration ability. Since the pioneering work by Julius Adler in the 1960's, bacterial chemotaxis has been predominantly studied as metabolism-independent. All available simulation models of bacterial chemotaxis endorse this assumption. Recent studies have shown, however, that many metabolism-dependent chemotactic patterns occur in bacteria. We hereby present the simplest artificial protocell model capable of performing metabolism-based chemotaxis. The model serves as a proof of concept to show how even the simplest metabolism can sustain chemotactic patterns of varying sophistication. It also reproduces a set of phenomena that have recently attracted attention on bacterial chemotaxis and provides insights about alternative mechanisms that could instantiate them. We conclude that relaxing the metabolism-independent assumption provides important theoretical advances, forces us to rethink some established pre-conceptions and may help us better understand unexplored and poorly understood aspects of bacterial chemotaxis.  相似文献

参考Friedman等分离人肝脏星形细胞(HSC)的方法,采用校正密度的淋巴细胞分离液进行一步梯度离心,成功地分离得到了正常人HSC。HSC的收获量约为1×10~7个/10克肝脏组织,存活率在95%以上,纯度达85%以上。传代后纯度接近100%。对人HSC的标志物进行检测发现,结蛋白不宜作为鉴定初分离的及原代培养初期人HSC纯度的指标,而α-平滑肌肌动蛋白可作为鉴定激活的人HSC的可靠指标。  相似文献

Summary A simple protocol for garlic in vitro management is described. It comprises a single medium for all developmental stages and genotypes tested, and the use of immature bulbs as source of axillary buds. Although genetic variability exists among the different accessions tested, for both the multiplication rate and bulblet size, acceptable values of multiplication were reached for all the accessions, and values of bulb formation approached 100% in the shoots produced.  相似文献

Swine coronaviruses affecting pigs have been studied sporadically in wildlife. In Argentina, epidemiological surveillance of TGEV/PRCV is conducted only in domestic pigs. The aim was to assess the prevalence of TGEV/PRCV in wild Suina. Antibodies against these diseases in wild boar and captive collared peccary were surveyed by ELISA. Antibodies against TGEV were found in three collared peccaries (n?=?87). No TGEV/PRCV antibodies were detected in wild boar (n?=?160). Preventive measures should be conducted in contact nodes where the transmission of agents may increase. Epidemiological surveillance in wildlife populations and in captive animals before their reintroduction should be attempted.

The biolistic particle delivery system was used to transfer the uidA gene into different garlic tissues, including regenerable calli. These tissues showed a high endogenous nuclease activity preventing exogenous DNA expression. After submerging the tissues for 24 h in a 2 mm solution of the nuclease inhibitor aurintricarboxylic acid, transient expression of the uidA gene was detected. Furthermore, vacuum infiltration of aurintricarboxylic acid into the garlic tissues significantly increased the percentage of explants showing transient expression. Twenty garlic Allium sativum (L.) genotypes were analysed for genetic variation in their ability to form callus (in one medium) and regenerate shoots (in four different media). Genotypes showed significant differences in the analysis of variance of all the traits tested. The accession Printanor showed the best general behaviour, with 83% callus-producing explants, 44.7% organogenic explants, and 15.35 shoots/g of callus. The best regeneration medium was MBO, without growth regulators. Shoot production capacity was examined with the additive main effects and multiplicative interaction model that proved to be a powerful tool for analysis and easy comprehension of the strong genotype×medium interactions frequently observed in in vitro culture systems.
